Herbal Ashwagandha Exerts Cytostatic Effect Against Multiple Myeloma In Vitro

Multiple myelomas (Myeloma) is a type of cancer originated from plasma cells in the bone marrow.

The disease is a condition of DNA alternation of white blood cell normally responsible for the production of antibodies to protect our body against infection with some characteristics.

Most common symptoms of MS are
* Bone pain
Abnormal growth of plasma cells can cause a collapse of the bone.

* Infection
Overcrowding of abnormal plasma cells reduces the functions of white blood cells produced by the bone marrow, leading to a weakened immune system. Most common infections found to be pneumonia and pyelonephritis.

* Anemia
As a result of malignant plasma cells has to interfere with the normal production of red blood cells.

* Bleeding
Due to the lower production of the platelet.

* Renal failure
Abnormal plasma cells can also affect the function of the kidney in getting rid of waste, causing urea and creatinine accumulated in the bloodstream.

* Neurological symptoms
Due to hypercalcemia, as a result of the breakdown of the bone cause of the release of calcium into the bloodstream.

However, most people with some of the above symptoms never developed MS.

On finding a potent compound for the treatment of tumor of the bone marrow, researchers examined the growth inhibitory effects of withanolide D (WND), a steroidal lactone isolated from Withania somnifera on drug-resistant subpopulation known as cancer stem cells (CSCs).

According to tested assays, WND exhibited similar cytostatic effects by inhibiting cancer cell growth and division. on drug-resistant and drug-sensitive cell lines in the nM range.

In cancer cell lines MM-CSCs and RPMI 8226, the administration of WND also induced cancer cell death and apoptosis.

However, , WND did not increase the accumulation of the fluorescent P-gp substrate rhodamine 123 in MM-CSCs associated with cancer cells drug resistance. More precisely, the result suggested that WND at the tested doses may not inhibit P-gp accumulation involving in the development of resistance of cancer cells to chemotherapeutic drugs.

Based on the findings, researchers said, " the WND-induced cytostatic effect may be independent of P-gp efflux".

Betacyanins, Protects the Proteins, Lipid and Cells Against Oxidative Stress

Inflammation is a natural response of the immune system that tries to protect the body against invasive pathogens.

In the acute phase of infection, after receiving the information transmitted by the central nervous system, the white blood cells of the first line of defense stimulate the production of blood platelets to cover the site of damage and pro-inflammatory cytokines, killing off the foreign invaders.

During the acute phase of infection, overproduction of proinflammatory cytokines may cause damage to nearby healthy cells, leading to the formation of scars.

Most cases of infection are stopped at the acute phase of infection. The wound is slowly healed within a few days or weeks.

However, if the immune system can not destroy all pathogens within a period in the range of 3 to 8 weeks, it will adapt to the new change, leading to chronic inflammation.

In other words, chronic inflammation happens when the immune response lingers, causing low-grade inflammation. Chronic inflammation over time may damage the body tissues and organs.

In some cases, the infection may be latent in which the pathogen remains within the body for a long time without producing any symptoms, such as the hepatitis virus.

Diseases associated with chronic inflammation include cancer, heart disease, rheumatoid arthritis, type 2 diabetes, and obesity.

Betacyanins are phytochemicals in the class of red and yellow indole-derived pigments, belonging to the group of Betalains, found abundantly in beets, chard, etc.

On finding a natural pigment for the prevention and treatment of disease associated with chronic inflammation, researchers examined the effects of Beetroot (Beta vulgaris L.) juice (BRJ, betacyanins, and betaxanthin) anti-inflammatory activity in vitro and in vivo.

The study included rats induced myocardial injury by isoproterenol (ISP) at 24 h intervals, followed by oral administration of BRJ for 28 days at doses of 150 and 300 mg/kg.

According to the tested assays, pretreatment with BRJ (150 and 300 mg/kg) not only reduced oxidative/nitrosative stress but also restored the cardiac endogenous antioxidants in myocardial injury rats.

ISP induced cardiac inflammatory cytokines such as NF-α, IL-6, and IL-10 and biomarkers of inflammatory promotion such as NF-κB also were inhibited by the pretreatment of BRJ.

