#Antioxidant #Betaxanthins Process Anti #Cancer Activities Through Multi-Target Action, According to Studies

By Kyle J. Norton

Cancer is a class of chronic diseases caused by cell growth and multiplies disorderedly and uncontrollably in the body.

Every day millions of cells grow to replace millions of dead cells to keep our body in balance and healthy. Sometimes, acceleration of cell growth is necessary to prevent further damage to the body organ, such as cuts, injuries, etc., but after fulfilling their purposes, the growth of cells is back to a normal rate again.

Under normal circumstances, cell growth is limited to several cycles before dying and replacing b new cells. However, in cancer, the invasive cell can grow indefinitely. In other words, the cancer cell ignores the cell growth inhibitor.

According to the statistics provided by the American Cancer Society, in 2019, 1,762,450 new cancer cases will be diagnosed in the US. The disease will also cause the death of 606,880 Americans in the same year.

Believe it or not, the study continued, "Over the past decade of data, the cancer incidence rate (2006-2015) was stable in women and declined by approximately 2% per year in men, whereas the cancer death rate (2007-2016) declined annually by 1.4% and 1.8%, respectively".

Depending on the type and grade of cancer, some cancers can be progressively worse and damage other healthy tissues. Sometimes cancers can spread to other tissues and organs in the body via lymph or blood.

The most common prevalent factors that induce cancer risk are
* Aging is a prevalent risk factor for cancer development as most patients with cancer are found in older adults. Some researchers suspected that free radicals may be one of the contributors.

* Tobacco use is associated with an increased risk of lung cancer, due to chemical-induced cells alternation

* Sunlight is the major cause of skin cancer,

*ionizing radiation: Any type of hazardous chemicals cause gene mutation

* Viral and bacterial infection

* Hormones: Abnormal production of certain hormones can increase the risk of sex-related cancers

* Family history.

Carcinoma cancer is the most common type of malignant tumor found in anywhere of the epithelial tissues lining the cavities, surfaces of structures, and glands, accounting for 90% of all cancer cases.

Betaxanthins are phytochemicals in the class of red and yellow indole-derived pigments, belonging to the group of Betalains, found abundantly in beets, Sicilian prickly pear, etc.

To find a potential compound for the treatment of inflammatory disease, researchers examined the anti-inflammatory effects of red beetroot betaxanthins.

Vitexin-2-O-xyloside found in beetroot, in combination with betaxanthins and betacyanins, exerted antiproliferative activity in breast, liver, colon, and bladder cancer cell lines.

This efficacy was attributed to the synergistic activity in inhibiting genes associated with pro-survivals, angiogenesis, and promoting apoptosis.

The combination inhibited the hypoxia-inducible factor 1A, which responded to the decrease in available oxygen in the cancer cellular environment.

These results strongly suggested that these phytochemicals enhance their anticancer activity through multi-target action.

According to the findings, researchers said, "Vitexin-2-O-xyloside, betaxanthins, and betacyanins can be combined with conventional anticancer drugs to reduce their toxicity and overcome the multidrug resistance of cancer cells".

Taken altogether, betaxanthins combined with other phytochemicals may be considered a remedy for the prevention and treatment of cancer, pending the confirmation of the larger sample size and multicenter human study.

#Herbal #ArtichokeLeafExtracts Process Anti Mesothelioma Properties, Researchers Show

By Kyle J. Norton

Mesothelioma is a type of cancer that develops from the thin layer of tissue known as the mesothelium.
Most cases of mesothelioma are found on the lining of the lungs and chest wall, compared to other organs. Pleural mesothelioma is a cancer of the pleural lining of the lung.

Exposure to asbestos which has been banned is one of the most common causes of the disease. Some researchers suggested that inhaling asbestos can cause mesothelioma in many ways, including
* Induced chronic low-grade inflammation of mesothelial cells as the immune system can not completely drive out the fibers that cause damage to the lung, leading to the compromise.

* Long-term exposure to asbestos can suppress the genes associated with limited cell cycle division and tumor suppression

* Long-term exposure is also found to alter the cell DNA, leading to cancer.

According to statistics, 2000 to 3,000 people are diagnosed with mesothelioma each year in the United States. The asbestos risk of pleural mesothelioma ranges from 2 to 10 percent.

