Green Tea and Its Bioactive Theanine That Promote Tobacco Cessation Natually

Green tea mixed in tobacco can significantly improve chronic cigarette smokers to quit smoke naturally, some scientists suggested.

Smoking has a history in many cultures dated back as early as 5000 BC. Europeans who arrived to America in the 16th century have quickly transformed the land to cultivated these plants for commercial profits and encourage consumption.

At of today statistic, tobacco smoking including cigarettes is causing over 3 million deaths a year worldwide, and if current smoking trends continue the annual mortality will exceed 10 million by around 2030, Dr. Anne Charlton, the author of the study suggested(4).

Continuously, the doctor said, " Tobacco leaves and the smoke generated when they are burned contain over 4 thousand chemicals, the best known of which is nicotine, first isolated from tobacco leaves in 1828 by Posselt and Reimann. It is the nicotine that causes smokers to become addicted to tobacco, and the chemical itself is lethal in small doses".

According to a statistic from CDC, more than 4 out of 10 (45.5%) of all high school students who smoke tried to stop smoking in the past 12 months. Since 2002, the number of former smokers has been greater than the number of current smokers in the US(5).

Truly, a high percentage of smokers try to quit, however, smokers who successfully quit smoke are quite low because nicotine is highly addictive.

Green tea contains more amount of antioxidants than any drinks or foods with the same volume, and is the leaves of Camellia sinensis, undergone minimal oxidation during processing, originated from China. 

In the study to test the effects of tea filter in promoted tobacco cessation in volunteers smokers, researchers found that smoking tobacco with tea filter successfully decreased cigarette consumption of participants by 56.5%, and 31.7% of them stopped smoking(1).

Further analysis of tea filter effects suggested the results may be attributed to the tea filter's bioactive chemical theanine in exerting anti-tobacco and nicotine addiction activities.

Dr. Yan J, the lead author said, "This work identified a new method and material, tea filter and theanine, which inhibit tobacco and nicotine addiction and provide an effective strategy for treating tobacco addiction"(1).

Additionally, in the review of clinical studies evaluating herbals use for smoking cessation from databases from its inception to October 2012. Studies are restricted to only English or Thai, researchers indicated that out a total of 7 studies met inclusion criteria; 6 studies were a randomized trial (RCT) evaluating a variety of herbals.

According to the smoking cessation measured with various methods including the number of cigarettes per day, abstinence rate, expired CO level, and urine cotinine level at the 3 and 6 months follow up, application of mix herbal tea showed a statistically significant effect in reduced numbers of cigarettes smoked per day, nicotine craving, and total withdrawal scale but not urinary cotinine excretion(2).

However, Dr. Kitikannakorn N, the lead scientists, after taking into account of other co and confounders said, "Evidence supporting the role of complementary medicines as an anti-smoking is limited. Further research in this field would benefit from the standardization of herbals products to implement on standard interventions and outcome measurement for smoking cessation".

Interestingly, according to the study by the Science China Life Sciences to test the effect of green tea lace in a cigarette in increased smoke cessation probability, researchers found that custom-designed cigarettes laced with green tea components exerted a significant effect in reduced smoking cravings.

Prince Philip, a lecturer in the Community Oncology Division of the Malabar Cancer Centre, said, "The study conducted in China shows that green tea may be an alternative to quit this addictive habit".

Taking all together, green tea used cigarette lace may be considered as a secondary and therapeutic treatment in increased the change of quit smoking. Black tea may have a therapeutic and positive effect in reduced risk and treatment of irritable bowel syndrome, some scientists suggested.

Black tea is a type of tea with more oxidized in compared to green tea, made from leaves of the shrub (or small tree) Camellia sinensis.

Irritable bowel syndrome is a common condition of digestive disorder, affecting the large intestine without inducing evidences of intestinal damage.

Scientists as of today do not the exact causes of irritable bowel syndrome. In the study of symptoms of  gastrointestinal (GI) motility in patients with irritable bowel syndrome who exhibited reduced levels of serotonin 5-HT3 which play an important role in regulating the motility in the entire GI tract, researchers found that application of black tea and its major polyphenolic pigment thearubigins (TR)  enhance movement of the gastrointestinal tract and stimulating gut emptying.

