Bromelain Inhibits the Parameters Associated with The Onset of Alzheimer's disease

 Alzheimer's disease (AD) is a neurodegenerative disease correlated with major reductions of neurons to the respective target areas, named after German physician Alois Alzheimer.

Alzheimer's destroys brain cells, causing cognitive modalities severe enough to affect language communication, memory, lifelong hobbies or social life. Alzheimer's gets worse over time, and it is fatal.

According to statistic, over 25 million people in the world today are affected by dementia and most are suffering from Alzheimer's disease. In Canada, approximately, over half a million Canadians living with dementia accompanied by the 25,000 new cases diagnosed every year.

Believe or not, in 2016, dementia had cost Canada over $10 billion, including the health-care system and out-of-pocket caregiver costs.

Epidemiologically, cigarette smoking, midlife high blood pressure and obesity, diabetes, and cerebrovascular lesions and psychosocial factors (eg, high education, active social engagement, physical exercise, and mentally stimulating activity) are associated with prevalent risk of AD  developing.

Some researchers in the evaluated the increased AD in the older adults in the Western world over the past few decades may be correlated to the promotion of the high-fat diet.

Dr. Peter Thériault, the lead researcher wrote, "WD promotes systemic inflammation by increasing the levels of blood-circulating monocytes and monocyte chemotactic protein-1 production, which is accompanied by an augmentation of oxidized-low density lipoprotein levels in blood circulation".

And, "At the blood-brain barrier (BBB), WD potentiates the age-induced increase of Aβ 1-40 accumulation and exacerbates the oxidative stress, specifically in cerebral microvasculature. These effects were accompanied by the dysfunction of pericytes, thus altering BBB functionality without compromising its integrity".

In other words, if you are high-diet follow, you are at high risk of AD developing.

Bromelain, a proteolytic enzyme found in pineapples (Ananas comosus) has been used in traditional medicine as an inflammatory agent and to treat pains, strains, and muscle aches and pains and ease back pain and chronic joint pain, skin diseases, etc.

On finding a potential compound for the treatment of Alzheimer's disease, researchers investigated the ability of bromelain, a plant cysteine protease derived from pineapple stems interaction with synthetic Aβ42 monomers and oligomers.

According to the tested assays, bromelain protected the neuron successfully by degrading Aβ42 monomers and low and high molecular weight oligomers.

Examined the effects of bromelain preparations of 3 and 6 mU added to the cerebrospinal fluid (CSF), the levels of Aβ42 monomers and oligomers were significantly reduced, compared to that od the same standard Aβ42 preparations incubated in CSF without bromelain

Furthermore, the concentration of bromelain incubations with 0.1, 0.5, and 1 mU/ml were not toxic to the central nervous system.

Based on the findings, researchers said, "these results represent an important insight into the action of bromelain on Aβ42 oligomers, suggesting its potential use in the therapy of AD".

In order to know more information about bromelain anti-Alzheimer's disease activity, researchers examined a beta-degrading activity of diverse alpha 2-macroglobulin (alpha 2-M)-proteinase including trypsin, alpha-chymotrypsin, and bromelain (125)I-A beta 1--42.

The complex showed a strong degradation of (125)I-A beta 1--42.

In Ex vivo, oral administration of high doses of proteinases displayed an enhanced degradation of (125)I-A beta 1--42 in the blood of volunteers.

In other words, the complexes exhibited a significant A beta catabolism, thus reducing the risk of developing AD by preventing A beta accumulation in brain and vasculature.

Taken altogether, bromelain used alone or combined with other herbal medicines may be considered supplements for the prevention and treatment of Alzheimer's disease, pending to the confirmation of the larger sample size and multicenter human study.

Intake of bromelain in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Phytochemical 3,3'-Diindolylmethane Kills Triple Negative Breast Cancer Cells in Vivo and Vitro

Unlike general breast cancer which processes either receptors for estrogen or progesterone and/or receptors with a protein called HER2, triple-negative breast cancer is a form of breast cancer that does not share the aforementioned characteristics.