Oxidative stress-induced hyperlipidemia level was significantly reduced by the BRJ pretreatment.

The results strongly suggested that BRJ pretreatment mitigates cardiac dysfunction and structural damages by decreasing oxidative stress, inflammation in cardiac tissues.

Dr. Raish M, the lead scientist said, "These results further support the use of BRJ in traditional medicine against cardiovascular diseases".

Furthermore, in rat induced renal damage by Paraquat injected intraperitoneally randomly divided into four groups: a control group, a paraquat group, and two paraquat groups that were treated with betanin at 25 and 100 mg/kg/d three days before and two days after paraquat administration.

According to the differentiation of the proteins associated with pro-inflammatory cytokines and pathways that promote inflammatory stimulation, betanin administration found in the family of betacyanins alleviated the paraquat-incurred acute kidney injury by decreasing oxidative stress associated with the inflammatory expression.

Taken altogether, betacyanins processed abundantly bioactive ingredient betanin in BRJ may be considered a remedy for the prevention and treatment of disease associated with inflammation, pending to the confirmation of the larger sample size and multicenter human study.

Intake of betacyanins in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Diabetes is a medical and chronic disease characterized by insufficient insulin entering the bloodstream to regulate glucose.

In vitro, in vitro CP and CP-SAB administration showed a significant inhibition of α-amylase, α-glucosidase, and angiotensin I-converting enzyme (ACE) associated with the management of hyperglycemia linked to type 2 diabetes.

At 5 mg/mL, CP-SAB exerted a maximum of 40% inhibition against α-amylase.

Levels of ACE were also significantly inhibited by CP-SAB at the concentration of 100 and 200 mg/mL sample concentrations.

Based on the results, researchers wrote, "These in vitro results indicate the potential of cranberry powders as a dietary supplement and food-based strategies for potential hyperglycemia management".

Based on the findings researchers said, "This paper gives an overview of the protective effects of curcumin against I/R injury in the liver and discusses the studies that have linked biological functions of curcumin with liver I/R injury improvement".

Ulcerative colitis (UC) is a type of inflammatory bowel disease induced chronic inflammation and ulcers (sores) in the digestive tract.

The other type of IBD is Crohn's disease, caused by inflammation of the lining of the digestive tract, other than the colon and rectum.

IBS affects the colon and the rectum, leading to bloody diarrhea accompanied by symptoms of belly pain, not feeling hungry, fatigue, weight loss, dehydration.

Some patients may also experience symptoms of joint pain or soreness and sudden urges to empty the colon right away.

Inflammation is a process of the immune system that responds to harmful pathogens.

Inflammation can be acute or chronic. In the acute phase of infection, the immune system after receiving information from the neurons, macrophages, the largest white blood cells in the first line of defense stimulate the production of blood palette to cover the wound and the production of inflammatory cytokines to kill off micro-organisms, leading to symptoms of fever, redness, and swelling.

The UC is caused by the immune system mistakenly attacks the cell of the colon, leading to low-grade inflammation.

Tomatoes provide about 80% of the lycopene in the world diet. In plants, lycopene protects the host against excessive photodamage and performs various functions in photosynthesis.

On finding a potential compound for the treatment of colitis researchers examined the protective effects of conventional and colon-targeted lycopene (TLC) and linalool (TLN) on acetic acid (AA)-induced ulcerative colitis (UC) in an animal model.

The study included conventional and colon-targeted LC (10 mg/kg) and LN (200 mg/kg) that were administered in vivo orally for 7 days and sulfasalazine (100 mg/kg) was also used as the reference drug, following by 4% AA to induce rats UC.


Based on the tested analysis, the levels of stress marker such as malondialdehyde (MDA), pro-inflammatory cytokines such as interleukin 1β (IL-1β), IL-6, cyclooxygenase-2 (COX-2) and nuclear factor kappa B (NF-κB) levels were decreased in the both targeted groups compared to the AA group.

Levels of nuclear factor-erythroid 2-related factor 2 (Nrf-2) associated with the increased antioxidant enzymes produced by the host was also increased comapred to control.