Taken together,  black tea may be considered as functional food for reduced risk, inhibited symptoms  and treatment of irritable bowel syndrome. However, intake of black tea extracts should be taken with extreme care, as acute liver toxicity was reported in some medical literature.

Herbal fennel may have a substantial and potential effect in optimized gastrointestinal function by balancing the ratio of bacteria in the digestive tract, some scientists suggested.

Taken together, there is no doubt that fennel and its bioactive compound anethole may be considered as a function food to promote gastrointestinal function in absorption and distribution of nutrients and fluids to the body.

However, further data collection on large example size and multi centers studies performed with human consumption of the whole food during the course of disease will be necessary to complete the picture of fennel anti digestive disorder possibilities.

The causes of type 2 diabetes are debatable as researchers do not know why some people are susceptible to develop insulin resistance while others do not.

However, genetics, unhealthy diet and lifestyle are the most common risk factors of type 2 diabetes(8).

Some researchers suggested that racial/ethnic group, overweight/obesity, elevated blood pressure, elevated casual blood glucose, and the presence of acanthosis nigricans)  are associated to the development of type 2 diabetes mellitus (T2DM)(9). 

If untreated, patients with type 2 diabetes may develop potential complications of heart disease.neuron, kidney, eye, foot, skin damage.

In some cases, these patients may be infected by plaques accumulated in neurons, leading to Alzheimer's disease.

According to 2015 statistic, in US, approximately 30.3 million Americans, or 9.4% of the population, had diabetes, including  1.25 million children and adults have type 1 diabetes.

Unfortunately, according of Center of Diseases Control (CDC), over 7.2 million people have diabetes but remain undiagnosed, while another 84.1 million adults 18 years and older have prediabetes.

Continuously, the CDC wrote, " The estimated diabetes cost in the United States in 2012 was $245 billion, including $176 billion for direct medical costs and $69 billion in indirect costs, such as disability, time lost from work, and premature death".

Cinnamon, a kitchen spicy in many cultures, may have gained many believers, including medical professions because of its anti diabetic effect, through regulating the blood glucose levels in healthy and diabetic subjects.

According to herbalists, intake of 1/4 tsp. in the morning and evening may reduce the blood sugar significantly with little or no adverse side effects.

The herb has been use in herbal and traditional medicine as anti-fungal and bacteria level to improve reproductive organ, prevent flatulence and intestinal cramping, treat indigestion, diarrhea, bad breath, headache, migraine, etc.(1).

In a total of 66 patients with type 2 diabetes study conducted by Xuhui District Central Hospital, researchers showed that cinnamon reduces significantly blood triglyceride levels even in the low-dose group, after 3 months of oral administration(2)

Its extract also improved fasting blood glucose levels and glucose tolerance without altering the insulin secretion, through lower triglyceride and increased liver glycogen content and improved insulin action in liver tissues(3).

Dr. Kim SH and Dr. Choung SY. told PubMed that “The effect of cinnamon in significantly increases insulin sensitivity, reduces serum, and hepatic lipids, and improves hyperglycemia and hyperlipidemia possibly by regulating the PPAR-medicated glucose and lipid metabolism”(4)

In support of the above, chromium(Cr) and polyphenols found in cinnamon(Cinnamomon cassia)  in patients with type 2 diabetes expressed an strong improvement of glucose, insulin, cholesterol and HbA1c following Cr supplementation, in a double-blind placebo-controlled study(7).

Unfortunately, according to the Western University of Health Sciences, consumption of cinnamon is associated with a statistically significant decrease of levels of fasting plasma glucose, but insisted that high degree of heterogeneity may limit the ability to apply these results to patient care, due to the unclear preferred dose and duration of therapy(5)

The randomized clinical trial in which 70 Patients with type II diabetesassigned randomly two groups (35 in cinnamon and 35 in placebo group for the testing of the effect of cinnamon on the glucose level in blood, also indicated an insignificant effect of using cinnamon on glucose level of diabetic patients and the herb should not be recommended to patients with type II diabetes(6).