In other words, triple-negative breast cancer does not have receptors of estrogen or progesterone or genetic preposition.

More importantly, triple negative breast cancer was found to infect women under the age of 40 and women of African or Asian ancestry in the US, compared to breast cancer in the White.

Particularly, according to the epidemiological study, younger women and in women of African ancestry are susceptible to a higher risk of basal-like triple negative breast cancers.

Compared to the malignant nature, triple-negative and basal-like breast cancers are highly invasive which can grow and spread quickly to other tissues and organs through lymph and blood.

Also compared to other breast cancer, with decreased risk after 5 years, triple-negative breast cancers have a substantial higher recurrent rate within 5 years after treatment.

Triple negative breast cancer shares the symptoms similar to those of breast cancer and risk factor except in the form of the genetic preposition.

According to Dr. Shimelis H, the lead scientist in the joint study led by the Mayo Clinic, said, "Pathogenic variants in BRIP1, RAD51C, and TP53 were associated with moderate risk (odds ratio > 2) of TNBC. Similar trends were observed for the African American population. Pathogenic variants in these TNBC genes were detected in 12.0% (3.7% non-BRCA1/2) of all participants".

And, "Multigene hereditary cancer panel testing can identify women with an elevated risk of TNBC due to mutations in BARD1, BRCA1, BRCA2, PALB2, and RAD51D". 

In other words, if you carry the mutated genes BARD1, BRCA1, BRCA2, PALB2, and RAD51D, you are at a higher risk of triple negative breast cancer.

In seeking a potential compound for the treatment of breast cancer, researchers reexamined the promising anticancer effects of novel C-substituted diindolylmethane (DIM) derivatives DIM-10 and DIM-14 in aggressive triple negative breast cancer (TNBC, MDA-MB-231) models.

In vitro studies, novel C-substituted diindolylmethane (DIM) derivatives DIM-10 and DIM-14 demonstrated strong anticancer effects.

NLC formulations. including DIM-10 and DIM-14 extended significant increase in oral bioavailability compared to poor permeability and poor oral bioavailability of DIM-10 and DIM-14 used alone in pharmacokinetic studies.

Futhermore, compared to DIM-10, DIM-10 used alone, the NLC formulation showed an increase in Cmax and AUC values by 4.73 and 11.19-folds.

Similar increased anti-aggressive triple negative breast cancer (TNBC) efficacy was also found in the DIM-14 NLC formulations.

In dogs, the DIM-10 NLC formulations showed an increase of 2.65 and 2.94-fold in Cmax and AUC, against triple negative breast cancer (TNBC) respectively.

Moreover, DIM-10 or DIM-14 NLC formulations demonstrated significant a reduction in tumor volumes TNBC models and treated animals.

Dr. Godugu C, the lead scientist after taking into account co and confounders wrote, "NLC formulation of both DIM-10 and 14 is effective in TNBC models".

The review study includes the search in electronic databases (ProQuest Medical Library, CINAHL Complete, and PUBMED) to locate and identify articles to meet a predetermined set of inclusion criteria, including articles published after 2007 in human participants 18 years or older.

Compared to Remicade in reducing TNF-a, the protein associated with inflammation in adults with CD, curcumin induced a 55 points mean reduction in the Crohn's Disease Activity Index, reduced the expression of IL-1 and levels of C-reactive protein (CRP), associated with elevated levels of proinflammatory cytokines in the colon, particularly in the reduced levels of TNF-a and PPMTase involved in the improvement of colorectal cancer outcomes.

Based on the findings, researchers said, "...Preliminary data would suggest using both would reduce LOR. Curcumin, even by itself, was found to be a cheap and safe way to reduce CD symptoms and inflammatory markers"

Furthermore, according to the study conducted by the Medical School of Marília in the search for articles associating curcuminoids and CD and UC on the database of MEDLINE-PubMed, curcumin demonstrated an intestinal protective effect on reducing oxidative stress and inhibiting the migration of neutrophils and inducible nitric oxide synthase in the intestine.

