#Healthyfood #Onions Inhibit Motor Incoordination and Memory Deficits INduced by Oxidativestress, According to Studies

By Kyle J. Norton

Neuroprotection is a process to preserve the neuronal structure and/or function against the death or gradual death of neurons, including the use of medicines and herbs.

In other words, neuroprotection protects the neuronal integrity against the reduction in the rate of neuronal loss over time inducing neurological disease, including neurodegenerative disease.

Neurodegenerative disease is the class of conditions associated with the progressive loss of neurons occurred mostly in older adults, including dementia. The most common dementia in the US is Alzheimer`s disease (AD).

Alzheimer's disease is an irreversible, progressive brain disorder caused by the slowly lost of neuron in the brain that affects memory and thinking skills and, eventually, the ability to carry out the simplest tasks as the disease progress into an advanced stage.

According to the statistic, dementia affects over 47.5 million people worldwide, approximately 58 percent are living in low and middle-income countries.

Believe it or not, 5.8 million Americans of all ages are living with Alzheimer's dementia in 2019 and two-thirds of people with Alzheimer's disease are women.

Conventionally, as of today, there is no cure for dementia. Treatment of dementia is focusing to improve dementia symptoms and slow down the progression of the disease.

Some researchers suggested that the increased incidence of the onset of Alzheimer's disease over the past few decades may be correlated to the promotion of a high-fat diet in the US.

Dr. Rollins CPE, the lead scientist wrote, "HFD-induced obesity during adolescence exacerbated brain volume alterations by adult life in the 3xTg mouse model in comparison to control-fed mice and mediated volumetric alterations of select brain regions, such as the hippocampus".

And, "Further, HFD-induced obesity aggravated memory in all mice, lowering certain memory measures of B6129s control mice to the level of 3xTg mice maintained on a CD. Moreover, a decline in the volumetric trajectories of hippocampal regions for all mice was associated with the degree of spatial memory impairments on the MWM.".

The onion is a plant in the genus Allium, belonging to the family Alliaceae, a close relation of garlic. It is often called the "king of vegetables" because of its pungent taste and found in a large number of recipes and preparations spanning almost the totality of the world's cultures.

Depending on the variety, an onion can be sharp, spicy, tangy, pungent, mild, or sweet.

On finding a potential compound for the treatment of cognitive impairment, researchers examined the neuroprotective potential of Allium cepa (A. cepa, Onion) in aluminum chloride-induced animal model neurotoxicity.

The study included mice induced neurotoxicity by aluminum chloride (50 mg/kg/day) orally were treated supplemented with different doses of A. cepa hydroethanolic extract for a period of 60 days.

Before the treatment of onion, aluminum chloride-treated mice resulted in significant motor incoordination and memory deficits, observed by the increased oxidative stress and elevated levels of acetylcholinesterase (AChE) and aluminum levels in the brain.

Onion supplement injection in aluminum-exposed animals not only significantly improved muscle coordination and memory deficits but also decreased the levels of oxidative stress, and AChE, and decreased abnormal aluminum deposition induced by the aluminum chloride.

Decreased vacuolated cytoplasm associated with the death of neurons and pyramidal cells in the hippocampal area of the mice brain caused by toxicity was also reversed by the injection of A. cepa supplementation.

The results strongly suggested that onion processes anti-neuro-toxicity induced cognitive impairment in an animal model.

Dr. Singh T, the lead scientist, after taking other factors into account wrote, "significant neuroprotective potential of A. cepa in aluminum-induced neurotoxicity".

Taken altogether, onion may be considered a neuroprotective remedy, pending the confirmation of the larger sample size and multicenter human study.

#Bromelain Found in #Pineapple Reduces the Symptoms and Improves the Quality of Life in Osteoarthritis (OA) Patients, Researchers Say

By Kyle J. Norton

Osteoarthritis is an inflammation of joints caused by the depletion of the cartilage on the ends of the bones due to wear and tear over time.
The disease is one most common forms of arthritis that affect millions of people worldwide.

According to the statistics, in the US, approximately 10% of men and 13% of women aged 60 years or older have osteoarthritis (OA).

The most common risk factors found in patients with OA are older adults, female gender, overweight and obesity, knee injury, repetitive use of joints, and low bone density.

