Ashwagandha Protects the Kidneys Against Nephrotoxicity

Kidneys are the bean-shaped organs located below the rib cage, one on each side of your spine with a function to filter out blood and remove waste through urinary secretion.

The kidney plays a critical in maintaining the levels of fluids in the body, by removing extra fluids to make urine.

Furthermore, the kidney also regulates electrolytes, blood pressure, maintains acid-base balance, produces hormone calcitriol, renin, and erythropoietin.

Nephrotoxicity or acute kidney failure is a condition of toxicity in the kidneys.

The disease is caused by a poisonous effect of prolonged use of certain medications and the accumulation of harmful chemicals.

According to the American physician association, long term use of penicillins, cephalosporins, sulfonamides, thiazide diuretics, furosemide, NSAIDs and rifampicin are associated to increase damage to the kidney.

And people with age over 60 are at increased risk of nephrotoxicity if the above medicines are prescribed for prolonged periods.

Continuously, the association wrote, "Drug-induced nephrotoxicity tends to be more common among certain patients and in specific clinical situations" and " Drugs cause approximately 20 percent of community-and hospital-acquired episodes of acute renal failure".

Ashwagandha also is known as Withania somnifera is a nightshade plant in the genus of Withania, belonging to the family Solanaceae, native to the dry parts of India, North Africa, Middle East, and the Mediterranean.

The herbal medicine has been considered as Indian ginseng and used in Ayurvedic medicine over 3000 years to treat tumors and tubercular glands, carbuncles, memory loss, and ulcers and considered as anti-stress, cognition-facilitating, anti-inflammatory, and anti-aging herbal medicine.

On finding a potent compound that processes anti Nephrotoxic activity, researchers examined the nephroprotective effect of W. coagulans extract and its modulatory effects against cisplatin-induced nephrotoxicity.

The experiment included rat induced nephrotoxicity by a single dose of cisplatin (6 mg/kg, intraperitoneal). treated by W. coagulans fruit extract at different doses.

According to the tested differentiation, Withaferin A at 3.56 mg/g of W. coagulans fruit extract significantly inhibited the rise in serum urea and creatinine level and also protected the rat kidneys against oxidative stress and free radical-induced DNA damage. 

Furthermore, the extracts also protected kidney cells' integrity by eliminating parameters associated with kidney toxicity against tubular swelling, cellular necrosis, and protein cast deposition in cisplatin-treated kidney tissues by enhancing the production of antioxidants such as glutathione content, activities of superoxide dismutase and catalase. 

Most importantly, the extract in a dose of 400 mg/kg restored all parameters to near-normal levels.

Based on the results, researchers suggested, "It can be justified that W. coagulans possess dose-dependent protective effect against cisplatin-induced kidney damages, primarily through its free radical scavenging and anti-inflammatory activity".


Taken altogether, Ashwagandha processed abundantly Withaferin A may be considered a functional herb for the prevention nephrotoxicity, pending to the result of larger sample size and multicenter study