Moreover, curcumin also improved micro and macroscopic lesions in the gut, prevented apoptosis of intestinal cells and induced the restoration of immune reaction. 

 Dr. Cunha Neto F, after taking into account of co and confounders wrote, "The use of curcumin is cheap, efficient and associated with no side effects, and may become an alternative to the IBD treatment".

Peyronie's disease is a rare condition caused by plaque, that forms inside the penis, leading to a bent erect penis, depending on where the plaque is located.

Patients with Peyronie's disease has been found to affect sexual desire due to a physical condition associated with painful sexual intercourse and erectile dysfunction.

Researchers do not know the exact causes of Peyronie's disease. However, according to the epidemiological studies, researchers suggested that injury that causes bleeding inside the penis may induce the built-up of scars, particularly in men with the genetic preposition of certain genes.

Most cases of pain in Peyronie’s disease are mild and often do not require treatment, however, over counter medicine may be needed occasionally.

Believe or not, most mild cases of men with Peyronie's disease do not require treatment as the condition has been found to go away within a year. In severe cases, surgery may be necessary to remove the scar tissues in the penis that cause the condition.

Bilberry is a species of low-growing shrubs in the genus Vaccinium, belonging to the family Ericaceae, native to Northern Europe.

The plant has been used as herbs in traditional medicine for the treatment of acute and chronic diarrhea, gastritis, gastric ulcer, and duodenal ulcer, enterocolitis, ulcerative colitis, anemia, cystitis, kidney disease, and psoriasis, diabetes, etc.

Researchers on finding a natural compound for the treatment of Peyronie's disease investigated various antioxidants, including bilberry in PD treatment with significantly increase the likelihood of therapeutic success.

By comparing the effects of silymarin, ginkgo biloba, vitamin E, bilberry, topical diclofenac sodium, and pentoxifylline (PTX) in patients with PD, researchers found that by administration of a single oral antioxidant; two oral antioxidants; three oral antioxidants; five oral antioxidants + local diclofenac; and five oral antioxidants + local diclofenac + PTX achieve better results when combining more than one agent.

According to the tested assays, the combination of more than a single compound reduced the symptoms in patients with PD, including the improvement and disappearance of penile pain; percentage of reduction in the volume of penile plaque; reduction in penile curvature; recovery of erectile function in patients with erectile dysfunction; increase in the International Index of Erectile Function score; and reduction of psychosexual impact.

Particularly, treatment of combined therapy including oral treatment with more than one antioxidant is greater when topical use of diclofenac gel and perilesional injection of PTX, compared to those of single conventional medicine.

In other words, combined natural or dietary antioxidants such as bilberry into the diet of patients with PD are more effective in inhibiting the symptoms compared to the use alone of any single ingredient therapy.

The final outcome of 6 months of treatment showed that combined antioxidant therapy is effective in treating PD. 

Dr. Paulis G, the lead scientist, after taking other factors into account wrote in the final report, " Although several articles have already been published reporting the effectiveness of combined treatment in PD, this is the first study clearly proving how, as the number of substances used in treatment rises, a proportionally greater therapeutic effect is achieved."

Taken altogether, bilberry combined with other antioxidants or conventional medicines may be considered function remedies for the treatment of Peyronie's disease (PD), pending to the validation of larger sample size and multicenter human study.

* Regenerating islet-derived protein 3 gammas (Reg3γ), an antibacterial lectin is a type of antimicrobial peptides found in the gut epithelial Paneth cells with a function to prevent microorganisms from coming into contact with the epithelial layer.

In other words, the intestinal mucosal barrier plays an essential role in the separation of the inside of the body from the outside environment, particularly in harmful microorganisms.

