#Bromelain Kills #Cholangiocarcinoma Cells in Vitro, Researchers Suggest

Cholangiocarcinoma, also known as bile duct cancer, is a medical condition that originated from irregular cell growth in the bile ducts.
Most cases of cholangiocarcinoma are caused by the alternation of cell DNA in the cell on the surface of the inner lining of the bile duct.

At the advanced stage, cancer cells in the bile duct may travel a distance away to infect other healthy tissues and organs through the circulation of blood and fluids.

Primary bile duct cancer has been found to spread to the regional lymph nodes and adjacent organs.

According to statistics, cholangiocarcinoma is rare cancer that occurred approximately 2500 Americans each year compared to 5000 cases of gallbladder cancer and 15,000 cases of hepatocellular cancer.

Believe it or not, the 5-year survival rate diagnosed with early-stage extrahepatic bile duct cancer is only 30%. If cancer has spread to the regional lymph nodes, the 5-year survival rate is 24%.

The most common type of cholangiocarcinoma found in the bile duct is perihilar cholangiocarcinoma, followed by the distal extra-hepatic and then the intrahepatic forms.

Epidemiologically parasitic infections, primary sclerosing cholangitis, biliary-duct cysts, hepatolithiasis, and toxins are some prevalent factors that cause the onset of cholangiocarcinoma.

Dr. Gia L. Tyson, the lead scientist wrote, "Cholangiocarcinoma is the second most common primary hepatic malignancy after hepatocellular cancer. It accounts for approximately 10–25% of all hepatobiliary malignancies".

And, "Other less-established, potential risk factors include inflammatory bowel disease, hepatitis C virus, hepatitis B virus, cirrhosis, diabetes, obesity, alcohol drinking, tobacco smoking, and host genetic polymorphisms".


However, most people with some of the above risk factors do not develop cholangiocarcinoma.

Bromelain, a proteolytic enzyme found in pineapples (Ananas comosus) has been used in traditional medicine as an inflammatory agent and to treat pains, strains, muscle aches and pains and ease back pain and chronic joint pain, skin diseases, etc.

With an aim to find a natural ingredient against Cholangiocarcinoma (CC), researchers investigated the anticancer effect of bromelain and papain in intra- and extrahepatic human CC cell lines.

According to the medical assays, bromelain and papain showed a similar effect inhibition of human CC cell growth, migration, and invasion.

Furthermore, according to cell proliferative assays, bromelain, and papain resulted in a decrease in the proliferation, invasion, and migration of CC cells.

Bromelain and papain-induced CC death by inhibiting protein-associated anti-apoptotic signaling as well as their downstream signaling proteins associated with CC proliferation.

Interestingly, compared to papain, bromelain showed an overall more effective inhibition of CC.

In additional differentiation of the anti-cancer activity, researchers found that bromelain and papain have cytotoxic effects on human CC cells.

Based on the findings, Dr. Müller A the lead scientist said, "Bromelain and papain have cytotoxic effects on human CC cell lines, and bromelain and partially papain in comparison impair tumor growth by NFκB/AMPK signaling".

And, " Especially bromelain can evolve as a promising, potential therapeutic option that might open new insights for the treatment of human CC".

Taken altogether, bromelain may be considered a supplement for the prevention and treatment of cholangiocarcinoma, pending the confirmation of the larger sample size and multicenter human study.

Intake of bromelain in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Fennel may be the next natural antibiotic used for the treatment of diseases caused by bacterial infection with no side effects, some researchers suggested.
Although most micro bacteria are harmless, some are pathogenic.

The most common bacterial pathogens that cause human diseases are Staphylococcus aureus and group A β-hemolytic streptococci.

According to statistics provided by the World Health Organization, approximately, 50,000 men, women, and children are dying from infectious diseases every day worldwide.

Some researchers suggested that over the span of decades or centuries, the epidemiological transition provides the central narrative of global health, including a reduction in mortality and the creation of larger and older populations in which the main causes of illness are chronic infectious diseases.