Besides pain in the joints, the other most common symptoms of OA include tenderness, stiffness, low flexibility, and grating sensation.

Conventionally, treatment of OA is focused on managing symptoms, by reducing pain, stiffness, and swelling through the use of painkillers, improving joint mobility and flexibility, and maintaining a healthy weight by exercise.

Some researchers suggested that the high prevalence of OA in the US has become a burden to the health care cost and economic burden.

Dr.Bitton R, the lead scientist at the Health Plan of Nevada wrote, "Approximately one-third of direct OA expenditures are allocated for medications, much of which goes toward pain-related agents. Hospitalization costs comprise nearly half of the direct costs, although these expenditures are consumed by only 5% of OA patients who undergo knee or hip replacement surgery".

And, "Indirect costs for OA are also high, largely a result of work-related losses and home-care costs".

Bromelain, a proteolytic enzyme found in pineapples (Ananas comosus) has been used in traditional medicine as an inflammatory agent and to treat pains, strains, muscle aches and pains and ease back pain and chronic joint pain, skin diseases, etc.

With an aim to find a natural compound for diseases associated with inflammation, researchers investigated the effects of bromelain in mild-to-moderate knee OA patients.

The randomized, single-blind, active-controlled pilot study included 40 knee OA patients randomized to receive oral bromelain (500 mg/day) or diclofenac (100 mg/day).

At week 4, both bromelain and diclofenac-treated groups showed an improvement in total Western Ontario and McMaster Universities Osteoarthritis Index (WOMAC) and pain subscales compared to the baseline.

According to the tested assays, bromelain and diclofenac groups exerted similar effects after 4 weeks.

At week 16 of bromelain treatment, patients had improved total WOMAC scores (12.2 versus 25.5), pain subscales (2.4 versus 5.6), stiffness subscales (0.8 versus 2.0), and function subscales (9.1 versus 17.9), and physical component of SF-36 (73.3 versus 65.4) as compared with baseline values.

In other words, bromelain reduced symptoms and improved quality of life in OA patients by inhibiting the levels of oxidative stress and overexpression of pro-inflammatory cytokines at week 16.

Based on the results, researchers after taking other factors into account wrote, "Our results show the inhibitory effect of enzyme therapy on collagen-induced arthritis in rats, as well as the efficacy of cyclosporin".

Taken altogether, bromelain may be considered supplement for the prevention and treatment of osteoarthritis (OA), pending the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#Antioxidant #Lycopenes Inhibtit Chemical and Natural Toxins That Cause Cellular DNA Damage, According to Studies

By Kyle J. Norton

Natural toxins are toxic chemicals produced by living organisms with the aim to protect themselves from predators. Therefore, these types of toxins do not cause harm to the host but can be very deadly to other organisms if they are consumed.
The most common types of natural toxins produced by plants are alkaloids such as pyrrolizidine alkaloids and glycoalkaloids, cyanogenic glycosides, lectins, saponins, and antinutrients.

Chemical toxins are chemicals or substances that not only are harmful to the environment but a hazard to human health if inhaled, ingested, or absorbed through the skin.

The most common types of chemical toxins are inorganic compounds such as mercury and carbon monoxide, and organic compounds, such as methyl alcohol.

The Institute of Natural Medicine suggested that exposure to chemical and natural toxins may cause* Enzyme dysfunction, including antioxidant enzymes that prevent the free-radical damage that causes damage to protein, lipid, and cell DNA.
* Reduced mineral absorption that can induce bone disease.
* Damage to the organs, particularly in the detox organs, such as the liver and kidney

* Damage cell's DNA and modify genetic expressions of the host that initiates the onset of cancer.

* Alternation of the body's hormones by inducing, inhibiting, mimicking, and blocking hormone production.

Lycopene is a phytochemical in the class of carotenoids, a natural pigment with no vitamin A activity found abundantly in tomatoes and other red fruits and vegetables, such as red carrots, watermelons, and papayas,

Tomatoes provide about 80% of the lycopene in the world's diet. In plants, lycopene protects the host against excessive photodamage and performs various functions in photosynthesis.