Furthermore, Tumor necrosis factor, a cell-signaling protein (cytokine) involved in systemic inflammation was significantly decreased and catalase activity (CAT) associated with levels of antioxidants increased in the TLC group, compared to the AA group 

The results strongly suggested conventional and colon-targeted lycopene showed a significant effect in the reduced risk of UC.

Based on the findings, researchers said, "sulfasalazine was effective on all parameters analyzed, but the colon-targeted pretreatments were more effective from sulfasalazine on some parameters. Therefore, colon-targeted plant-derived therapies might be alternative approaches to provide protection against UC,".

Taken altogether, lycopene found in tomato combined with conventional medicine may be considered a remedy for the prevention and treatment of UC, pending on the confirmation of the larger sample size and multicenter human study.

Intake of lycopene in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Turmeric, the Anti Hypercholesterolemic Herbal Remedy, Scientists Find

Cholesterol, a waxy substance produced by the liver or the dietary plays a critical role to aid digestive function in absorbing nutrients and fluids, build strong cell membranes, produce steroid hormones and vitamin D. However, excessive amounts in the blood have been found to induce plaque built-up on the arterial wall, leading to hypercholesterolemia, a leading cause of heart disease, and stroke.

Hypercholesterolemia is a condition characterized by abnormally high levels of cholesterol in the bloodstream.

The condition is a result caused by either overexpression of low-density lipoprotein (LDL) or low levels of high-density lipoprotein (HDL).


 People with hypercholesterolemia at the early stage are asymptomatic. However, patients with familial hypercholesterol may experience symptoms of chest pain with activity. xanthomas, and cholesterol deposits around the eyelids.

According to the statistics provided by the CDC, hypercholesterolemia is a major risk factor of heart disease, a leading cause of death in the US.

In the US, approximately, 71 million American adults (33.5%) have high low-density lipoprotein (LDL), or “bad,” cholesterol. and only 1 out of every 3 adults with high LDL cholesterol has the condition under control.

Sadly, only 50% of people hypercholesterolemia are getting treatment.

Out of many risk factors associated with the risk of hypercholesterolemia, some researchers suggested that the promotion of a high-fat diet over the past few decades may have a strong impact on the increased incidence of hypercholesterolemia in the US.

Dr. ROBERT B. KELLY, the lead scientist wrote, "The most beneficial changes result from reducing intake of saturated and trans fats; increasing intake of polyunsaturated and monounsaturated fats; fortifying foods with plant stanols or sterols; isocalorically adding tree nuts to the diet; consuming one or two alcoholic drinks per day; and adopting a Portfolio, Mediterranean, low-carbohydrate, or low-fat diet".

Turmeric is a perennial plant in the genus Curcuma, belongings to the family Zingiberaceae, native to tropical South Asia.


The herb has been used in traditional medicine as anti-oxidant, hypoglycemic, colorant, antiseptic, wound healing agent, and to treat flatulence, bloating, and appetite loss, ulcers, eczema, inflammations, etc.

On findings a potential compound for the treatment of hyperlipidemia, researchers examined the efficacy of curcumin on hypercholesterolemia, one of the most important risk factors for atherosclerosis and subsequent cardiovascular diseases.

The experimental study included selected 24 male Sprague-Dawley rats that were divided into six groups, namely normal group diet, high-cholesterol diet (HCD) 2%, HCD + 100 mg/kg b.w. curcuminoid fraction, HCD + 300 mg/kg b.w. curcuminoid fraction, HCD + cider 1% v/v, and HCD + curcuminoid cider 2% v/v for 4 weeks.

According to the tested analysis of total cholesterol levels measured at day 1, 14, and 28, curcuminoid cider at 2% v/v significantly lowered total cholesterol levels compared to the positive control.

Furthermore, curcuminoid fractions at 100 and 300 mg/kg) and curcuminoid cider (1 and 2% v/v) inhibited the gene expression of CD44 associated with membrane cholesterol and intercellular adhesion molecule (ICAM)-1involved in the measurement of lipid metabolic impairment.

