#Herbal #Arnica Inhibits the Gene Expression that Causes Colon Cancer, According to Studies

By Kyle J. Norton

Arnica may have a profound and therapeutic effect in ameliorated risk and treatment of colon cancer, some scientists suggested.

The results of the investigation were carried out by some respectable institutes, including the University of São Paulo and the University of Münster, and published in online medical literature.

Colon cancer is a medical condition characterized by cell growth irregularly and uncontrollably in the colon tissue. At the later stage, the cancerous cells may travel a distance away from the original site to infect other healthy tissue.

Arnica is a herbaceous species, genus perennial, belongings to the family Asteraceae, native to Europe and Asia, and used in herbal medicine for centuries to treat strains, sprains, and bruises.
Wound healing is a process of repairing the damage of the skin, tissues, organs, etc.

In the study to reconfirm the use of the technique in detecting the early stage of colon cancer through examining the expression of aberrant crypt foci (ACF) and colon-rectal mucosal epithelial cell proliferation researchers at the University of São Paulo conducted an experiment of 50 male Wistar rats randomly divided into 5 groups, as indicated below

* Groups G1 to G4 was given 4 injections of the carcinogen 1,2-dimethylhydrazine (DMH)

* The G2 group received Lychnophora ericoides (LE) extracts for 6 wk.

* The groups G3 and G4 received LE for 4 wk and 2 wk, respectively, at the postinitiation and initiation phases of colonic carcinogenesis. Group G5 was the control.

Observation on the 42nd day after the first injections of DMH for the neoplasia induction found that the Arnica treatment group (LE), including G2, 3, and 4 demonstrated a statistically significant decrease in the number of aberrant crypt foci (ACF) and a reduction of cell proliferation induced by DMH.

These indications suggested that Arnica consisted of a protective influence on the process of colon carcinogenesis, through suppressing both the initiation and the promotion of colonic carcinogenesis.

Furthermore, in the reexamine of the cytotoxic activity of phytochemical compound isolated from Arnica, against COLO 320, a human colorectal cancer cell line using the microculture tetrazolium (MTT) assay, researchers found that isolated flavonoids showed moderate to low cytotoxicity, in compared to the reference compound cisplatin at 2.9 microM.

Cisplatin is a conventional chemo medication used to treat a number of cancers.

In 2 hours of injection, sesquiterpene lactones, helenalin at the dose of 1.0 microM, possessing both the reactive alpha-methylene-gamma-lactone moiety and a reactive alpha,beta-unsubstituted cyclopentenone ring, displayed the strongest cytotoxicity against the tested colon cell line in compared to other compounds.

In comparison to tested GLC4, a human small-cell lung carcinoma cell line, COLO 320 was more sensitive to many flavonoids (especially for flavones) but more resistant to the cytotoxic effect of the sesquiterpene lactones.

Interestingly, in the evaluation of the Arnica effect in the deregulation of c-myb expression involved in the development of human tumors, especially certain types of leukemia and breast and colon cancer, researchers indicated that application of sesquiterpene lactones isolated from Arnica suppress Myb-dependent gene expression with potential anti-colon cancer activity with the IC50 values were in a range between 0.7 and >30 μM.

The C-myb is a member of the terminal domain with the function of regulating the expression of hematopoiesis, cell proliferation, and differentiation in tumorigenesis.

In comparison to other isolated bioactive compounds, furanoheliangolide goyazensolide and the pseudoguaianolide helenalin acetate with IC50 = 0.6 and 0.7 μM, respectively, represented the most active inhibitors of c-Myb dependent gene expression.

Observed by the control measurements for cell viability (MTS assay), researchers also found that c-Myb-dependent gene expression is not a function of cytotoxicity/unspecific cell damage.

All these results indicated that Arnica processed a significant effect in preventing early-onset, progression, and treatment of colon cancer through its bioactive compounds in the inhibited onset of aberrant crypt foci (ACF) and colon-rectal mucosal epithelial cell and colon cancer cells proliferation and gene expression in the risk of colon cancer.

