Phytochemical Polysulfides, the Natural and Potential Chemo- Therapeutic Agent for colon Cancer with No Side Effects

Phytochemical Polysulfides may be considered a functional supplement in reducing the risk and treatment of colon cancer.

Colon cancer is a very slowly growing medical condition characterized by irregular cell growth in the colonic tissue.

At the advanced stage, cancerous cells can travel a distance away from the colon to infect other healthy tissue and organ through lymph and blood.

The colon is a part of the digestive system involved the absorption of water from the small bowel contents and broken down of certain materials of the feces into substances of which some of them to be reabsorbed and reused by the body.

According to the statistics,  in Canada, every year approximately over 17,000 new cases of colorectal cancer are diagnosed causing over 6500 death

Colorectal cancer ranks third overall in the number of both new cancer cases and cancer deaths in Canada.

The causes of colon cancer are unknown. however, people who are obese, follow the Western diet and long-term exposure to ionizing radiation due to occupation are some of the most prevalent risk factors that cause colon cancer.

The “Western” diet, characterized by high intakes of saturated fat and trans fat, red and processed meats, sweets, refined grains, and desserts increased the risk and recurrent risk of colon cancer, according to the study published on the Aug. 15 issue of the Journal of the American Medical Association.

Dr. the lead scientist in the comparison of diet pattern said, " Compared to the Western-Southern pattern, the fruit-vegetable and metropolitan patterns were associated with more healthful dietary behaviors (e.g., higher vegetable intake and lower red meat consumption), and demographic/lifestyle characteristics typically correlated with low colon cancer risk, for example, lower BMI, higher education, and higher NSAID use".

Therefore, by changing diet pattern from the Western diet to traditional diet with more fruits and vegetable, reducing weight and actively engaging in a healthy lifestyle, risk of colon cancer are decreased substantially.

Polysulfides are phytochemicals in a class of chemical compounds containing chains of sulfur atoms, belonging to the group ofOrganosulfides found abundantly in an oxidized product, including beer, wine, whiskey, garlic oil, etc.

In the finding, the new anti-colon cancer drug with the same effect of diallyl polysulfides are well known as anticancer drugs with no side effects, researchers at the University of the Saarland conducted an experiment to examine the effect of the combined coumarins with polysulfides in the form of di-coumarin polysulfides in HCT116 colorectal cancer cell line.

Application of combination reduced cell viability of cancer cells in a time and concentration-dependent manner.

The combined coumarins with polysulfides exerted apoptosis through the G(2)/M phase of the cell cycle.

The apoptosis activity was attributed to the effect of the combined coumarins with polysulfides in the activation of the proteins that program cell death.

In the comparison of these new coumarin compounds with the well-known diallyl polysulfides, researchers found that the coumarin disulfides are more active than the corresponding diallyl disulfides.

Dr. Saidu NE, the lead author in the final report said, "coumarin polysulfides also down-regulated the level of Cell Division Cycle 25C,(cdc25C), which also contributed to the arrest in the G(2)-phase of the cell cycle".

Futhermore, in the evaluation of the garlic-derived organosulfur compounds such as diallyl sulfide, a member of polysulfides, researchers revealed that the compound exerts a significant influence on cell viability, cell cycle arrest and induction of apoptosis in HCT116 human colon cancer cells.

Diallyl trisulfide is even more efficiently diallyl tetrasulfide treatment of HCT116 cells that lead to a reduced cell viability, cell cycle arrest, and apoptosis.

Additional analysis suggested that the efficacy of the compound was attributed to the function in inhibition of the redox state of the cell, and its anti-oxidants that prevent sulfide-induced apoptosis.

The growth arrest and induction of apoptosis are associated with a considerable reduction of the level of cdc25C similar to those of the previous study.

Taken together, the bioactive polysulfides may be considered a functional phytochemical for the prevention of colon cancer and combined with primary therapy for the treatment of colon cancer.