Dr. Oshima T the lead scientist in the examination of light junctions (TJs), the most important component for the construction of a constitutive barrier of epithelial cells that regulates the permeability of the barrier by tightly sealing the cell-cell junctions, wrote, "Gastrointestinal diseases including reflux esophagitis, inflammatory bowel disease, functional gastrointestinal disorders, and cancers may be regulated by these molecules, and disruption of their functions leads to chronic inflammatory conditions and chronic or progressive disease".

Phytochemical Alliin Gobbles Up Plagues Against the Onset of Atherosclerosis

Atherosclerosis is a medical condition induced arterial narrowing due to the accumulation of plague that weakens the arterial wall.

Atherosclerosis is the most common cause of heart disease and stroke in the Western world, depending on the location of oxygenated rich blood flow blockage.

Plague is made up of fat, cholesterol, calcium, and other substances in the blood.

According to the epidemiological studies, diseases associated with atherosclerosis include coronary heart disease (plaque in arteries in or leading to the heart), angina (chest pain from reduced blood flow to the heart muscle), carotid artery disease (plaque in neck arteries supplying blood to the brain),  and peripheral artery disease (plaque in arteries of the extremities, especially the legs).

According to the statistics provided by the CDC, coronary heart disease (CHD)  caused by atherosclerosis is the most common type of heart disease, that kills over 370,000 people annually.

Every year about 735,000 Americans has a heart attack. Of these, 525,000 are a first heart attack and 210,000 happen in people who have already had a heart attack

Although there are many risk factors that increase the risk of plaque accumulation, some researchers suggested people who follow the high-fat diet without injection of an adequate amount of fruits and vegetables are at a higher risk of atherosclerosis.

Dr. Zhao Y, the lead scientist in the team of cardiovascular disease at the Chinese PLA General Hospital wrote, "Coronary artery disease has a significant genetic predisposition, which mainly results from atherosclerosis".

And, "(Pig fed with high-fat diet) At 6 months, plaque lesions appeared in the abdominal aorta and iliac artery, while at 8 months, they appeared in the coronary artery. At 12 months, atherosclerotic lesions could be found in all major arteries, while lipid core, cholesterol precipitation, and calcium deposition appeared in the most serious sites".

Alliin (S-allyl-L-cysteine-S-oxide) is a phytochemical compound sulfoxide,. a derivative of the amino acid cysteine, belonging to the class of sulfur compounds, found abundantly in fresh garlic and onion.

With an aim to find potential treatment for atherosclerosis, a leading cause of cardiovascular disease, researchers evaluated the capacity of Allium ursinum (wild garlic) leaf lyophilisate (WGLL; alliin content: 0.261%) to mitigate cardiovascular damage in hypercholesterolemic rabbits.

Selected tested New Zealand rabbits incldued in the study were divided into three groups: (i) cholesterol-free rabbit chow (control); (ii) rabbit chow containing 2% cholesterol (hypercholesterolemic, HC); (iii) rabbit chow containing 2% cholesterol + 2% WGLL (hypercholesterolemic treated, HCT); for eight weeks.

According to the differentiation at the zero- and eight-week time points, WGLL treatment protects the cardio functioning by mediating the increase in fractional shortening, right ventricular function; peak systolic velocity; tricuspid annular systolic velocity in live animals.


Furthermore, WGLL improved aortic and coronary flow.

In other words,  WGLL exerted cardiovascular protective activity by promoting the function of ventricles in blood circulation via regulating the blood pressure.

Moreover, according to the Western blot analysis, revealed WGLL-associated increases in HO-1 protein associated with reduced cardio oxidative stress and decreases in SOD-1 protein involved oxygen metabolism.

Interestingly, WGLL also expressed a significant decrease in aortic atherosclerotic plaque coverage, plasma ApoB, an important component of many lipoproteins involved in atherosclerosis and cardiovascular disease. and the activity of LDH and CK (creatine kinase) in plasma.

Particularly, the chemical compound also inhibited the plasma LDL.

Based on the findings, researchers wrote in the final report, "The results clearly demonstrate that WGLL has complex cardioprotective effects, suggesting future strategies for its use in prevention and therapy for atherosclerotic disorders".