With an aim to protect the body against environmental, occupational, and therapeutic toxins, researchers examined the lycopene anti-toxic agent's activity.

A high intake of lycopene administration has been effective in decreasing the risk of both natural toxins including mycotoxins, bacterial toxins, and chemical toxins including heavy metals, pesticides as well as herbicides.

Phytochemical carotenoids processed by abundantly bioactive compound lycopene have also been found to process antioxidative activity against long-term overexpression of free radicals, antiapoptotic activity against cancer cells, radical scavenging activity against the initiation of free radical chain reaction, and chelating activity against chemical toxins.

Furthermore, lycopene also played a critical role in the modulation of inflammatory pathways that facilitate the onset of protein, lipid, and cellular DNA damage.

Based on the results, researchers said, " This review summarizes available data from several recent studies about lycopene and its role against chemical and natural toxicants".

Taken altogether, lycopene found in carotenoids may be considered supplements for the prevention and treatment of diseases associated with chemical and natural toxins, pending the confirmation of the larger sample size and multicenter human study.

Intake of lycopene in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#Enzyme #Bromelain In the Prevention of #OvarianCancer, According to Studies

By Kyle J. Norton

Ovarian cancer is a chronic condition characterized by irregular cell growth in the tissue of the ovary.

Most cases of ovarian cancer start in the cells on the surface of the inner lining of the tissue of the ovary.

In the very early stage of ovarian cancer, patients may not experience any specific symptoms. However, after penetrating into the deeper layers and increasing in size, ovarian tumors may cause symptoms of an abdominal mass, bloating, back pain, urinary urgency, constipation, pelvic pain, abnormal vaginal bleeding, and buildup of fluid in the abdominal cavity, accompanied with general symptoms shared by other types of cancer.

At the advanced stage, the cancer cells can also travel a distance away from the ovary to infect other healthy tissues and organs, leading to secondary metastasis.

Ovarian cancer is classified into 5 stages
1. Stage 0,
If the cancerous cell has not penetrated deeper tissue but into the surface of the ovarian lining.

2. Stage I
In stage I, The cancerous cells are no longer on the surface they have invaded into deep ovary lining, but still completely inside the ovaries.
a. Stage IA1
The cancer is not ≤ 3 mm (1/8 inch) deep and ≤ 7 mm (1/4 inch) wide.
a.1. Stage IA1:
The spreading is < 3mm(1/8 inch) deep and & less than 7mm (1/4 inch) wide.
a.2. Stage IA2: The invasion area is ≥ 3 mm but ≤ 5 mm (about 1/5 inch) deep and & less than 7 mm (about 1/4 inch) wide.
b. Stage IB:
Cancer in this stage has invaded the connective tissue, & less than 5cm (2 inches).
b.1. Stage IB1:
Cancer is 4 cm large (1 3/4 inches).
b.2. Stage IB2:
Cancer is ≥ 4 cm (1 3/4 inches) but & less than 5cm (2 inch)

3. Stage II
In stage II, the cancerous cells have spread to distant tissues but are still within the lung.

4. Stage III
In this stage, cancerous cells have spread to the tissues immediately surrounding the lung.

5. Stage IV
In this stage, cancerous cells have spread to the tissues immediately outside of the ovaries, other distant parts of the body

Bromelain, a proteolytic enzyme found in pineapples (Ananas comosus) has been used in traditional medicine as an inflammatory agent and to treat pains, strains, muscle aches and pains and ease back pain and chronic joint pain, skin diseases, etc.

With an aim to find a potential compound for the treatment of colon cancer, researchers evaluated the effect of a high quantity of bromelain from pineapple anticarcinogenic activity against A2780 ovarian cancer cell lines.

In order to prevent the homogenization, ultrafiltration, precipitation, and dialysis from contributing to heavy loss of bromelain, fresh pineapple juices from the flesh (PJ-F), core (PJ-C), and stem (PJ-S) were selected as a source of crude bromelain.

Bromelain concentrations of PJ-F, PJ-C, and PJ-S (1 μg/ml, 10 μg/ml, 100 μg/ml, and 1000 μg/ml) were selected for the study.

According to the tested assays, a 100 μg/ml concentration of bromelain showed exceptional efficiency in inhibiting the cancer cells without affecting the nearby normal cells.