Most cases of malignant mesothelioma are closely associated with past occupational exposure.
However, asbestos is found persistently in our natural and built environments.

Besides most common cancer symptoms, patients with pleural mesothelioma at the advanced stage may also experience symptoms of dry cough or wheezing, shortness of breath (dyspnea), respiratory complications, and pain in the chest or abdomen.

Some patients may also develop pleural effusion (fluid around the lungs), fever or night sweats, fatigue, and muscle weakness

Artichoke is a perennial thistle of Cynara cardunculus species of the Cynara genus, belonging to the family Carduoideae native to Southern Europe around the Mediterranean.

The herbal plant has been used in traditional medicine as a liver protective and detoxified agent and for the treatment of digestive disorders, abdominal pain gas and bloating, etc.

In the concern of the malignant pleural mesothelioma, a poorly treated neoplasia by conventional medicine, arising from the pleural mesothelial lining, researchers examined leaf extract of Cynara scolymus in vitro and in vivo.

The study included tMSTO-211H, MPP-89, and NCI-H28 mesothelioma cell lines and untransformed mesothelial cells HMC, treated with increasing concentrations (ranging from 3 to 200 μg/ml) of artichoke leaf extract for 72 hrs.

Injection of the leaf extract not only inhibited strong cell proliferation but also affected the migration and tumor engraftment in the mesothelioma cell lines.

More precisely, MSTO-211H cells treated with the artichoke extract showed a change in the cellular morphology in a short-term assay, compared to more resistant resistance in the HMC cells.

In MSTO-211H, MPP-89, and NCI-H28 cells, injection of artichoke leaf extract inhibited the formation of cancer cell colony colonies.

Astonishingly, dietary feeding with Cynara scolymus leaf extract showed a similar inhibition against the growth of mesothelioma xenografted tumors comparable to those of pemetrexed.

Where pemetrexed is a chemo medicine used for the treatment of cancer, including mesothelioma.

Dr. Claudio Pulito and colleagues suggested, "leaf extract of Cynara scolymus holds therapeutic potential for the treatment of mesothelioma".

Taken altogether, artichoke leaf extract may be considered a functioning remedy for the prevention and treatment of mesothelioma, pending the confirmation of a large sample size and multicenter human study.

#Bromelain Processes #PlateletInhibition Properties, According to Studies

By Kyle J. Norton

Fibrinolysis is the condition associated with the enzymatic breakdown of fibrin in blood clots.
Where fibrin plays an essential role in hemostasis as the primary product of the coagulation cascade of fibrinolysis.

In other words, fibrinolysis is a process that prevents blood clots from growing to induce certain health problems such as cardiovascular disease.

Platelet aggregation is a condition characterized by platelets clumping together in the blood. The process also is necessary for the cover of vascular injury. However, overexpression of platelet counts in the blood is problematic for human health.

In other words, the production of blood platelets is to protect tissue injury against infection.
Furthermore, platelet disorders can lead to abnormal bleeding. Most common types of the genetic proposition of platelet disorders including

* Disorder of platelet adhesion, also known as Bernard-Soulier Syndrome is a condition in which the platelets fail to adhere together, leading to frequent bruises, nose bleeds, and bleeding in the mouth and gums.

* Disorder of platelet aggregation also known as Glanzmann Thrombasthenia is a condition that the platelets not only fail to cover at the site of an injury but also prolong the bleeding.

* Disorder of platelet secretion is a condition that causes slows down normal platelet adhesion, aggregation, and repair of the blood vessel.

* Disorder of platelets procoagulant activity, also known as Scott Syndrome is a condition in which the platelets fail to promote activation of the blood clotting proteins.

Conventionally, certain medications with blood-thinning activity such as aspirin (ASA) and other drugs containing aspirin non-steroidal anti-inflammatory drugs, and some antibiotics may have a strong implication for the formation of platelets coagulation.

Dr. Wolberg AS in the examination of fibrinolysis in the changes in the local concentrations of pro- and anti-coagulants and cellular activities may affect the clot-forming wrote, "patterns of abnormal thrombin generation produce clots with altered fibrin structure and that these changes are associated with an increased risk of bleeding or thrombosis".