#Herbal #Cascarasagrada Reduces the Risk of #PancreaticCancer in Vivo and Vitro, Scientists Say

By Kyle J. Norton

Herbal Cascara sagrada may process a major bioactive compound targeted by pharmaceutical companies for the development of conventional medicine in the treatment of pancreatic cancer, some scientists suggested.
Primary pancreatic cancer is cancer that originates in the pancreas as the result of the alternation of DNA of the cell.

Most cases of pancreas cancer start in the cell on the surface of the inner lining tissue. However, as the cancer progress, the malignant cells of the pancreas, although unusual may travel a far distance away to infect other healthy tissue and organs.

The pancreas cancer was found to be the site of metastasis from renal cell cancer, and lung, colon, and breast cancers.

Scientists do not know the exact causes of pancreatic cancers, but they do know several risk factors that can make a person more likely to get the disease, including the genetic and acquired genetic mutation of p16 and TP53 and KRAS, BRAF, and DPC4 (SMAD4).

Other prevalent risk factors, including long-term use of tobacco, exposure to certain chemicals, overweight, and obesity, and unpreventable risk factors of family history, gender, the increase of age, and ethnicity are associated with the early onset of pancreatic cancer.

However, most people with some of the above risk factors do not get the disease.

Researchers at the John Hoskins Medical Center suggested an unhealthy diet high in meats, cholesterol, fried foods, and nitrosamines may increase risk, while diets high in fruits and vegetables reduce the risk of pancreatic cancer.

In the change of lifestyle in the reduced risk of pancreatic cancer, the research team in the cancer program at the Ústav preventivního lékařství, after analyzing the total of 529 subjects, including 309 cases and 220 control subjects newly diagnosed patients with pancreatic cancer who lived in the region selected in hospitals in three centers (University Hospital Olomouc, University Hospital Ostrava, Hospital Ceske Budejovice) said, "Physical activity, dietary measures that will lead to weight loss and education to non-smoking can have a significant impact on the primary prevention of cancer."

These results suggested that if you follow a healthy diet and lifestyle with moderate exercise, your risk of pancreatic cancer is substantially lower compared to others.

Cascara sagrada is a species of buckthorn, genus Rhamnus, belonging to the family Rhamnaceae, native to western North America,

The herbal bark has been used in traditional medicine for gastrointestinal support. and it is thought to have a laxative and natural cleansing, etc., effect.

Besides the main compound of anthraquinones, other compounds of Cascara sagrada include anthraquinone glycosides, (emodin, frangulin, iso-emodin, aloe-emodin, and chrysophanol), rhein, aloin, malic acid, tannic acid, cascarosides A, B, C, and D, and hydroxy anthracene derivatives (HAD), etc.

In the reassessment of the effects of Emodin in inducing the apoptosis of pancreatic cancer cells and inhibiting tumor growth, researchers at the Zhejiang University School of Medicine conducted an investigation to examine pathways activated by Emodin during the anti-pancreatic cancer process.

In vivo and vitro, the application of Emodin up-regulated the function of cancer cell death programmer and down-regulated the expression of proteins that are responsible for the production of the new blood vessels to provide energy to the pancreatic cancer tumor.

Emodins also reduced the expression of eNOS phosphorylation in regulating the vascular angiogenesis which was promoted by the tumor observed by both immunohistochemistry and western blot analysis.

However, Emodin had no direct effect on VEGFR expression in vascular angiogenesis, in vivo

These results suggested that Emodin may be a potential anti-tumor effect on pancreatic cancer via apoptosis and suppression of angiogenesis.

Additionally, in the study to explore the role of XIAP and NF-κB in potentiating the antitumor effect of gemcitabine by Emodin in pancreatic cancer, researchers found that
* In Human pancreatic cancer cells SW1990, Emodin or their combination (gemcitabine plus emodin) induces cellular proliferation and apoptosis.

* The combination therapy more significantly inhibited SW1990 cell growth and induced a higher percentage of apoptosis, compared to those of Emodin treatment.