Although in comparison to the third world, the prevalence of infectious diseases in the UK has reduced dramatically over the last century, the rate of infectious diseases caused by pathogen still has a strong and negative impact on major health, social and economic issues.
Each year in the UK, infectious diseases were found to cause 576 of days lost due to disability, out of 1000 population, according to statistics reported by Somerset Intelligence.

The most common symptoms of infectious diseases include fever, diarrhea, fatigue, tiredness, muscle aches,..... coughing. However, signs and symptoms may vary, depending on the pathogen, causing the infection.

Fennel is a species of Foeniculum Vulgare, belonging to the family Apiaceae (Umbelliferae), native to the shores of the Mediterranean. It is now widely cultivated all around the globe to use as food and herb.

In the study to examine 3 different essential oils from grown cultivars of Egyptian fennel, researchers found that according to chromatography/mass spectrometry analysis, all chemical compounds isolated from 3 essential oil processing a strong antioxidant activity in inhibited pathogenic bacteria including two species of fungi, two species of Gram-negative and two species of Gram-positive bacteria.

Further analysis also suggested that the efficacy of compounds isolated from essential oil from 3 species of fennel was attributed to its free radical scavenging, anti-lipid peroxidation, and metal chelating properties.

Additionally, in the observation of essential oil's antioxidant activity from Foeniculum vulgare commercial aerial parts and fruits isolated by hydrodistillation, researchers at the Universidade do Algarve conducted an experiment, using different distillation times (30 min, 1 h, 2 h, and 3 h), and analyzed by GC and GC-MS in isolated chemical compounds from obtained fennel essential oil.

According to the analysis of the agar diffusion method. trans-Anethole (31-36%), alpha-pinene (14-20%) and limonene (11-13%), the main components of the essentials oil isolated from F. vulgare dried aerial parts and exerted a significant antibacterial activity compared to methyl chavicol (= estragole) (79-88%) dominant in the fruit oils.

In the DPPH method, according to the measurement of oxidative stress, plant essential oils showed better antioxidant activity compared to fruit oils.

Dr, Miguel MG, the lead author wrote at the end of the experimental report, "None of the oils showed a hydroxyl radical scavenging capacity > 50%, but they showed an ability to inhibit 5-lipoxygenase. The essential oils showed very low antimicrobial activity. In general, the essential oils isolated during 2 h were as effective, from the biological activity point of view, as those isolated during 3 h".

These results suggested that the effect of chemical compounds in inhibited pathogenic bacteria is isolated time dependable. Only those isolated less than 2 hours are processed strong antibacterial capability.

Hypertension or high blood pressure, a condition associated with the cluster of metabolic syndrome is a medical condition associated with abnormally high blood pressure.
Blood pressure is the force of blood pushing against the walls of the arteries as the heart pumps out blood and changes constantly depending on activity. However, persistent hypertension due to poor management over time can lead to cardiovascular disease.

According to the statistics provided by the American Heart Association., nearly half of all adults in the United States are living with high blood pressure, or 1 in 3 Americans are hypertensive.

Hypertension is the leading risk factor for heart disease and stroke in the US.

Cardiovascular diseases (CVD) including heart disease and stroke are a class of conditions associated with the dysfunction of the arteries and heart, affecting the heart functioning.

According to the statistics provided by the CDC, heart disease is the leading cause of death for both men and women. Men are associated with 1/2 of the deaths due to heart disease in 2015 in men, compared to women.

Believe it or not, more than 63,000 Americans die from heart disease each year or 1 in every 4 deaths are caused by heart disease.


The most common prevalent factors associated with hypertension onset are family history, aging, being male, being overweight or obese, ethnicity, physical inactivity, unhealthy diet, and medical condition such as chronic kidney disease.

Conventionally, blood pressure medications have been found to prevent many deaths from heart disease and stroke over the past 30 years, however, long-term intake of such medications has caused many side effects such as damaging the kidney.

Some researchers suggested by making changes to lifestyle, maintaining an ideal weight, and diet with low salt, you could do just fine without the drugs.

Dr. L. J. Beilin wrote, "Lifestyle factors are critical determinants of blood pressure levels operating against a background of genetic susceptibility. Excess body fat is a predominant cause of hypertension with additive effects of dietary salt, alcohol, and physical inactivity."
And "Controlled trials in hypertensives show blood pressure lowering effects of supplemental potassium, fiber, n-3 fatty acids, and diets rich in fruit and vegetables and low in saturated fats".