These results strongly outline the importance of fibrin in the formation of a blood clot in the presence of thrombin that converts fibrinogen to fibrin.

Bromelain, a proteolytic enzyme found in pineapples (Ananas comosus) has been used in traditional medicine as an inflammatory agent and to treat pains, strains, muscle aches and pains and ease back pain and chronic joint pain, skin diseases, etc.

In finding a potential compound for the treatment of fibrinolysis, researchers examined the influence of bromelain on platelet count, platelet aggregation, and, platelet activity in vitro.

The study included blood samples taken from the antecubital vein of 10 healthy male non-smokers.

According to the tested observation, platelet count decreased after incubation with 2.5 and 5 mg bromelain/ml.

Interestingly, according to the flow cytometry, the percentage of the total number of platelets or unstimulated CD62P positive platelets.

In other words, under normal conditions, bromelain does not have any effect on unstimulated CD62P positive platelets even with higher incubation with 5 mg bromelain/ml compared to control.

However, after TRAP-6 stimulation with platelet aggregation, the incubation with 5 mg bromelain/ml led to a remarkable decrease, compared to the untreated control.

The changes of CD62P (TRAP-stimulated) and the results of platelet aggregation after incubation with bromelain in vitro may demonstrate the potential of bromelain as a substance for platelet inhibition.

Based on the findings, researchers said, "The changes of CD62P (TRAP-stimulated) and the results of platelet aggregation after incubation with bromelain in vitro may demonstrate the potential of bromelain as a substance for platelet inhibition".

These results suggested that bromelain has a strong effect on the prevention and treatment of cardiovascular disease associated with platelet aggregation.

Taken altogether, bromelain may be considered a remedy for the prevention and treatment of fibrinolysis,, pending the confirmation of the larger sample size and multicenter human study.

#Healthyfood #Greenbean Decreases Blood Glucose in VIvos, Researchers Find

By Kyle J. Norton

Hyperglycemia is a condition of abnormally high blood sugar in the bloodstream, inducing diabetic complications in patients with type 2 diabetes.
In order words, hyperglycemia is a result of the body which does not properly use or make the hormone insulin.

The condition is also considered a hallmark of patients with type 2 diabetes. Another hallmark is insulin resistance.

Hyperglycemia occurs mostly due to poor control and management in patients with diabetes such as
a poor diet, physical inactivity choices, symptoms, and medicines intake.

Some patients with diabetes also experience symptoms of hyperglycemia due to skipping or not taking enough glucose-lowering medication.

The most common symptoms of hyperglycemia are frequent urination, increased thirst, blurred vision, persistent fatigue, and headache.

According to epidemiological studies, untreated or long-term persistently hyperglycemia in patients with diabetes has been found to induce serious symptoms such as shortness of breath, and confusion.

In serious cases, untreated hyperglycemia also induces long-term complications of cardiovascular disease, nerve damage associated with skin infections, and ulcerations.

Green bean is a genus of Phaseolus Sensu Stricto, containing 55 different species, belonging to the family Fabaceae and native to the New World tropics and grown in Europe for its attractive flowers and fleshy immature pods. The veggie can be classified into two major groups, bush beans, and pole beans.

In finding a potent veggie for the prevention of hyperglycemia, researchers examined the effects of bean extract on overexpression of free radicals and higher reactive oxidative species (ROS) production induced high blood glucose.

The randomized controlled trial (RCT) study included mice (Micetus norvegicus) of Wistar strains randomized in five groups: P1-negative control, positive P2-control, P-3 ethanol extract group of 200 mg/kg BW, P4-group 400 mg/kg BW, P5- group 600 mg/kg BW.

According to the differences in blood sugar levels and superoxide enzyme levels of intergroup dismutase, ethanol extract of green beans 600 mg/kg BW from day 14 to day 28 (p <0.05) showed a statistically significant decrease in blood glucose levels.

However, ethanol extract of green beans 200 mg/kg BW and 400 mg/kg GB from day 7 until day 28 (p> 0.05) did not have any effect on the decreasing blood glucose in the tested rats.

The tested ethanol extract of green beans showed no effect on the levels of activity of the superoxide dismutase enzyme (p = 0.830).