Based on the basis of morphological alterations using fluorescent and inverted phase-contrast microscopes, all concentration-induced cancer cell death through apoptosis.

Additionally, the efficacy of fresh pineapple juices that kills cancer cells on A2780 cells was attributed to the inhibition of tumor colony-forming abilities.

In other words, PJ-F, PJ-C, and PJ-S effectively suppressed colony formation in cancer cells.

Based on the results, Dr. Madihah Binti Abdul Gani, the lead scientist said, "PJ-F, PJ-C, and PJ-S may have the potential to induce anticarcinogenic effects through apoptosis to A2780 and ... in vitro".

Taken altogether, bromelain found in fresh pineapple juices may be considered supplements for the prevention and treatment of ovarian cancer, pending the confirmation of the larger sample size and multicenter human study.

Intake of bromelain in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#Herbal #Burdock Exerts #Antibiotic-Like Activities with No Side Effects, Researchers Suggest

By Kyle J. Norton

Herbal Burdock may be the next pharmaceutical target to extract a single ingredient for the treatment of infectious diseases, some scientists found.
Infectious diseases are a class of diseases caused by the invasion of pathogenic microorganisms, such as bacteria, viruses, parasites, or fungi.

Bacteria is a type of biological cell consisting of a single cell with a simple internal structure.

The most common diseases caused by bacteria include strep throat, urinary tract infections, and tuberculosis.
Infection occurs when the bacteria successfully invade the body and start to multiply. Most cases of infectious diseases caused by bacteria are treated with antibiotics.

And, most cases of infectious disease are transmitted airborne you can get sick if you breathe into the air that carries infectious bacteria in air particles, vector-borne if you are stunk by the insect that the bacteria, and vehicle-borne if you are eating food or taking the materials contaminated by the bacteria.

Most cases of bacterial invasion are met with the first line of immune white blood cells. In the active division of bacteria in the acute phase of infection, the immune system response activated the production of the proinflammatory cytokine to the site of infection with an aim to kill off all bacteria.

However, if the immune fails to kill off all the bacteria within the 3 - 8 weeks period, it then will adapt to the change, leading to chronic inflammatory and infectious diseases.

Not all bacteria are bad, some play an important role in boosting immune function. Dr. Hsin-Jung Wu, the lead author in the study on the effect of gut microbiota in immune homeostasis and autoimmunity, said, " The gut microbiota that resides in the gastrointestinal tract provides essential health benefits to its host, particularly by regulating immune homeostasis" and " gut microbiota regulates innate and adaptive immune homeostasis, which in turn can affect the development of not only intestinal but also systemic autoimmune diseases:.

In other words, gut microbiota promotes and balances the immune system in eliminating invading pathogens, while still maintaining self-tolerance to avoid autoimmunity, the critical role of the immune system for the body’s health.

Burdock is a plant in the group of biennial thistles, genus Arctium, belonging to the family Asteraceae, native to the Euro.

The plant has been used over thousands of years in China and other traditional herbal medicine as a diuretic, diaphoretic, and blood-purifying agent and to treat wounds and infections stomach ulcers, and other digestive problems.

The ingredients of herbal Burdock include carbonated water, sugar,, manloid colorings, phosphoric acid, citric acid, and a good amount of dietary fiber, calcium, potassium, and amino acids and chemicals compounds of guaianolide, arctinone, arctinal, methyl arctate,11-tridecadien-3,5,7,9-tetraynde-hydrocostus lactone, dehydrodihydrocostus lactone, 3-octenoic acid, 3-hexenoic acid, etc.

In finding a natural ingredient or whole foods to replace the conventional antibiotics for the treatment of bacterial infection, the research team of infectious diseases at the University of Ribeirão Preto evaluated the antibacterial activity of a phytotherapeutic agent prepared from an ethyl acetate fraction (AcOEt) extracted from Arctium lappa.

The study includes 27 maxillary canines inoculated with a mixed bacterial suspension of Pseudomonas aeruginosa, Escherichia coli, Lactobacillus acidophilus, Streptococcus mutants, and Candida albicans.