* Emodin or emodin plus gemcitabine downregulated the expression of XIAP and NF-κB similar to the above study, compared to the control group in vitro

* The combination of emodin and gemcitabine showed a more significant reduction in the T/NT ratio, an indication of the risk of cancer onset, SUV, and an indication of the response to cancer treatment and tumor weight compared to monotherapy.

* Emodin-enhanced tumor apoptosis induced by gemcitabine in vivo.

These results suggested that Emodin enhances the antitumor effect of gemcitabine in SW1990 pancreatic cancer in vitro and in vivo by downregulation of NF-κB expression, thus inhibiting the expression of XIAP.

Where X-linked inhibitor of apoptosis protein (XIAP) plays a key role in survival and also promotes invasiveness. in cancer cells.

Taken altogether, herbal Cascara sagrada processed with high levels of bioactive compound Emodin may be considered a functional herb for the prevention of pancreatic cancer and an adjunct therapy combined with primary medicine for the treatment of pancreatic cancer.

Further data collection large example size and multi-center studies performed with human consumption of the whole food or its bioactive compounds during the course of the disease will be necessary to complete the picture of herbal Cascara sagrada anti-pancreatic cancer possibilities.

#Antioxidant #Lycopene Is Inversely Associated with The Risk of #ChronicObstructiveLungDiseases (#COPD), Researchers Say

By Kyle J. Norton

The lungs are a pair of organs located on either side of the chest (thorax) with the function to bring fresh oxygen into the body and remove carbon dioxide and other waste gases from the body.

According to the statistics provided by the Lung Association over 3 million Canadians are affected by one of five serious respiratory diseases – asthma, chronic obstructive pulmonary disease (COPD), lung cancer, tuberculosis (TB), and cystic fibrosis.

In 2017, 28,600 Canadians were diagnosed with lung cancer, representing 14% of all new cancer cases. Lung cancer caused the death of 21,100 Canadians in the same year.

The exact causes of lung disease are not identified. However, epidemiological studies suggested
being overweight, smoking, having a family history, and smoking are some of the most prevalent factors associated with the onset of the disease.

The most common lung diseases in North America include asthma, chronic obstructive pulmonary disease (COPD), interstitial lung disease, pulmonary hypertension, and cystic fibrosis.

Symptoms of lung disease are totally depending on the type of disease.

The most common diseases associated with allergic airway inflammation are
* Asthma is a chronic inflammatory disease affecting the airway of the lung with recurring symptoms, such as wheezing, chest tightness, shortness of breath, and coughing. The disease affects people of all ages and mostly starts during childhood.

*Chronic obstructive pulmonary disease (COPD), the third leading cause of death in the US can be classified into
** Emphysema caused by damage to tissues of the air sacs (alveoli) in the lungs is a type of chronic obstructive pulmonary disease (COPD)associated with a long-term and progressive condition caused by shortness of breath but depending on the stage of lung function.

** Chronic bronchitis is a chronic inflammation of the lung’s bronchi that causes the increased production of mucus in the lung-induced symptoms of difficulty breathing.

Lycopene is a phytochemical found in tomatoes in the class of carotenoids, a natural pigment with no vitamin A activity found abundantly in tomatoes and other red fruits and vegetables, such as red carrots, watermelons, and papayas,

Tomatoes provide about 80% of the lycopene in the world's diet. In plants, lycopene protects the host against excessive photodamage and performs various functions in photosynthesis.

In finding a potential compound for the treatment of asthma researchers examined the effects of ascorbic acid (AA), carotenoids (lutein, zeaxanthin, β-cryptoxanthin, α-carotene, β-carotene, and lycopene), retinol and α-tocopherol levels in Japanese patients with chronic obstructive lung diseases (COPD).

Based on the tested analysis, compared to other phytochemicals, plasma AA level was significantly lower in COPD subjects than that in healthy elderly people.

Levels of plasma lycopene and total carotenoid levels associated with levels of antioxidant enzymes such as GSH content produced by the host were significantly lower in COPD subjects comapred to those of healthy elderly people without affecting the redox markers such as GSSG, GSH/GSSG ratio associated with the oxidative stress in the lung tissues.