Taken altogether, cinnamon in low dose may be effective in controlling the levels of blood glucose in patient with diabetes without altering the insulin level. according to DR. Georgakopoulou EA(7).

Oral intake of Cinnamon may cause Stomatitis, and burning sensation in mouth with large amount. The traditional Chinese medicine also insisted that Overdoses of cinnamon can be toxic and damaged to liver(1).

According to viral classification, there are 8 types of herpesvirus, including human herpesvirus 1 that causes cold sores around the mouth. and 2 that causes genital herpes, herpes zoster that causes chickenpox and shingles, Epstein-Barr virus that causes infectious mononucleosis, cytomegalovirus (CMV) that causes mononucleosis, Human herpesvirus 6 and 7(HHV6 - 7) that causes roseola and Human herpesvirus 8 (HHV8) that causes cancer, such as Kaposi's Sarcoma.

According to statistic, in the United States, half of teens and adults under age 50 are still infected with the oral herpes virus, and about 1 in 8 have an infection with the genital herpes virus.

All herpesviruses like other virus are nuclear-replicating, infecting healthy cells nucleus through replication of viral DNA and transcription of viral mRNA.

People infected by may experience symptoms.of painful sores in the genital area, anus, buttocks, thighs, itching, painful urination, vaginal discharge,..... and tender lumps in the groin.

However, some people may not show any symptoms at all and sometimes, the diseases may be misdiagnosed as a skin condition.

EGCG and other purified green tea catechins as topical agents inactivate multiple clinical isolates of HSV-1 and HSV-2(1). 

Application of EGCG in 10 to 20 min and reduced HSV-1 titers by the same amount in 30 to 40 min reduced HSV-2 titers by ≥1,000-fold(1).

Dr. Charles E, the lead scientists said, "(The efficacy in ameliorated risk of HSV), because EGCG would be able to directly inactivate extracellular HSV virions in the vaginal lumen" and " At a concentration range that can easily be delivered to the female reproductive tract".

Interestingly, in the study of Epigallocatechin-3-gallate (EGCG) effects on primary effusion lymphoma (PEL), an aggressive neoplasm caused by human herpesvirus 8 (HHV8), researchers found that EGCG inhibits induced cell death and ROS generation in PEL cells in a dose-dependent manner(2).

Furthermore, application of EGCG reduced the production of progeny virus from PEL cells without causing HHV8 reactivation(2). 

These results suggest that EGCG may have a therapeutic treatment of HHV8 infection and HHV8 that cause lymphomas.

Most importantly, according to the joint study conducted by the Institute of Medical Microbiology, University Hospital Essen, University of Duisburg-Essen, epigallocatechin-3-gallate (EGCG), processed significant anti-infective properties. EGCG exerted different modes of action in inhibited the diverse families of viruses(3)

In details(3), EGCG 
* inhibited HCV through viral entry by interference with binding to target cells.
* Inhibited integrase, an enzyme produced by a retrovirus HCV by binding to envelope and CD4 and interference.
* Reduced HBV antigen expression, extracellular HBV DNA and cccDNA
* Induced damage and inactivation of virions of HSV-1/HSV-2, probably by binding to envelope proteins
* Inhibited EBV transcription of immediate-early genes Rta, Zta, and EA-D
* Inactivated Adenovirus particles, inhibited Adenovirus intracellular virus growth and viral protease
* Alternated influenza virus physical integrity of virus particles, inhibited entry by binding to haemagglutinin
* Suppressed enterovirus replication via modulation of cellular redox milieu

Taking all together, green tea and its bioactive constituents may be used in combined with conventional medicine for the prevention and treatment of herpesvirus. 

However, further data collection on large example size and multi-centers studies performed with human consumption of the green tea and its bioactive polyphenol epigallocatechin 3-gallate (EGCG), during the course of herpesviral infection will be necessary to complete the picture of their anti herpesviral possibilities.

