Phytochemical Indole-3-carbinol (I3C), the Potent Anti Cervical Cancer Plant Base Agent

Indole-3-carbinol may be the next pharmaceutical ingredient for the prevention and treatment of cervical cancer with no side effects, some studies suggested.

Indole-3-carbinol is a phytochemical in the class of Indoles, by the breaking down of the glucosinolate glucobrassicin, found abundantly in cabbage, kale, Brussels sprouts, rutabaga, mustard greens, broccoli, etc.

Cervical cancer is a malignant neoplasm of the cervix uteri or cervical area caused by abnormal cells growth with alternation of cells DNA.

Researchers do not know what causes cervical cancer. Neither they can explain that women with the same health condition, family history, some are susceptible to the disease while others do not.

Most cases of cervical cancer start in the cell on the surface of the inner lining tissue of the cervix. At this stage, most women with cervical cancer do not experience any symptoms.

At the advanced stage, invasive cancer can induce symptoms of abnormal bleeding, pain during sexual intercourse, increased vaginal discharge. If the tumor has suppressed the nearby nerve cells, cervical cancer can also cause localized severe pain.

According to the American Cancer Society's, in 2014, 12,360 new cases of invasive cervical cancer will be diagnosed with the death of 4,020 patients.

The risk of cervical cancer is higher in Hispanic women followed by African-Americans, Asians and Pacific Islanders, and whites.

Epidemiological also suggested that HPV infection, sexual activity, smoking, giving birth many times. Infection with the human immunodeficiency virus (HIV, history of sexually transmitted infections (STIs) and long-term use of oral contraceptives are some prevalent risk factors of cervical cancer.

However, most women with some of the risk factors have never developed cervical cancer.

According to the statistic, the 5-year survival rate for women with invasive cervical cancer is 92%. and approximately, 46% of women with cervical cancer are diagnosed at an early stage.

With an aim to find a natural ingredient or whole food for the prevention and treatment of cervical cancer, researchers investigated the anticancer effect of I3C and its major acid-catalyzed condensation product diindolylmethane (DIM), against the human cervical cancer cell lines.

In vitro, both I3C and DIM caused accumulation of DNA strand breaks and induction of apoptosis in three cervical cancer cell lines.

In C33A cervical cancer cells, DIM was more potent than I3C  and faster acting.

I3C reduced the expression of a protein associated with the cancer cell proliferation and survival in a time- and dose-dependent manner. 

Futhermore, I3C  also promoted the protein associated with the programming cancer cell death.

Moreover, Mice that express transgenes for human papillomavirus type 16 under a keratin 14 promoter (K14-HPV16 mice) induced cervical cancer by given 17beta-estradiol chronically, researchers found that indole-3-carbinol (I3C) administered at physiological doses, would prevent cervical-vaginal cancer promoted by high doses of estrogen.

Compared to mice fed a control diet with those fed a diet supplemented with 2000 ppm I3C, researchers surprisingly discovered that only 2 mice of 24 in the group fed the I3C supplemented diet developed cervical cancer. I3C also reduced dysplasia in the nontransgenic mice.

Dr. Jin L after taking into account co and confounders said, " I3C is a useful preventive for cervical-vaginal cancer and, possibly, other cancers with a papillomavirus component."

Also, in a cervical cancer cell line, application of 13C exhibited the genes associated with cancer suppression through a number of mechanisms, including cancer cell adhesion, spreading, and invasion.

These results reconfirmed that the diminished PTEN expression during the progression from low-grade to high-grade cervical dysplasia in humans and in a mouse model for cervical cancer.

Taken altogether, Indole-3-carbinol (I3C) may be used for the prevention and treatment of cervical cancer with no side effects, pending to large sample size and multicenter human study.