With an aim to find a natural food for the prevention and treatment of hypertension, researchers examined the effect of daily supplementation of kale juice can modulate blood pressure (BP).

The study included 84 subclinical hypertensive patients with systolic BP over 130 mmHg or diastolic BP over 85 mmHg who received 300 ml/day of kale juice for 6 weeks.

According to the blood samples obtained on 0-week and 6-week, kale supplements showed a significantly decreased systolic and diastolic blood pressure in all patients regardless of their GSTM1 or GSTT1 polymorphisms.

According to a joint study led by the Isfahan University of Medical Sciences, the GSTT1 and GSTM1 gene increases arterial pressure; hence, it can predict susceptibility to cardiovascular disease.

In other words, kale supplements inhibited the hyperactive parameters without being affected by the gene expression of GSTM1 or GSTT1 polymorphisms.

Furthermore, kale supplements exerted no effect on the plasma lipid profiles of patients with GSTM1-null and GSTM1-present genotypes.

More importantly, the supplement also increased plasma HDL-C and lowered LDL-C in the GSTT1-present type, while blood glucose was decreased only in the GSTT1-null genotype.

Based on the findings, researchers suggested, kale processes a strong effect in lowering blood pressure through other mechanisms, regardless of GSTT1 and GSTM1 gene types.

Taken altogether, kale juice may be considered a supplement for the prevention and treatment of hypertension, pending the confirmation of the larger sample size and multicenter human study.

Eggplant may have a potential and substantial effect in reduced risk and treatment of prostate cancer, some scientists suggested.

Eggplant is a species of S. melongena with deep purple color, belongings to the family Solanaceae and native to India, cultivated in southern and eastern Asia since prehistory.

Prostate cancer is a medical condition characterized by irregular cell growth in the tissues of the prostate. At the later stage, the cancerous cell may invade other healthy tissues and organs a distance away from the original site.

Indeed, in the human prostate cancer cell PC-3. administration of a non-toxic dose of α-solanine initiated cancer suppression and reduced the viability of the human cancer cell through significantly elevated epithelial marker E-cadherin gene expression in target mutated cells in the induction of cell proliferation and invasion and decreased mesenchymal marker vimentin expression in initiated cell metastasis.

Further differentiation, also suggested that α-Solanine ameliorated cancer cell proliferation and tumor initiation through reduced mRNA levels of matrix metalloproteinase-2 (MMP-2) and 9 (MMP-9) in stimulated cancer invasion and metastasis and increased the expression of reversion-inducing cysteine-rich protein with kazal motifs (RECK) in suppression of prostate tumor progression and tissue inhibitor of metalloproteinase-1 (TIMP-1) and TIMP-2 in the regulation of MMP activity, particular in the induction of cancer cell overexpression.

According to the observation of the Immunoblotting assays, injection of α-solanine is effective in suppressing phosphorylation of phosphatidylinositide-3 kinase (PI3K) which plays an important role in modulated cellular functions such as cell growth, proliferation, differentiation, motility, survival and intracellular trafficking involved cancer progression, Akt phosphorylates expression with primary functions is to promote cell growth and protein synthesis and The extracellular signal-regulated kinase (ERK) signaling pathway phosphorylates with function in the control of diverse cellular processes including such as proliferation, survival, differentiation, and motility.

Moreover, α-solanine reduced levels of microRNA-21 (miR-21) in the expression of cancer progression. Higher levels of miR-21 are correlated to more advanced malignancies and elevated tumor suppressor miR-138 expression in induced prostate cancer cells apoptosis through cell cycle arrest at G1/S transition.

Taken together, eggplant with abundant phytochemicals including glycoalkaloids derivatives α-solamargine and α-solasonine may be considered as a functional food in reduced risk and treatment of prostate cancer.

In other words, over time, increased intake of onion may have significant activity in reducing prostate cancer risk.

The results strongly suggested, over time, regular consumption may decrease the risk of prostate cancer, depending on the amount of onion intake daily.