Taken altogether, alliin may be considered supplements for the prevention and treatment of atherosclerosis, pending to the confirmation of the larger sample size and multicenter human study.

Intake of alliin in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Phytochemical Lycopene Inhibits Cancer Metastasis in Vitro and Vivo

Cancer is a class of disease associated with irregular cell growth on the tissue of the specific organ.

Most cases of invasive cancer can spread to other organs in the body via lymph or blood.

According to the statistics provided by the Cancer Institue, between n 2011–2015, the number of new cases of cancer is 439.2 per 100,000 men and women per year (based on 2011–2015 cases). 

The number of cancer deaths (cancer mortality) is 163.5 per 100,000 men and women per year.

Under normal circumstance, cell growth is a natural process without even notice, every day millions of cell grow to replace millions of dead cells and to keep our body in balance and healthy.

Sometimes, acceleration of cells growth is necessary to prevent further damage to the body organ, such as cut, injure, etc., but after fulfilling their purposes, the growth of cells are back to a normal rate again due to our body growth inhibitors. Unfortunately, skin cells sometimes overgrow while healing, leading to scar to the skin.

In cancer cells, due to the alternation of DNA, can grow indefinitely. 

Cancer cell metastasis is the process that cancer cells break away from the primary site and travel through the blood or fluid circulation and form new tumors (metastatic tumors) in other parts of the body, similar to those of primary cancer.

Dr. RUI-AN WANG, the lead scientist in the explanation of the reasons for cancer metastasis, wrote, "Over several years, scientists investigating cancer have focused their efforts on elucidating the mechanisms underlying cancer metastasis, with the aim of finding a way to inhibit this process".

And, "Cancer metastasizes due to several factors, namely attack by the immune system, lack of oxygen and necessary nutrients, large amounts of lactic acid produced by glycolysis and increased cell death. Therefore, the majority of the presently available treatments for cancer also bear the potential to induce metastasis".

In other words, for the tumor to survive that must metastasize.

Lycopene is a phytochemical in the class of carotenoid, a natural pigment with no vitamin A activity found abundantly in tomatoes and other red fruits and vegetables, such as red carrots, watermelons, and papayas,

Tomatoes provide about 80% of the lycopene in the world diet. In plants, lycopene protects the host against excessive photodamage and perform various functions in photosynthesis.

In the urgency to find a natural ingredient for the inhibition of cancer cell metastasis, researchers examined the apo-10'-lycopenoic acid (ALA), a derivative of lycopene through cleavage by carotene-9',10'-oxygenase, on tumor progression and metastasis in both liver and lung cancer animal models.

According to the tested assays, ALA showed significant activity in a dose-dependent manner in inhibiting migration and invasion of liver and lung cancer cells (HuH7 and A549).

In other words, ALA constrained lung cancer cell from the induction of mechanisms associated with cancer cell spreading.

Furthermore, in HuH7 and immortalized lung BEAS-2B cells, ALA suppressing the function of remodeling actin in stimulating cancer cell mobility and membrane formation.

More importantly, ALA also inhibited both HuH7 and A549 cells. proliferation and migration by suppressing the genes associated with angiogenesis.

Moreover, according to the tested assays, ALA also decreased the expression of enzyme which plays a pivotal role in cell migration during physiological and pathological processes.

In human THE-2 liver cells, ALA in a dose-dependently increased the mRNA expression and protein levels of peroxisome proliferator‑activated receptor‑γ (PPARγ) involved in controlling angiogenesis, tumor progression and metastasis.

Based on the findings, Dr. Cheng J., the lead scientist wrote, "ALA inhibits cancer cell motility and angiogenesis and induces PPARγ expression, which could be one of the potential mechanisms for ALA protecting against tumor progression".

Taken altogether, phytochemical lycopene may be considered supplements for the prevention of cancer cell metastasis, pending on the confirmation of the larger sample size and multicenter human study.