The teeth were divided into three groups and their canals were filled with:
* Group 1: calcium hydroxide and propylene glycol;

* Group 2, a paste containing AcOEt fraction of A. lappa and propylene glycol; and

* Group 3, propylene glycol (control).

Observation of the 7, 14, and 30 days, when three teeth from each group were opened, researchers found that group 2 treated with the Burdock extract showed severe growth at 7 days, but no growth at 14 and 30 days, compared to mild growth in group 1 and control.

These results clearly indicated the phytotherapeutic agent extracted from an AcOEt fraction of the herbal Burdock process has a vigorous antibacterial activity against the growth of all the microorganisms.

In bacterial straits Enterococcus faecalis, Staphylococcus aureus, Pseudomonas aeruginosa, Bacillus subtilis, and Candida albicans were used for the investigation of the antimicrobial activity of rough extracts from leaves of Arctium lappa and their phases, researchers at the University of Ribeirão Preto (UNAERP) showed some bioactive compounds with chemical structures of different polarities were found to exert the effect of the extract through different phases of inhibition, depending on the target microorganisms.

Dr. Holetz FB, the lead scientist at the Universidade Estadual de Maringá in the investigation of the ethnobotanical use of the herbal species for the treatment of various infectious diseases, explained, herbal Burdock, one of the Brazilian folk medicine that may be used for the treatment of chronic infectious diseases induced by bacteria and yeasts.

Herbal Burdock was one of 13 herbal medicine studied for its antimicrobial activity against bacteria and yeasts, including Pseudomonas aeruginosa, and Escherichia coli.

The evidence showed that herbal Burdock processed with abundant bioactive constituents may be considered a functional herb for the prevention and treatment of infectious diseases without inducing any side effects

#Phytochemical #Indicaxanthin Inhibits the Formation of Plagues that Cause #Atherosclerosis, Researchers Say

By Kyle J. Norton

Atherosclerosis is a medical condition caused by plaque accumulated on the wall of arteries, leading to the narrowing of the artery and reducing oxygen risk blood flow to the heart and brain.
Depending on the location of the blockage, if the reduced blood flow occurred in the brain, it will cause the death of the brain cells, leading to stroke.

However, if the reduced blood flow occurred in the heart, it will cause damage to the heart tissues, leading to heart disease.

The most common disease associated with atherosclerosis is coronary artery disease. Plaques are made up of cholesterol, calcium, and other chemical substances in the artery

Most people with atherosclerosis are asymptomatic at the early stage.

Some researchers suggested that atherosclerosis is a lipoprotein-driven disease. Therefore, people with high blood cholesterol are at an increased risk of atherosclerosis. Overuse of calcium supplements is also found to elevate the condition risk, particularly in the elderly.

Other risk factors that have been found epidemiologically by studies are high blood pressure, triglycerides, smoking, insulin resistance, obesity or diabetes, and inflammatory diseases, such as arthritis, lupus, or infections.

Most cases of atherosclerosis can be reversed by making a change of lifestyle and moderate exercise.
Dr. Singh RB, the lead scientist wrote, "Adherence to diet and exercise was obtained through questionnaires, and information obtained was quantified into a formula. After 24 weeks, the overall score of diet and exercises were significantly higher in group A(intervention diet( than in group B(control). There was a significant decrease in serum total cholesterol (13.3%), low-density lipoprotein cholesterol (16.9%), triglycerides (19.2%), fasting blood glucose (19.5%), and blood pressures (11.5/6.2 mm Hg) in the intervention group compared with initial levels and changes in group B".

Indicaxanthin is a phytochemical in the class of red and yellow indole-derived pigments of betacyanins, belonging to the group of betalains, found abundantly in beets, chard, etc.

With an aim to find a natural compound for the treatment of atherosclerosis, researchers at the Università di Palermo investigated the effect of phytochemical indicaxanthin (Ild) a major compound found in beetroot on human atheromas.

7-Ketocholesterol (7-KC)-induced the development of human atheromas, according to the kinetic measurements within 24 h, before injection of indicaxanthin showed the early formation of intracellular reactive oxygen species over basal levels, preceding NADPH oxidase-4 (NOX-4) over-expression and elevation of cytosolic Ca²⁺, involved in reactive oxygen species (ROS) production. and associated with the progressive depletion of the antioxidant activity of total thiols.

