#Herbal #Artichokes Inhibit the Parameters that Cause #NonalcoholicFattyLiverDisease (#NAFLD), Researchers Say

Artichoke may be used as a herbal remedy for the treatment of nonalcoholic fatty liver disease with no side effects, according to studies.
Nonalcoholic fatty liver disease is a fatty liver disease caused by fat accumulated in the liver other than those of excessive alcohol drinking.

The disease, in most cases, is asymptomatic, however, it has been seen to slowly progress to end-stage liver disease.

According to the statistics, non-alcoholic fatty liver disease affects over 25% of the US population.

Epidemiological studies suggested that most cases of nonalcoholic fatty liver disease are found in obese and extremely overweight adults. However, people with diabetes, high cholesterol, or high triglycerides are associated with an increased risk of the disease. Indeed, 75% of obese have steatosis and 20% to 35% have nonalcoholic steatohepatitis (NASH).

As of today, there is no effective treatment in patients with NAFLD, change of diet and lifestyle to induce weight loss are the most common nonmedical treatment conventionally for the management of the disease.

Some researchers suggested that the cause of NAFLD may be associated with the promotion of the Western diet over the past few decades.

Dr. Michael D. Roberts, the lead scientist in the evaluation of the Western diet risk in NAFLD, wrote, "...sub-chronic WD feeding appears to increase hepatic steatosis development over a 6-week period but only induces select inflammation-related liver transcripts, mostly acute phase response genes".

And, "diet-induced liver fibrosis and inflammation may either: a) occur much later with more chronic WD rat feeding paradigms, and/or b) may be initiated by other factors that precede eventual perturbations in local and systemic pro-inflammatory signalers with chronic WD feeding".

The results strongly suggested the Western diet pattern has a strong impact on the development of non-alcoholic fatty liver disease.

Artichoke is a perennial thistle of Cynara cardunculus species of the Cynara genus, belonging to the family Carduoideae, native to Southern Europe around the Mediterranean.

The herbal plant has been used in traditional medicine as a liver protective and detoxified agent and for the treatment of digestive disorders, abdominal pain gas and bloating, etc.


Scientists on finding a natural ingredient for the treatment of nonalcoholic fatty liver disease examined the artichoke (JA) potential in attenuating lipid disturbances in an animal model.

The study included Wistar of rats fed a control diet, a 60 % fructose-enriched diet (FRU), or an FRU with 10 % JA (n 6-7) for 4 weeks.

After 21 days of the experiment, according to the liver samples collected, the JA treatment group showed a significant improvement in a hepatic TAG accumulation, an indication of a fatty liver.

The expression of fat accumulation in the tested rats was observed by the expression of malic enzyme 1 (Me1), associated with fatty acid synthesis; decorin (Dcn), related to fibrosis altered by the FRU.

Additionally, the injection of the supplement also improved the gene associated with liver inflammation caused by FRU.

Furthermore, in order to reveal more information about artichoke anti-NAFLD, researchers conducted a randomized double-blind placebo-controlled parallel-group trial in ultrasound-diagnosed NAFLD.

100 subjects were randomized to either ALE 600 mg daily or placebo for a 2-month period.

Injection of AlE showed increased hepatic vein flow (p < .001), reduced portal vein diameter (p < .001), and liver size and levels of liver oxidative stress markers.

The ALE-treated group improved liver function by reducing the expression of the levels of bilirubin.

More importantly, ALE supplementation also reduced total cholesterol, low-density lipoprotein cholesterol, high-density lipoprotein cholesterol, non-high-density lipoprotein cholesterol, and triglyceride concentrations.

The results showed that artichoke anti-NALFD via several mechanisms, including the inhibition of levels of cholesterol and liver stress markers.

Taken altogether, artichoke may be considered a functional remedy for the prevention and treatment of non-alcoholic fatty liver disease, pending the confirmation of a larger sample size and multicenter human study.

* Furthermore, BSH injection increased mean relative mRNA levels of the NRF2 target HBG1, compared to the baseline at the end of treatment, without affecting the levels of HbF protein.


In other words, BSH displays a similar effect compared to that of sulforaphane in the promotion of the production of antioxidant enzymes against sickle cell disease (SCD).

Based on the findings, researchers said, "BSH, in the provided doses, is safe in stable SCD patients and may induce changes in gene expression levels".

Taken altogether, broccoli sprouts processed abundantly SFN may be considered a functional food for the treatment of SCD, pending the confirmation of the larger sample size and multicenter human study.