In other words, green bean extract inhibited the expression of hyperglycemia without affecting the levels of antioxidant enzymes produced by the host in a dose depending manner.

Based on the findings, researchers said, "Giving ethanol extract of green beans 600 mg/kg BW effectively decreases blood sugar in male Wistar mice (Micetus norvegicus) from day 14 to day 28".

Taken altogether, green beans may be used as an adjunct therapy for the prevention of diabetes by inhibiting the expression of hyperglycemia, pending the confirmation of a larger sample size and multicenter human study.

#Greentea #Polyphenols Show a Significant Effect on Delayed Against #Cartilage Reduction and Ioint Damage in Patients of Arthritis

By Kyle J. Norton

Green tea may have a therapeutic and positive effect in the protection of the cartilage against the degeneration of arthritis in the joint, some scientists suggested.
Green tea is a precious drink that processes several health benefits known to almost everyone in Asia and the Western world.

Cartilage is a connected tissue, rubber-like padding between the joint of the bone with the function to protect against bones rubbing against each other at the endpoints.

In the study of adolescent male Wistar rats with 12-week exposure to Cd and Pb (7mg Cd and 50mg Pb in 1kg of the diet randomly assigned to 12 to each group, as positive control received without Cd, Pb, and teas, a negative control group received Cd and Pb, and groups supplemented additionally with green (GT), black (BT), red (RT), and white tea (WT), application of tea significant inhibited the decreased levels of the geometric(shape) and densitometric(density) parameters and the total thickness of articular cartilage caused by Cd and Pb in compared to the negative control group.

The tea injection group also displayed a huge improvement in mechanical endurance, growth plate thickness, and trabecular histomorphometry depending on the tea type.

Due to the limitation of the study, dr. Tomaszewska E, the lead researcher said, " It is difficult to indicate which tea has the best protective effects on bone and hyaline cartilage against heavy metal action".
Some scientists insisted that green tea with bioactive epigallocatechin-3-gallate, a green tea polyphenol, maybe the one was contributed to the cartilage protection against various types of influence.

Application of green tea EGCG in drinking water of mice was found to prevent collagen-induced arthritis by inhibiting the production of certain pro-inflammatory factors in association with damage of collagen such as tumor necrosis factor-alpha (TNFα), a cell-signaling cytokine with pro-inflammatory involvement in systemic inflammation and cyclooxygenase-2, an enzyme in response to inflammation and pain.

In EGCG treatment of human chondrocytes derived from OA cartilage, the application inhibited and modulated IL-1β cytokines in induced activity and expression of cyclooxygenase-2 through its immune and inflammatory signaling and production of nitric oxide in the contribution of T lymphocyte dysfunction. and prostaglandin E2 in the promotion of inflammation in chondrocytes.

According to the study, the inhibition of IL-1β by EGCG in human chondrocytes was also associated with its inhibitory effects on the activation of the nuclear translocation of NF-κB in the expression of proinflammatory activity.

Dr. Santosh K Katiya, the lead author, after taking into account other co-founders said, "These data thus provide a mechanistic link in the prevention of arthritis responses as well as potential therapeutic value for EGCG/GTPs inhibiting cartilage resorption in arthritic joints".

Truly, in investigating the effects of catechins found in green tea on cartilage extracellular matrix components using bovine nasal and metacarpophalangeal cartilage as well as human nondiseasednondiseased, osteoarthritis and rheumatoid cartilage cultured with and without reagents known to accelerate cartilage matrix breakdown in vitro, researchers found that catechins, containing gallate ester, were most effective in inhibited proteoglycan and type II collagen breakdown without inducing adverse effects.

These results suggested that some green tea catechins may have a significant effect in protecting or delayed against cartilage reduction and joint damage through the protection of chondrocytes.

Dr. Adcocks C, the lead author said, "Green tea may be prophylactic for arthritis and may benefit the arthritis patient by reducing inflammation and slowing cartilage breakdown".

Taken together, green tea with bioactive polyphenols may be considered a functional food for reduced or delayed progression of cartilage damage against type II collagen degradation and used the combination with standard medicine for the treatment of cartilage inside the joints in arthritis patients.