Lycopene and total carotenoid levels did not lower the marker of the stories of urinary 8-OHdG detected in COPD subjects compared to healthy elderly people.

In other words, increased levels of lycopene and total carotenoid levels in the body may have a strong impact on reducing the risk of COPD

Based on the findings, researchers said, "COPD of Japanese patients may develop partly because of oxidative stress derived from a shortage of antioxidant nutrients, especially of AA and lycopene".

Taken altogether, lycopene found in tomatoes may be considered a remedy for the prevention of COPD, pending the confirmation of the larger sample size and multicenter human study.

Intake of lycopene in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#Antioxidant #Betanin Kills #Cancer Cells by Inducing #Oxidative-Cyoxicity Through Its Antioxidant Activities, Sdudies Suggest

By Kyle J. Norton

Colon cancer is a medical and chronic condition caused by the irregular growth of cells in the colon, due to the alternation of cell DNA.
Colon-formed part of the digestive tract, including the large intestine with the function to reabsorb fluid before passing them back to the blood and remove waste through the anus.

Colon cancer is one most slow cancer compared to other cancers. In some patients, colon cancer may exist in the colon for more than 20 years before it is discovered.

According to the statistic provided by the American Cancer Society, in 2019, approximately, 101,420 new cases of colon cancer will be diagnosed. Colon cancer also causes the death of 51,020 deaths. during 2019.

The lifetime risk associated with the onset of colorectal cancer is 1 in 22 (4.49%) for men and 1 in 24 (4.15%) for women.

Out of many risk factors involved in the onset of colorectal cancer, aging is one of the major culprits, according to epidemiological studies.

The median age diagnosed with colon cancer is 68 in men and 72 in women compared to the age of 63 of rectal cancer in both men and women.

However, over the past few decades, the incidence of colon cancer has been found to rise in the younger age groups, probably due to the promotion of a high-fat diet.

Dr. Robb Bird, the editor at the worldwide health wrote, "Poor diet is associated with about 80 percent of colorectal cancer cases. Even 20 years ago studies were attributing colon cancers to high-fat diets. Colorectal cancer is the third most common cancer in the United States with more than 130,000 cases reported every year. These cancers are a result of a combination of several causes including genetic, epigenetic, and environmental causes, such as diet. Hopefully, with more research, the exact pathways by which diet leads to cancer can be discovered. Once discovered, blocking these pathways can hopefully reduce the negative impact of a high-fat diet on colon cancer risk".

Betanin is a Phytochemical in the class of red and yellow indole-derived pigments of Betacyanins, belonging to the group of Betalains, found abundantly in beets, chard, etc.

Om finding a potential compound for the treatment of continuing colonic inflammation, an important risk factor in the development of colorectal cancer, researchers examined the phytochemical compound betanin isolated from beetroot (Beta vulgaris var. rubra) effects against colon cancer.

In patients with inflammatory bowel disease, such as ulcerative colitis (UC) and Crohn's disease (CD), administration of betanin exerted an inhibition against the oxidative DNA damage and induced apoptosis in neutrophils from the blood, compared to those obtained in colon carcinoma-derived Caco-2 cells.

In IBD patients' neutrophils, betanin treatment at the concentration of 100 μM for 24 h modulated the immune function by exerting anti-inflammatory activity, assessed by comet assay.

In Caco-2 cells, injection of H2O2 caused a 4-fold increase of DNA strand breaks in comparison to untreated cells, but pre-treatment with betanin reduced DNA damage in these cells.

Betanin also exhibited the protein associated with pro-apoptotic function accompanied by the loss of mitochondrial transmembrane potential in the energy transformation.

In other words, betanin promotes ROS cytotoxicity that leads to colon cancer cell death without harming the nearby cells through its antioxidant activity.

The findings strongly suggested betanin exerts anti-inflammatory and potentially cancer-preventive activity.

Taken altogether, betanin isolated from beetroot may be considered a supplement for the prevention and treatment of colon cancer, pending the confirmation of the larger sample size and multicenter human study.