Intake of SFN in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Uveitis is a type of eye inflammation, affecting the middle layer of tissue in the eye wall (uvea).
The condition has been found to onset suddenly and get worse quickly. The most common symptoms of uveitis are eye redness, pain, and blurred vision that can affect one or both eyes.

In some cases, patients with the condition also experience symptoms of light sensitivity, floaters, and decreased vision.

The exact cause of uveitis is unknown. However, some researchers suggested that people with a compromised immune system associated with inflammatory disease and infected diseases are at an increased risk of uveitis.

Other risk factors, epidemiologically such as eye injury, diseases that affect the eye such as diabetes are also found in patients with uveitis.

Most cases of uveitis are treated by prescription eye drops in combination with anti-inflammatory medications. If all fail, surgery may be necessary.

Untreated uveitis has been found to induce complications of glaucoma, cataracts, optic nerve damage, and retinal detachment.

In finding a natural compound for the treatment of uveitis, researchers examined the effect on turmeric administered 4 h after induction of E. coli lipopolysaccharide-induced uveitis in rats.
Compared to the control group, turmeric showed significantly suppressed inflammation with a significantly lower mean clinical grade, histopathological grade, and aqueous humor (AH) protein level, second to the conventional medicine prednisolone.

The turmeric administered for 4 h did not affect proinflammatory cytokines levels of TNF-alpha levels.

However, when the aqueous extract was administered starting 3 days before induction of uveitis, the treatment group not only showed a similar effect on the mean clinical and histopathological grade, as well as AH protein but also reduced TNF-alpha levels, comparable to the treatment group.

Based on the findings, researchers said, "topically applied a standardized aqueous extract of C. longa suppresses endotoxin-induced uveitis in rats by reducing TNF-alpha activity".

In order to reveal more information about turmeric anti-uveitis properties, researchers examined the anterior uveitis induced in rabbits by intravitreal injection of lipopolysaccharide from Escherichia coli after pretreatment with C. longa and B. aristata aqueous extracts.

According to the clinical and histopathologic grading, anterior segment inflammation in the control group was significantly higher compared to both the extract-treated groups.

More precisely, the inflammatory cell count in the control group was 30.75 cells/mL compared to 2.39 and 11.56 cells/mL in the C. longa- and B. aristata-treated groups, respectively.

Similarly, C. longa- and B. aristata-treated groups also exerted strong effects on the protein content and TNF-alpha level as of that in the aforementioned study.

Taken altogether, turmeric may be considered a supplement for the prevention and treatment of uveitis, pending the confirmation of the larger sample size and multicenter human study.

Phytochemicals may be the next potential source of new medications for the treatment of diseases with little or no side effects.
Scientists may have found a dietary and bioactive ingredient for the prevention and treatment of diseases associated with oxidative stress.

Allicin is phytochemical containing sulfur in the class of organosulfur compounds, found abundantly in onion and garlic.

Oxidative stress is a condition characterized by the overexpression of free radicals in the host body.

Most cases of oxidative stress are caused by the inability of the host liver in stimulating the production of antioxidant enzymes which play a critical role in inhibiting the free radical expression, causing oxidative stress.

In a healthy individual, the ratio of free radicals and antioxidant enzymes is always balanced.

However, an individual whose antioxidant enzymes were suppressed due to damage to the antioxidant enzymes' productive system or other sources may have a greater risk of the onset of oxidative stress.

Dietary antioxidant intake such as fruits and vegetables may be helpful to keep the free radical in check.

Oxidative stress has been found to cause damage to cell lipids, proteins, and DNA, leading to many chronic diseases, including atherosclerosis, cancer, diabetics, rheumatoid arthritis, post-ischemic perfusion injury, myocardial infarction, cardiovascular diseases, chronic inflammation, and stroke.

The oxidative stress that causes chronic ailment is a result of a chain reaction started by free radical donating or stealing an electron from another atom, leading to a domino effect that can not be stopped until all outer ring electron of free radicals is paired or inhibited by antioxidants.

A free radical is any atom or molecule that has a single unpaired electron in an outer shell and highly reactive to react with another cell.

Most free radicals and other ROS are derived internally either from normal essential metabolic processes in the human body or from external sources such as exposure to X-rays, ozone, cigarette smoking, air pollutants, and industrial chemicals.



Antioxidants are biomolecules that inhibited oxidation.
On finding a natural ingredient and whole food for the prevention and treatment of diseases associated with oxidative stress, researchers investigated the effect of allicin (the active ingredient of garlic) contained in the commercial preparation of Garlicin against hydroxyl radicals (.OH) using high-pressure liquid chromatographic (HPLC) method.