Taken altogether, onion used alone or combined with conventional medicine may be considered a remedy for the prevention and treatment of adjuvant rheumatic diseases, pending the confirmation of the larger sample size and multicenter human study.

A fatty liver is a liver condition associated with the accumulation of excess fat in the liver.
Non-alcoholic fatty liver (NAFL) is the early stage of nonalcoholic steatohepatitis (NASH) caused by the accumulation of fat in the liver, other than excessive use of alcohol.

Nonalcoholic steatohepatitis (NASH) is a type of inflammatory non-alcoholic fatty liver disease caused by excessive fat accumulated in the liver.

Nonalcoholic steatohepatitis (NASH) is asymptomatic until the condition is developed into cirrhosis, the condition that causes advanced liver scars and reduced the performance of the liver.

In other words, if more than 5% – 10% percent of your liver’s weight is fat, you have a fatty liver (steatosis).

According to the statistics, non-alcoholic fatty liver disease (NAFLD) is one of the most common causes of liver disease affecting 100 million people in the US. Out of that number, approximately 20% of people have NASH.

Medical conditions such as obesity, metabolic syndrome, and type 2 diabetes are some of the metabolic risk factors that have been found to induce the development of NASH.

As of today, there is no effective treatment for NASH. Depending on the condition people with overweight or obese are recommended to lose weight. The aim of medical treatment is to reduce the symptoms and delay the progression of the condition and improve the quality of life.

Out of many prevalent factors that cause the onset of NASH, some researchers suggested that the promotion of a high-fat diet over the past few decades induced a high percentage of the overweight and obese population may be the major culprit associated with the rise of NASH.

Dr. Jensen VS, the lead scientist wrote, "Hepatic dysfunction and inflammation were confirmed in HFD-fed rats by elevated levels of hepatic MCP-1 (p < 0.0001), TNF-alpha (p < 0.001) and plasma β-hydroxybutyrate (p < 0.0001), and in NASH-fed rats by elevated levels of hepatic MCP-1 (p < 0.01), increased hepatic macrophage infiltration (p < 0.001), and higher plasma levels of alanine aminotransferase (p < 0.0001) aspartate aminotransferase (p < 0.05), haptoglobin (p < 0.001) and TIMP-1 (p < 0.01) compared to Control".

And, " dietary fat and cholesterol are the primary drivers of NAFLD development and progression in rats, while fructose mostly exerts its effect on the circulating lipid pool".

Betaxanthins are phytochemicals in the class of red and yellow indole-derived pigments, belonging to the group of Betalains, found abundantly in beets, Sicilian prickly pear, etc.

In finding a potent ingredient for the treatment of liver disease, researchers examined the protecting effects of bioactive agents of the lyophilized table beet and carrot powder on fatty liver.

The shortened-term study included male Wistar rats fed with chow with or without high fat (2% cholesterol, 0.5% cholic acid, 20% sunflower oil) and treated with 0.1 or 1 g/bwkg/day natural product for ten days parallel with the feedings.

Based on the tested analysis, compared to other feedings, the higher dose of the natural product better decreased the induced free radical reactions, cyclooxygenase-2, and proteins associated with the production of pro-inflammatory cytokines in normal and fatty liver tissues.

More precisely, the natural treatment of beet and carrot powder inhibited liver oxidative stress by inhibiting the inflammatory expression without causing significant changes in all global antioxidant parameters.

However, the antioxidant activity of mobilized methyl group concentrations associated with liver stress also increased by fatty liver tissues.

Based on the evidence, researchers said, "Beet and carrot proved to be a "functional food" in rats with alimentary fat-induced fatty liver. It cannot be ruled out that this beneficial effect may have clinical relevance".

Taken altogether, beet and carrot may be considered functional remedies for the prevention of fatty liver disease, pending the confirmation of the larger sample size and multicenter human study.

Cancer is a class of diseases associated with irregular cell growth in the tissue of a specific organ due to the alternation of cell DNA.

Compared to normal cells with a limited cell cycle division before replacing by a set of new cells, malignant cells have an unlimited cycle division. In other words, cancerous cells ignore cell division inhibitors.