#Phytochemical #Betanin, Shows A Chemopreventive Activity and Reduces Side Effects of Chemo-Drugs in the Treatment of #Cancer

By Kyle J. Norton

Cancer is a class of chronic diseases associated with the alternation of cell DNA.
Most cases of cancer start in the cell on the surface of the inner lining of the organ tissue, before penetrating deeper into other layers to form a tumor.

Epidemiologically, most cases of cancer at the very early stage are asymptomatic. However, at the advanced stage, primary cancer cells can travel a distance away from the original site to infect other healthy tissues and organs.

Types of secondary metastasis can be classified into

1. Local spreading, lymph vessel, blood vessel

* Local spreading as soon as the cancer cells grow to their tumor size, will invade the nearby area.

* Spread through lymph vessel
If some cancer cells detached from the primary tumor and travel, they can grow into secondary cancer but most of the time they are trapped in the nearby lymph nodes closest to the original tumor.

* Spread through blood circulation
Similar to spread through lymph vessels, cancer cells detached from an original tumor can enter the bloodstream by slipping past the blood vessel wall and start growing into the secondary tumor as they are trapped in some small capillaries.

2. Travel of clumps of cancer along the channels
Sometimes, cancer cells detached from the primary tumor can travel through other channels and spread to some parts of the body that are more vulnerable to becoming metastatic sites than others.

3. The arrival of cancer cells at a distance site
The development of the survival of cancer cells gets trapped and grows into the second tumor.

4. Angiogenesis
Angiogenesis is a process involving the growth of new blood vessels from pre-existing vessels. The secondary tumor can persuade the adjacent tissue to build blood vessels by providing it with vital nutrients and oxygen.

At this stage, the overgrowth of the tumor may also press on the nerve cells, blood vessels, and healthy tissues, leading to specific symptoms, depending on the types of tumor accompanied by the general symptoms shared by all cancers.

Betanin is phytochemical in the class of red and yellow indole-derived pigments of Betacyanins, belonging to the group of Betalains, found abundantly in beets, chard, etc.

In the urgency to find a potential and natural compound for the treatment of cancer with no side effects, researchers examined the effects of betanin (BET) in reducing the side effects of potent anticancer drugs e.g. doxorubicin (DOX) on the normal tissues in co-administration.

To overcome the incomplete oral absorption and low stability of BET that limits its application, the synergistic effect of DOX@BET by a new pH-responsive nanocarrier via decoration of gelatin nanoparticles (GNPs) (methoxy poly (ethylene glycol)-poly ((2-dimethylamino) ethyl methacrylate-co-itaconic acid) (PGNPs) was designed.

According to the triggered release ability of drugs assays, the GNPs not only control the release of DOX and BET by adjusting the pH of the environment but also decreased the cell viability amount of breast cancer cells (MCF-7) compared to the free form of DOX or BET.

In other words, BET combined with DOX not only enhanced the chemo drug toxicity against cancer but also reduced the side effect found in the free form of DOX.

Furthermore, in two mouse lung tumor models, researchers at the Washington University School of Medicine investigated the chemopreventive activity of betanin by oral consumption.

The experiment included vinyl carbamate (VC) and benzo(a)pyrene (B(a)P) induced lung cancer female A/J mice with lung cancer were treated with betanin in drinking water.

According to the tested assays, injection of betanin significantly decreased tumor multiplicity and tumor load induced by both carcinogens by 20% and 39% in the VC lung model, and by 46% and 65% in the B(a)P lung model, respectively.

The efficacy of betanin in both lung cancer models has been attributed to the phytochemical lung tumor inhibitory effects in the induction of apoptosis and inhibition of angiogenesis.

Dr. Zhang Q, the lead scientist after taking other factors into account wrote, " Our data show that betanin significantly inhibits lung tumorigenesis in A/J mice and merits investigation as a chemopreventive agent for human lung cancer:.

Taken altogether, betanin may be considered a supplement for the prevention and treatment of cancer, pending the confirmation of a larger sample size and multicenter human study.

Intake of betanin in the form of a supplement should be taken with extreme care to prevent overdose acute liver toxicity.