Allicin is equivalent in Garlicin (1.8, 3.6, 7.2, 14.4, 21.6, 28.8, and 36 micrograms) produced concentration-dependent decreases in the formation of 2,3-DHBA and 2,5-DHBA, the OH adduct products.

The inhibition of the formation of 2,3-DHBA and 2,5-DHBA with 1.8 micrograms/ml was 32.36% and 43.2% respectively while with 36.0 micrograms/ml, the inhibition was approximately 94.0% and 90.0% respectively.

These results suggested that allicin is highly active against the OH adduct products through its free radical scavenging activity.

Furthermore, Allicin also exerted a significant antioxidant activity by preventing the lipid peroxidation of liver homogenate in a concentration-dependent manner.

Additionally, by examing the antioxidant effects of four main chemical classes, alliin, allyl cysteine, allyl disulfide, and allicin, prepared by chemical synthesis or purification, researchers discovered
* Alliin scavenged superoxide, while allyl cysteine and allyl disulfide did not react with superoxide.

* Allicin suppressed the formation of superoxide by the xanthine/xanthine oxidase system, probably via a thiol exchange mechanism.

* Allicin did not exhibit hydroxyl radical scavenging activity in this study, probably due to some unknown mechanisms.

The finding suggested that allyl disulfide, alliin, allicin, and allyl cysteine exhibit different patterns of antioxidant activities as protective compounds against free radical damage.

Taken altogether, allicin-processed high levels of free radicals scavenging activity may be used for the prevention of the overexpression of ROS in the initiation of oxidative stress.

#WildCarrots Exhibit the Apoptosis of #SkinCancer in Vivos, Researchers Suggest

Carrots may have a profound and substantial effect in reducing the risk, progression, and treatment of skin cancer, some scientists suggested.

Skin cancer is a condition characterized by cell growth disorderly and uncontrollably in skin tissue. At the later stage, the cancerous cells may infect other healthy tissue and organ a distance away from the original site.

Carrot, a root vegetable with orange color is a subspecies of Daucus carota, belongings to the family Apiaceae, native to Asia and Europe.

In the investigation of essential oil isolated from wild carrot umbels effect in mice induced skin cancer through injection of 7,12-dimethyl benz(a)anthracene (DMBA), after 20 weeks of administered to animals via gavage (0.02 mL of 100% oil), intraperitoneal (0.3 mL of 2% oil), and topical (0.2 mL of 5, 50, and 100% oil) with half-maximal inhibitory concentration IC50, researchers showed that topical administration of 100% mice group exerted in delay skin cancer appearance and inhibited tumor incidence of 89%.

With the concentration of IC40, topical 50% and 5% treatment inhibited tumor incidence by 83% and 36%, respectively.


Topical ingestion of 100, 50, and 5% oil, also revealed a significant decrease of tumor volume by 99, 91, and 70%, respectively.

Intraperitoneally, treatment mice displayed an inhibited tumor by 43% and decreased tumor volume by 85%, whereas gavage treatment showed minimal effects on both.

These results suggested that intraperitoneal and topical treatment of wild carrot essential oil decreased infiltration and proliferation with an increase in abnormal thickening of the outer layer of the skin.

Further analysis of the wild carrot essential oil anti-cancer effect on human epidermal keratinocytes and the chemopreventive activity of the pentane diethyl ether fraction on DMBA/TPA-induced skin carcinogenesis in mice, scientists in the joint study lead by the Lebanese American University suggested that application of all fractions isolated from the essential oil exhibited significant cytotoxicity, with HaCaT cells being 2.4-3 times less sensitive than HaCaT-ras A5 (benign tumorigenic), and HaCaT-ras II4 (malignant) cells observed by western blot.

In Gas chromatography-mass spectrometry, 2-himacharlen-6-ol. a derivative isolated from pentane/diethyl ether fraction (F2), demonstrated strong inhibition of HaCaT-ras A5 and HaCaT-ras II4 cells through cell cycle arrest in the accumulation of cells of the sub-G1 apoptotic phase and apoptosis in decreased the population of cells in the S and G2/M phases.

These results were attributed to the F2 fraction in enhanced expression of pro-apoptotic (Bax) in induction cellular apoptosis and attenuated expression of anti-apoptotic (Bcl2) proteins in promoted cancer proliferation.

Interestingly, all fractions also decreased the phosphorylation of AKT, (AKT is an essential component of the PI3K (phosphatidylinositol 3-kinase pathway) in monitoring skin cancer cell growth and resistance to apoptosis and ERK(components of mitogen-Activated Protein Kinase (MAPK) signaling pathways) in mediated different anti-proliferative events, such as apoptosis, autophagy, and senescence.