Moreover, curcuminoid fractions at 100 and 300 mg/kg) and curcuminoid cider (1 and 2% v/v) also lower the levels of iNOS, which has been found to mediate HCD-induced liver fibrosis.

Additionally, the applications also inhibited LOX-1 associated with high circulating cholesterol levels.

In other words, curcuminoid fractions at 100 and 300 mg/kg) and curcuminoid cider (1 and 2% v/v) displayed their hypocholesterolemic effects via attenuating genes related to oxidative stress in vivo.

Taken altogether, turmeric processed abundantly bioactive compound curcumin may be considered supplements for the prevention and treatment of hypercholesterolemia, pending to the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Herbal Ashwagandha Protects the Beta Cells of Pancreas Against the Onset of Diabetes in Vivo

Diabetes is a chronic condition caused by insufficient insulin entering the bloodstream. Insulin resistance and hyperglycemia are hallmarks of diabetes.

In other words, insulin produced by the pancreas is either blocked by the clogging-up of the insulin receptors by fat and cholesterol or the death of beta cells in the pancreas. In some cases, diabetes can also be associated with autoimmune disease, attacking the beta cells.

There are some major types of diabetes, including the best known, type 1 and 2.
* Type 1 diabetes is associated with a genetic proposition that leads to the immune system to attack insulin-producing cells of the pancreas.

* Type 2 diabetes (acquired diabetes), is a condition caused by either the pancreas doesn't produce enough insulin, or the insulin does not work properly due to dysfunction of the insulin receptors.

In pregnancy, gestational diabetes is a type of diabetes occurred during the second or third trimester of pregnancy. Gestation diabetes goes away after giving birth.

However, omen with gestation diabetes is associated with a substantial risk of diabetic developing sometimes in the future.

Epidemiologically, obesity, an unhealthy lifestyle with physical inactivity are some of the most common risk factor associated with the slow onset of diabetes.

Interestingly, some researchers suggested, night shifts workers accompanied by unhealthy lifestyles are linking to a high risk of type 2 diabetes.

Ashwagandha also is known as Withania somnifera is a nightshade plant in the genus of Withania, belonging to the family Solanaceae, native to the dry parts of India, North Africa, Middle East, and the Mediterranean.
The herbal medicine has been considered as Indian ginseng and used in Ayurvedic medicine over 3000 years to treat tumors and tubercular glands, carbuncles, memory loss, and ulcers and considered as anti-stress, cognition-facilitating, anti-inflammatory, and anti-aging herbal medicine.

With an aim to find a potent compound for the treatment of diabetes, researchers examined the effects of Withania somnifera (WS) on lipid peroxidation (LPO) and pancreas in type 2 diabetic rats.

The study included type 2 diabetes induced by a single intraperitoneal injection of STZ (100 mg/kg) to 2 days old rat pups pretreated by WS.

According to the tested analysis, untreated rats group showed significant levels of lipid peroxidation (LPO) observed by reducing levels of antioxidant enzymes produced by the host including glutathione (GSH) contents and by enzymatic activities of glutathione peroxidase (GPx), glutathione reductase (GR), glutathione-S-transferase (GST), superoxide dismutase (SOD) and catalase (CAT).

However, injection of WS not only caused a significant decrease in blood glucose and tissue LPO levels casued by the application of STZ but also a significant increase in GSH contents, compared to type 2 diabetic control rats.

Furthermore, antioxidant enzymes reduced in the untreated rat such as GPx, GR, GST, SOD, and CAT were also increased compared to type 2 diabetic control rats.

Most importantly, the significant reduction in the number and size of pancreatic beta-cells in STZ rats was normalized upon the administration of WS, evident from the histopathological examination.

In other words, WS protects the cells in the pancreas against oxidation and maintains the integrity of beta cells in insulin production in type 2 diabetes rats.

Based on the findings, researchers said, "WS has shown strong free radical scavenging activity and helped in improving the non-enzymatic and enzymatic antioxidants in type 2 diabetic rats".


Taken altogether, Ashwagandha WS may be considered a functional herb for the prevention and an adjunct therapy combined with the primary medicine for the treatment of type 2 diabetes, pending to the results of large example size and multicenter study.