Green Tea Mixture (NM) Which Kills 100% of Hemangioendotheliomas Cells, Biomedical Literature Suggest

Green tea may have a therapeutic and positive effect in reduced development of hemangiomas, some scientists suggested.

The results were carried out by some respectable intistutes including the Dr. Rath Research Institute and published on online medical literature.

Hemangioma is a benign tumor formed by a collection of excess blood vessels. The disease is a birth mark usually growth during the first year of life, and recedes over time

Hemangioendotheliomas are chronic condition caused by abnormal growth of blood vessel cells, with no exact underlying cause. The disease is a rare vascular tumor, described for the first time in 1975 by Dail and Liebow as an aggressive bronchoalveolar cell carcinoma.

The disease most commonly affect the soft tissues, liver, lungs and bones.

Localized epithelioid hemangioendothelioma can be removed surgically.

In differentiation of the efficacy of a nutrient mixture (NM) containing lysine, proline, ascorbic acid and green tea extract against the development of hemangiomas researchers at the Dr. Rath Research Institute, conducted a experiment using mouse in hemangioendothelioma model injected by Matrigel.

NM had shown a significant effect in inhibited hemangioendothelioma cell proliferation and cell division, in vitro through evaluating cell viability, MMP secretion, invasion, morphology and apoptosis.

Athymic nude mice, 5-6 weeks old, inoculated with 3 x10(6) EOMA cells subcutaneously and randomly divided into two groups,
* Group A was fed with a regular diet and
* Group B was fed with a regular diet supplemented with 0.5% NM.

At the end of the experiment, researchers found that mice fed with NM inhibits 50% of tumors growth.

Further analysis suggested that the efficacy of NM in inhibited the growth of tumor was attributed to its cytotoxicity linearly in induced apoptosis of EOMA cells, in dose dependent manner.

Injection of Matrigel in induction of tumor was inhibited by green tea mixture at 50, 100 and 500 microg/ml by 25%, 30% and 100% respectively

Dr. Roomi MW, the lead author, after taking into account of other co and con founders said, " NM may have therapeutic potential in treating infantile hemangioendotheliomas and, perhaps, other cutaneous vascular tumors".

Taken together, green tea mixture NM may have a profound effect in reduced risk and treatment of hemangioendotheliomas.

However, further data collection on large example size and multi centers studies performed with human consumption of green tea and its bioactive phytochemicals, during the course of the condition will be necessary to complete the picture of their anti hemangioendothelioma possibilities.

Green tea contains more amount of antioxidants than any drinks or foods with the same volume, and is the leaves of Camellia sinensis, undergone minimal oxidation during processing, originated from China.

Gallocatechin, containing catechin is phytochemicals of Flavan-3-ols, in the group of Flavonoids (polyphenols), found abundantly in green tea, almonds, black diamond plums, black tea, cocoa beans, Fuji apples, golden delicious apple, etc.


1. Bone metabolism

In the investigation of three tea catechins, epigallocatechin (EGC), gallocatechin (GC), and gallocatechin gallate (GCG) for their effects on bone metabolism, researchers found that EGC significantly inhibit osteoclast formations from RAW 264.7 cells upon receptor activation of nuclear factor-kappaB ligand induction on the fourth day of treatment, at a concentration of 10 microM(1).

Receptor activator of nuclear factor (NF-kappaB) ligand (RANKL) is the essential factor for osteoclast formation, fusion, activation, and survival, thus resulting in bone resorption and bone loss.

EGC also at dose-dependently inhibited the mRNA expression of tatrate-resistant acid phosphatase(1) which plays a critical role in many biological processes including skeletal development, collagen synthesis and degradation, the mineralisation of bone(14),
Further more, GC and GCG could decrease osteoclastogenesis at 20 microM.

Dr. Chun Hay Ko, the lead author said, "tea catechins, EGC in particular, had positive effects on bone metabolism through a double process of promoting osteoblastic activity(bone formation) and inhibiting osteoclast differentiation(bone re absorption)"(1).