#GreenTea #Epigallocatechin3gallate (#EGCG) Inhibits the Risk, Progression of #Fibromyalgia Caused by Inflammatory Rheumatic Disorders

By Kyle J. Norton

Green tea may have a therapeutic and positive effect in reduced onset, progression, and treatment of inflammatory rheumatic disorders induced fibromyalgia, some scientists suggested.
Green tea is a precious drink that processed a number of health benefits known to almost everyone in Asia and the Western world.

Fibromyalgia (FM) is a chronic condition characterized by muscle and soft tissue pain, affecting over 10 million (3.6% of the population) people in the US alone.

Inflammatory rheumatic disorder is a condition of autoimmune diseases caused by inflammatory connective tissues, joints, and vessels of the body, including rheumatoid arthritis, systemic lupus erythematosus, and vasculitis.

According to Cairo University, as many as 15-30% of patients with classic inflammatory rheumatic disorders also have co-morbid fibromyalgia syndrome (FMS) compared to the rate of only 2% of the prevalence of FMS in the general population (2%).

These results suggested that rheumatological syndromes have a strong implication in the development of fibromyalgia syndrome (FMS) compared to other initiators such as stress.

Therefore, patients with chronic rheumatic diseases are capable of triggering the condition of fibromyalgia syndrome (FMS).

Due to the highly correlated rheumatologic disease and FMS component, researchers postulated "Considerations of the FMS component in the management of rheumatologic diseases increase the likelihood of the success of the treatment" and "(prevented) FMS in misdiagnosed as an autoimmune disorder"

In the study of the effect of green tea's active ingredient, epigallocatechin 3-gallate (EGCG) on the risk of inflammatory rheumatic disorders in collagen-induced arthritis (CIA) in mice, researchers found that application of green tea demonstrates a substantially marked inhibition of the inflammatory mediator's cyclooxygenase (COX) -2 in converting arachidonic acid in stimulated production of proinflammatory cytokine prostaglandin H2, in catalyzation of symptoms of inflammation in the muscles.

Furthermore, green tea EGCG oral ingestion also inhibited levels of interferon-gamma in precipitated inflammation by modulating T-cells and natural killer cells activated by antigens, mitogens, or alloantigens to the site of infection.

Tumor necrosis factor-alpha (TNFα) involved in systemic inflammation, one of the cytokines that make up the acute phase reaction in mice was also inhibited by injection of green tea EGCG.

These results suggested that green tea ameliorated symptoms and expression of inflammatory rheumatic disorders, through attenuating production of pro-inflammatory factors without reducing the healing progression of injured tissue.

The above differentiation was supported by the study of green tea extract GTE in modestly ameliorated rat adjuvant-induced arthritis.

According to researchers, a daily per-oral administration of GTE (200 mg/kg) reduced expression of RA through reduced levels of MCP-1/CCL2 in expression (which can also be treated by biological agents, such as antibodies and inhibitory peptides) of a variety of diseases characterized by mononuclear cell infiltration involved substantial biological and genetic evidence, including RA.
and GROα/CXCL1 in the recruitment of inflammatory cells in the wound healing process after injury in angiogenesis and information of granulation tissue of wounds.

Additionally, the application of green tea besides attenuating the expression of proinflammatory factors also exhibited the production of chemokine receptors CCR-1, CCR-2, and CCR-5 in reduced direct communication between the innate and adaptive immune responses in initiating the development of inflammatory responses. through inactivation of leukocytes in inflamed tissues in contribution to the pathogenesis of inflammatory and autoimmune diseases.

And CXCR1 receptor binding to IL-8 in reduced the expression of some inflammatory cytokines in initiated inflammation such as epidermal growth factor (EGF) and tumor necrosis factor-α (TNF-α).

Some researchers suggested that the application of green tea extract may also reduce the symptoms of arthritis through the expression of IL-6 levels, as increased levels of Il-6 were associated with the presence and progression of rheumatoid arthritis (RA) found in conventional clinical and laboratory indices and MMP-2 activity in activated immune-inflammatory response through inducing the production of pro-inflammatory factors.

Taken together, green tea and its bioactive compound such as EGCG may have a profound and therapeutic effect in reducing the onset, progression, and treatment of fibromyalgia induced by inflammatory rheumatic disorders through modulating the production of pro and anti-inflammatory proteins. However, the intake of green tea supplements should be taken with extreme care due to recent reports of acute liver toxicity.