Intake of betanin in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

#Healthyfood #Gingers Reduce the Symptoms of Nausea and Vomiting In Early Pregancy, Scientists Show

By Kyle J. Norton

Scientists may have discovered a pungent and kitchen spice that processes a significant effect in reducing nausea and vomiting with no side effects, according to some studies.

Nausea and vomiting are the natural protection of the digestive system reacted to many causes, including food poisoning, gastritis (inflammation of the stomach lining), an ulcer, or bulimia.

Morning sickness or nausea and vomiting are common experiences in pregnancy, affecting 70–80% of all pregnant women.

Nausea and vomiting that often develop by five to six weeks of pregnancy are the results of the rise of the hormone estrogen in the early stage of pregnancy. The syndrome may get even worse in women have a sensitive stomach which tries to adapt to the changes of pregnancy.

Morning sickness in the pregnancy may seem obvious in general population, however in a group of women with severe nausea and vomiting during pregnancy due to hyperemesis gravidarum (HG), an entity distinct from nausea and vomiting of pregnancy (NVP), may lead to significant maternal and fetal morbidity if untreated.

Dr.Noel M. Lee, the lead author at the University of Wisconsin School of Medicine and Public Health in the study " Nausea and Vomiting of Pregnancy" said, "Various metabolic and neuromuscular factors have been implicated in the pathogenesis of NVP and HG; however, their exact cause is unknown. Consequently, treatment of NVP and HG can be difficult as neither the optimal targets for treatment nor the full effects of potential treatments on the developing fetus are known".

Where hyperemesis gravidarum (HG) is a pregnancy complication characterized by severe nausea, vomiting, weight loss, and possibly dehydration.

Other causes of nausea and vomiting are due to prolonged period of exposure to stress and long-term fatigue.

Patients who are having chemotherapy are associated with an increased risk of nausea and vomiting because of the injection of chemo drugs can affect parts of the stomach and brain that detect toxic (poisonous) substances.

Ginger (Zingiber officinale) or ginger root, the second superfood used for thousands of years by mankind, is the genus Zingiber, belonging to the family Zingiberaceae, native to Tamil.

The pungent spice has been used in traditional and Chinese medicine to treat dyspepsia, gastroparesis, constipation, edema, difficult urination, colic, etc.

With an aim to clarify the effect of ginger extract in treating chemotherapy-induced nausea and vomiting (CINV), a joint study led by Taipei Medical University-Shuang Ho Hospital was conducted to determine whether ginger could be used to treat CINV, which was interpreted using the PICOS (patient, problem/population, intervention, comparison, outcome, study) framework, with P being patients who underwent chemotherapy.

After searching the studies in English from 2000 to 2017 databases. 2 on a total of 10 studies with complete data. was included in the meta-analysis using Comprehensive Meta-analysis.

Ginger exerted a significant effect in reducing the symptoms of nausea and vomiting with the odds ratio (OR) of 0.71

Further analysis also discovered that ginger could only reduce acute CINV in patients, particularly acute vomiting.

After taking additional ]assessment of the co and confounders, Dr. Chang WP the lead scientist said, "Ginger displayed significant efficacy with regard to controlling CINV in the experimental groups".

In the reaffirming the effect of ginger in the early pregnancy symptoms of nausea and vomiting of pregnancy (NVP), a joint study led by the Iran University of Medical Sciences launched an evaluation to compare the effects of ginger, pyridoxine (vitamin B6), and placebo.

The triple-blind clinical trial on pregnant women suffering mild to moderate NVP between 6 and 16 weeks of pregnancy.

In these women ginger, 500 mg twice daily, vitamin B6 40 mg twice daily, and placebo twice daily were administered for 4 d.

The severity of NVP was evaluated 24 h before entering the study and up to 4 d after using medications and results were compared among the three groups.

The women of the three groups did not have significant differences according to age, gestational age, parity, and severity of each symptom before treatment and educational status.

After assessing the returned questionnaires, researchers indicated
* Ginger and vitamin B6 could reduce the severity of all items of the Rhodes questionnaire significantly.