2. Antimetastatic effects
In the evaluation of the antimetastatic effects of P urinaria L extracts (PUE), containing polyphenols including gallic acid, methyl gallate, epicatechin, epigallocatechin-3-gallate, gallocatechin-3-gallate, rutin, epicatechin-3-gallate, and naringin, researchers found that PUE inhibits the transcription of  of MMP-2 osteoblasts  that regulates osteopontin and bone protein expression in bone metastasis of osteotropic cancers(2).

Further more, PUE also exerted an inhibitory effect on the DNA-binding activity and the nuclear translocation of NF-κB and AP-1 in induction of secondary cancer and growth of LLC cells in vivo(2)


"(These results suggested that) PUE could be applied applied to be a potential antimetastatic agent", Dr. Tseng HH, the lead scientists at the Antai Tian-Sheng Memorial Hospital said(2).


3. Anti skin cancer
In the determination of the effect of green tea catechins on the invasive potential of human melanoma cells and the molecular mechanisms underlying these effects using A375 (BRAF-mutated) and Hs294t (Non-BRAF-mutated) melanoma cell lines in vitro model, researchers found that application of green tea catechins inhibits melanoma cell migration, through transition of mesenchymal stage to epithelial stage in induction of tumor growth to advanced stage(3).

These results indicate that EGCG, a major green tea catechin, has the ability to inhibit melanoma cell invasion/migration, an essential step of metastasis, by targeting endogenous expression of COX-2, which plays an important role in increased production of PGE2, the hormone that modulates skin caner cell proliferation from epithelial-to-mesenchymal transition(3).

4. Antidiabetic activity
In the observation of Terminalia sericea stem bark extract and theirs effect against alpha-glucosidase and alpha-amylase enzymes, four known compounds namely beta-sitosterol (1), beta-sitosterol-3-acetate (2), lupeol (3), and stigma-4-ene-3-one (4), in addition to two inseparable sets of mixtures of isomers [epicatechin-catechin (M1), and gallocatechin-epigallocatechin (M2). scientists found that  1 and 3 show the best inhibitory activity on alpha-glucosidase (IC50:54.5 and 66.5 microM) located in the brush border of the small intestine with function in stimulated digestion of carbohydrates that facilitates onset of diabetes mellitus type 2(4).

Bio-evaluation of the inhibitory activity of the purified compounds on alpha-amylase showed that 3 and 1 exhibit IC50 values of 140.7 and 216.02 microM, respectively against alpha-amylase, a protein enzyme that -links polysaccharides in induction of glucose and maltose with no side effects(4).

5. Anti-uveal melanoma and cancers activity

In the study of  the new compound gallocatechin 5-O-gallate isolated to MeOH extract of Acacia nilotica pods, scientists found that the newly found compound exerts a significant effect in inhibited uveal melanoma, a cancer (melanoma) of the eye involving the iris(5).

Further more, the antiproliferative activities of the isolated compounds and the related compound epigallocatechin 3-O-gallate (EGCG) also have a profound effect in protected healthy cells against the development of cutaneous melanoma, ovarian cancer, glioblastoma and normal retinal pigmented cells(5).


6. Degenerative diseases

In the investigation of whether green tea and its components can regulate the osteogenic and adipogenic differentiation in pluripotent rat mesenchymal stem cells (MSCs)in MSCs isolated from the bone marrow of tibiae and femora in rat, researchers found that among six tested tea polyphenols, epigallocatechin (EGC) processes the most effective in promoting osteogenic differentiation.

At 20 μM, EGC increased ALP levels and Ca deposition significantly in promote bone denesity by 2.3- and 1.7-fold, respectively,

When compared with the control group. EGC also increased the mRNA expression of bone formation markers runt-related transcription factor 2, ALP, osteonectin, and osteopontin(6)

7. Antioxidants
In the identification of glucose-6-phosphate dehydrogenase (G6PD) and its important roles in the maintenance of cellular redox balance, researchers found that pretreatment with green tea polyphenol epigallocatechin-3-gallate (EGCG) effectively blocks peroxynitrite-induced glutathione depletion(antioxidant enzyme in the host), p53 accumulation in suppressed irregular cell growth and  apoptosis in both normal and G6PD-deficient cells(7).