Based on the information fonding, Dr. Shebaby WN, the lead author said, " F2 fraction has remarkable antitumor activity against DMBA/TPA-induced skin carcinogenesis, an effect that may be mediated through inhibition of the MAPK/ERK and PI3K/AKT pathways".

The above differentiation was supported by the joint study led by the University of Surrey in the evaluation of Daucus carota oil extract (DCOE) activity in the inhibition of HaCaT-ras II-4 epidermal squamous cells.

According to the experiment, β-2-himachalen-6-ol isolated from the wild carrot essential oil at dose-dependent decreased in skin cell survival with an IC50 and IC90 of 8 and 30 μg/ml, respectively.

Observation of Flow cytometry analysis also revealed that treatment of β-2-himachalen-6-ol is linearly associated with volume injection, as the dose of 10 μg/ml HC significantly increased the number of cells undergoing late apoptosis (28%), while 25 μg/ml caused a larger cell shift towards late apoptosis (46.6%) and necrosis (39%).

#Phytochemical 3,3'-Diindolylmethane Inhibits the Risk of #ProstateCancer, According to Researchers

Scientists may have found a plan-based bioactive compound for the treatment of prostate cancer with no side effects, according to studies.
Prostate cancer is a medical condition caused by cell growth disorderly in the tissue of the prostate glands. Prostate cancer is one of the most slow-growing cancers that take years to develop.

The disease often is discovered during the rectum examination annually. Most cases of prostate cancer start in the cells on the surface of the inner lining of prostate tissue.

In the early stage, patients may not experience any symptoms. However, at the advanced stage, prostate cancer can induce symptoms of blood in the urine and semen, disruption of urinary passing, and frequent and urge urinary.

In severe cases, prostate cancer can also cause erectile dysfunction.

According to statistics, 1 man in 9 will be diagnosed with prostate cancer during his lifetime.

Most cases ( about 60%) of prostate cancer are found in older men, particularly in the African-American. The 5-year survival rate of local or regional prostate cancer is nearly 100.

3,3'-Diindolylmethane or DIM are phytochemicals derived from the digestion of indole-3-carbinol, belonging to the group of Indoles, found abundantly in broccoli, Brussels sprouts, cabbage, kale, etc.

Researchers on finding a natural compound for the treatment of prostate cancer examined the effect of 3,3'-Diindolylmethane (DIM) as tumor inhibitors in multiple malignant tumors on microRNAs (miRNAs).

Tetrachlorodibenzo-p-dioxin (TCDD) and 3,3'-Diindolylmethane (DIM) application showed a significant effect in the inhibition of proliferation and invasion of prostate cancer cells.

Furthermore, Tetrachlorodibenzo-p-dioxin (TCDD) and 3,3'-Diindolylmethane (DIM) treatment induced cancer death by inhibiting the expression level of CYP1A1, an enzyme associated with the activation of the compounds with carcinogenic properties.

In other words, Tetrachlorodibenzo-p-dioxin (TCDD) and 3,3'-Diindolylmethane (DIM) treatment activated the expression of MAP3K12 in the promotion of cancer cells by targeting the mRNA of miR-150-5p in prostate cancer cells.

Moreover, in order to reveal more information about DIM anti-prostate cancer activity, researchers examine the modulation of the immune system by cancer-protective food bio actives, indole-3-carbinol (I3C) and 3,3'-diindolylmethane (DIM).

Injection of indole-3-carbinol (I3C) and 3,3'-diindolylmethane (DIM), exerted prostate cell protective effect by the regulation of inflammatory responses in monocytes and macrophages of the immune system.

In other words, I3C and DIM inhibited the lipopolysaccharide (LPS), the major component of the outer membrane of Gram-negative bacteria in the induction of IL-1β mRNA associated with the immune response to infection and protein in the monocyte form of the white blood cells.


I3C and DIM also exerted anti-inflammatory activity by inhibiting the expression of gene CD84 mRNA in signaling T-cell activation by regulating the proteins associated with the expression of the gene.

Dr. Wang TTY and colleagues after taking into account co and confounders, wrote, "cross-talk between AHR(protein associated with regulating gene expression) and inflammation-mediated pathways, but not CD84-mediated pathways, in monocytes but not macrophages may contribute to the modulation of tumor environments by I3C and DIM in prostate cancer'.

The results strongly suggested that DIM modulated the immune function against the process of prostate cancer development and progression.

Taken altogether, DIM may be considered a supplement for the prevention and combined with primary medicine for the treatment of prostate cancer, pending the confirmation of a larger sample size and multicenter human study.


Intake of DIM in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.