* Ginger and vitamin B6 were more effective than the placebo

* total score of Rhodes did not show a significant difference between ginger and vitamin B6 (p = .128). Ginger was more effective for nausea (intensity and distress).

These results once against illustrate the efficacy of ginger in the treatment of nausea and vomiting in early pregnancy.

Taken together, there is no doubt that ginger may be used as a functional food for the treatment of nausea and vomiting in both early pregnancy and the side effects of chemo treatment.

#Antioxidant #Allicins Process Anti-#GastricCancer Activities in Vitros, According to Studies

By Kyle J. Norton

Gastric cancer is a condition caused by abnormal growth of the mucus-producing cells of the inside lining of the stomach. Adenocarcinoma is the most common type of stomach cancer.
At the later stage, liver cancer cells may travel a distance away from the liver to infect other healthy tissue, leading to secondary metastasis through blood and fluid circulation.

According to the American Cancer Society, every year, approximately 26,000 cases of stomach cancer are diagnosed (16,000 in men and 9,000 in women). Cancer causes the death of over 10,000 people, including 6,000 men and 4,000 women).

The cause of gastric cancer is a result of the alternation of cell DNA in the stomach. Researchers do not know what causes the cell DNA alternation, but they do know certain risk factors such as smoking, family history, genetic preposition, certain diseases associated with the stomach, and previous stomach surgery are involved in the onset of the disease.

However, some researchers suggested that infection caused by Helicobacter pylori is the most important risk factor for gastric cancer.

Dr. Lydia E. Wroblewski, the lead author examined the effects of Helicobacter pylori on the risk of gastric cancer and wrote, "Helicobacter pylori is a gastric pathogen that colonizes approximately 50% of the world's population. Infection with H. pylori causes chronic inflammation and significantly increases the risk of developing duodenal and gastric ulcer disease and gastric cancer".

And, "Infection with H. pylori is the strongest known risk factor for gastric cancer, which is the second leading cause of cancer-related deaths worldwide".

Allicin is phytochemical containing sulfur in the class of organosulfur compounds, found abundantly in onion and garlic.

To find the natural compound for treating gastric cancer with no side effects, researchers evaluated allicin, an active compound derived from garlic with anti-cancer effects.

Allicin injection reduced cell viability in a dose- and time-dependent manner, partly through the induction of apoptosis in gastric cancer cells.

At the molecular level, allicin induced cancer death through the degradation of cancer cells through activation of the proteins associated with the stimulation of cell apoptosis, including the caspase-3, -8, and -9 and Bax and fas expression.

Allicin treatment also inhibited proliferation and induced apoptosis in SGC-7901 cancer cells.

Dr. the lead researcher said, "Data from the current study demonstrated that allicin should be further investigated as novel cancer preventive or therapeutic agent in control of gastric cancer, with potential uses in other tumor types".

In a total of patients with progressive gastric adenocarcinoma, whose diagnosis was by gastroscopy and pathological examination assigned to 2 groups, 40 in each group, researchers examined the effects of local application of allicin via gastroscopy on progressive gastric carcinoma.

Allicin group showed the cell apoptosis rate of 9.60 +/- 1.52%, the percentage of cell in G0/G1 phase of 72.12 +/- 8.35%, in G2/M phase 9.54 +/- 3.20%, and PI 27.80 +/- 8.35, compared to the rate of 2.20 +/- 0.58%, 69.56 +/- 5.15%, 13.20 +/- 3.05%, and 30.40 +/- 5.15 in control.

The efficacy of allicin was attributed to the inhibition of the function of a protein (gene Bcl-2) associated with cancer cell anti-apoptosis and the exhibition of the function of proteins (gene Bax and Fas) associated with the pro-apoptosis.

These results suggested that local application of allicin via gastroscopy can inhibit the cell growth and proliferation of progressive gastric carcinoma. They can also promote gastric carcinoma cell apoptosis, according to the final report by Fourth Affiliated Hospital of Hebei Medical University.