Primary colon cancer is a medical condition caused by abnormal cell growth in the colon tissue. Most cases of colon cancer begin on the surface of the inner lining tissue of the colon, before penetrating into deeper layers to form a tumor through proliferation.

At the advanced stage, the cancerous cells can dispatch from the original site to infect other healthy tissue and organs far away from the colon, leading to secondary metastasis.

The exact causes of colon cancer are unknown. researchers do not know why people with the same health condition, family history, and diet, are susceptible to the onset of the disease while others do not.

According to the statistic, colorectal cancer(cancer that starts in the colon or rectum is the third most commonly diagnosed cancer and the second leading cause of cancer death in men and women combined in the United States.

The Colorectal Cancer Alliance suggested, on average, the lifetime risk of developing colon cancer is about one in 23 for men and women combined (4.5%), however, this varies widely according to individual risk factors.

Out of many risk factors associated with the onset of colon cancer, some researchers suggested that the risk of colon cancer may have a direct implication from the promotion of a high-fat diet over the past few decades

Dr. Milly Ryan-Harshman wrote, "Whether red meat is a culprit in causing CRC remains unanswered, although any effect it might have is likely moderate and related to processing or cooking"

And "The effect of dietary fiber on the risk of CRC has also been difficult to determine because fiber intake is generally low. Evidence that folic acid, calcium, and vitamin D reduce the risk of CRC is stronger"

These results strongly suggested that by reducing the intake of red meat and processed foods, and increasing the intake of nutrients from fruits and vegetables including folic calcium and vitamin D, the risk of colorectal cancer can be substantially decreased.

However, in the study conducted by the team led by Dr. Dana-Farber, Cancer Center oncologist Jeffrey Meyerhardt, MD appeared in the Aug. 15 issue of the Journal of the American Medical Association, the Western diet not only associated with the colon cancer risk but also elevate the risk of recurrent colon cancer in colon cancer patients.

The small, white navy bean, also called pea bean or haricot, popular in both dry and green forms, is very popular in Britain and the US, native to Peru. It is now grown and consumed across the world, due to its numerous health benefits.

With an aim to find a natural and dietary compound for the treatment of colon cancer, researchers examined the effects of navy beans on mice-induced colon cancer.

The study included cooked navy beans (whole beans), the insoluble fraction (bean residue) or soluble fraction of the 60% (vol: vol) ethanol extract of cooked navy beans (bean extract), or a modified AIN-93G diet (16.6% fat including 12.9% lard) as control fed to 160 male obese ob/ob mice after 2 azoxymethane injections.

According to the tested assays mice fed with navy beans showed a reduction of dysplasia, adenomas, or adenocarcinomas in fewer mice compared to control.


More precisely, whole beans 54%, P=0.10; bean residue 81%, P=0.003; bean extract 91%, P=0.007), inhibited all types of colon lesions, including focal hyperplasia compared to control.

Based on the findings researchers wrote, "suggest that both the soluble and the insoluble fraction of the extract contribute to the cancer-protective effect of cooked navy beans".

In order to reveal additional information about navy bean anti-colon cancer activity, researchers evaluated whether black beans and/or navy beans would reduce colon carcinogenesis in rats.

The study included rats fed a modified AIN-93G diet (control) or diets containing 75% black beans or 75% navy beans for 4 wk before colon cancer was initiated by the administration of two injections of azoxymethane 1 wk apart.

At 31 wk after the second injection, the incidence of colon adenocarcinomas was significantly lower in navy bean (14%) diets compared to rats fed the black bean (9%) diets and control diet (36%).

Furthermore, total tumor multiplicity was also significantly lower in rats fed the navy bean (1.0) diets compared to the black bean (1.1) and control diet (2.2).

Based on the additional analysis, researchers found that the 44-75% reduction in colon carcinogenesis in rats fed beans was attributed to more controlled appetites and much greater concentrations of butyrate in the distal colon.

The results strongly suggested eating....navy beans significantly lowered colon cancer incidence and multiplicity.

Taken altogether, the navy bean may be considered a remedy for the prevention and treatment of colon cancer, pending the confirmation of the larger sample size and multicenter human study. larger sample size and multicenter human study.