Furthermore, 7-KC- also induced the expression of NF-κB (nuclear factor kappa-light-chain-enhancer of activated B cells) associated with the production of proinflammatory cytokines and innate and adaptive immune response.

Where 7-Ketocholesterol (7-KC) induced apoptosis of macrophages has been found to elevate the plaque progression and endoplasmic reticulum stress caused by the secretion of pro-inflammatory, procoagulant, and proteolytic molecules by macrophages.

Believe it or not, Co-incubation of 2·5 μm of indicaxanthin completely prevented 7-KC-induced pro-apoptotic events.

In other words, indicaxanthin protects the macrophage's integrity against apoptosis, thus reducing the risk of plaque accumulating on the wall of the arteries in facilitating the onset of atherosclerosis.

Based on the findings, researchers concluded, "The effects of Ind may be ascribed to inhibition of NOX-4 basal activity and over-expression, inhibition of NF-κB activation, maintaining cell redox balance and Ca homeostasis, with the prevention of mitochondrial damage and consequently apoptosis".

Taken altogether, phytochemical indicaxanthin (Ild) found in beetroot may be considered supplements for the prevention and treatment of atherosclerosis, pending the confirmation of a larger sample size and multicenter human study.

Intake of indicaxanthin (Ild) in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#Antioxidant #Helenalin Found in #Herbal #Arnica Exhibits the Immunoregulatory Activities, According to Studies

By Kyle J. Norton

Arnica may have a therapeutic effect in enhanced immune function, some scientists suggested.

The results of the analysis were conducted by a number of respectable universities, including the University Medical Center Freiburg, and published in several medical online literatures.

Arnica is a herbaceous species, genus perennial, belongings to the family Asteraceae, native to Europe and Asia, and used in herbal medicine for centuries to treat strains, sprains, and bruises.
Wound healing is a process of repairing the damage to the skin, tissues, organs, etc.

In the investigation to compare Anise tinctures and sesquiterpene lactones (SL), the secondary plant metabolites isolated form of Arnica, anti-inflammatory effects in inhibited expression of nuclear factor (NF)-kappaB activation, researchers found that application of Arnica tinctures and SL potently suppressed NF-kappaB activation and IL-12 production in dendritic cells at high concentrations but has immunostimulatory effects at low concentrations.

The nuclear factor NF-κB pathway is considered as a prototypical proinflammatory signaling pathway and IL-12 is an immune response signaling pathway in response to antigenic stimulation.

Furthermore, in contrast to general belief, the application of Arnica tinctures exerted an effect in suppressed contact hypersensitivity to sensitizer trinitrochlorobenzene and activation of dendritic cells.

These results indicated that herbal Anise inhibits the induction of contact hypersensitivity through its anti-inflammatorily immunosuppressive effect via immune regulation.

In impairing protein and DNA synthesis, injection of helenalin also displayed a deregulated and unwanted T cell-mediated immune responses, through induced apoptosis in activated CD4+ T cells, by triggering the mitochondrial pathway of apoptosis.

CD4+ T cells expression is a displaying CD4 molecule on their surface induced by glycoprotein found on the surface of immune cells in sending signals to T helper cells to initiate an immune pro-inflammatory response to the infectious site.

More precisely, helenalin deactivated the production of CD4+ T cells through inhibition of proliferation by G2/M cell cycle arrest accompanied by the accumulation of cell cycle regulatory proteins.

Further examination also found that helenalin injection enhanced the function of p53 in inhibited CD4+ T cell cycle division through its ability as a tumor suppression as well as increased expression of reactive oxygen species in facilitated cytotoxicity.

Moreover, helenalin also deactivated CD4+ T cell expression by suppressing nuclear translocation of NFATc2 with function in response to the activation of CD4+ T cells.

Dr. Berges C1, the lead author, at the end of the experiment said, "Helenalin can be defined as a new immunosuppressive compound suited for the treatment of deregulated and unwanted T cell-mediated immune responses".

The evidence suggested that Arnica and its bioactive compound helenalin may be considered as a functional herb in improved immune function. However, intake of Arnica extract and supplement should be taken with extreme care as toxicity was found in case of overdose.