EGCG, administered to cells alone or as pretreatment, caused activation of protein kinase B (PKB), also known as Akt, that plays a key role in multiple cellular process in promoted survival and growth in response to extracellular signals.

The protective effect was abolished by phosphatidylinositol 3-kinase (PI3K) inhibitors, wortmannin, and LY294002, a steroid metabolite with function as immune suppressors(7).

8. Anti HIV
In the investigation of Epigallocatechin gallate (EGCG), the most abundant catechin in green tea and its effect on HIV-1, scientists found that EGCG appears to act mainly as an allosteric reverse transcriptase inhibitor with mechanisms different from those of currently approved NNRTIs used in the treatment of human immunodeficiency virus (HIV)(8).

Thus, EGCG is a good candidate for use as an additional or supportive anti-HIV agent derived from natural plants, according to "Epigallocatechin gallate inhibits the HIV reverse transcription step" by Li S, Hattori T, Kodama EN.(8)

9. Anti inflammatory activities
In the evaluation of the radioprotective efficacy of green tea polyphenols and the component ingredients against irradiated-induced damage in mice and elucidate the underlying mechanisms,  researchers found that GTP and its bioactive components (catechin, epigallocatechin and epigallocatechin-3-gallate) assists in decreasing the leukocytopenia, disease that decreases number of white blood cells (leukocytes) found in the blood(9).

Injection of GTP and its bioactive components, significantly reduced the elevated serum inflammatory cytokines (TNF-α, IL-1β, and IL-6)(9).

These results suggested that green tea polyphenols have a potential to be developed as radioprotective agents against irradiated-induced toxicity(9).

Furthermore, the antioxidant and anti-inflammatory activities of GTP can be attributed to the interaction of the different components through multiple and synergistic mechanisms.

10. Anti microbial effects
In the evaluation of the anti-inflammatory and antimicrobial effect of nanocatechin on CBP and plasma concentration of catechins in an animal model, researchers showed that the use of ciprofloxacin, catechin, and nanocatechin statistically significant decrease in bacterial growth and improvement in prostatic inflammation compared with the control  and catechin group..

Plasma concentrations of epicatechin, gallocatechingallate, and epigallocatechin gallate, were significantly higher in the nanocatechin group than those in the catechin group.

These results suggest that nanocatechin has better antimicrobial and anti-inflammatory effects on rat CBP than catechin due to higher absorption into the body(10).

11. Ultraviolet B irradiation protection
In the investigation of the protective effect of epigallocatechin gallate (EGCG) on the immune function of dendritic cells (DCs) after ultraviolet B irradiation (UVB), scientists found that the inhibition rate of DCs was improved to some extent after treatment with 200 microg/mL of EGCG.

UVB showed no significant influence on the secretion of IL-10 and IL-12 from DCs, while EGCG was able to down-regulate the secretion level of IL-12 and up-regulate that of IL-10.(11).

12. Antiviral effect
In the identification of tea polyphenols ability to inhibit enterovirus 71 (EV71) replication in Vero cell culture, researchers found that the viral inhibitory effect correlated well with the antioxidant capacity of polyphenol(12).

Mechanistically, EV71 infection led to increased oxidative stress, as shown by increased dichlorofluorescein and MitoSOX Red fluorescence(12).

Upon EGCG treatment, reactive oxygen species (ROS) generation was significantly reduced.

Consistent with this, EV71 replication was enhanced in glucose-6-phosphate dehydrogenase deficient cells, and such enhancement was largely reversed by EGCG(12).

13. Neuroprotective effect
In the research of beta-Amyloid (Abeta) peptide, a major component of senile plaques that play a crucial role in the development and neuropathogenesis of Alzheimer's disease (AD), researchers found that EGCG may have preventive and/or therapeutic potential in AD patients by augmenting cellular antioxidant defense capacity and attenuating Abeta-mediated oxidative and/or nitrosative cell death(13).
