Herbal Turmeric Inhibits Human Papillomavirus-Positive and Negative Squamous Carcinoma Cells

Cancer is a group of chronic diseases associated with cell growth disorderly in the tissue of some specific organ. Depending on the grade and stage, most cases of cancer in the advanced stage has been found to induce a higher mortality rate compared to cancer diagnosed in the localized stage.

Head and neck cancer is a group of cancers that starts in the cell on the surface of the inner lining of the mouth, nose, throat, larynx, sinuses, or salivary glands.

Squamous cell carcinoma is cancer arisen from the squamous cells found in the outer layer of skin and in the mucous membranes, which are the moist tissues that line body cavities.

 Head and neck squamous cell carcinoma (HNSCC) develops in the mucous membranes of the mouth, nose, and throat.

Head and neck squamous cell carcinoma (HNSCC) epidemiologically ahs have been found to spread to the lymph nodes or lungs. More than 90% of the head and neck cancer diagnosed are squamous carcinomas.

Symptoms for head and neck cancer may include a lump or sore that does not heal, a sore throat that does not go away, trouble swallowing, or a change in the voice.

According to the statistics provided by the Western General Hospital, squamous cell cancer of the head and neck is common worldwide accounting for 4% of all cancers in the United States and  5% in the United Kingdom.

The exact causes of head and neck squamous cell carcinoma (HNSCC) are unknown. However, geographically, smoking tobacco, drinking alcohol, and long term poor diet is important to risk factors in the West compared to risk factors of chewing betel or areca nuts, smoking bidis, and taking snuff are important in the Indian subcontinent. 

More importantly, the genetic preposition of genome 9p21, which may indicate loss of a gene that suppresses tumors accounts for a total of 70% of all cases.

Turmeric is a perennial plant in the genus Curcuma, belonging to the family Zingiberaceae, native to tropical South Asia.

The herb has been used in traditional medicine as anti-oxidant, hypoglycemic, colorant, antiseptic, wound healing agent, and to treat flatulence, bloating, and appetite loss, ulcers, eczema, inflammations, etc.

On finding a potential ingredient for the treatment of head and neck cancer, researchers examined the effect of curcumin on head and neck squamous cell carcinoma (HNSCC) using a panel of cell lines with and without human papillomavirus (HPV+, HPV-).


According to the tested differentiation, curcumin treatment of HNSCC cell lines not only interfered RNA of Skp2, that causes cancer tumorigenesis and radiation tolerance but also induced concomitant accumulation of p21 and p27 associated with a cells cycle arrest in HPV+, HPV- cells.

Furthermore, curcumin inhibits cells viability and induces apoptosis in a dose-dependent manner in the tested cell line with and without human papillomavirus (HPV+, HPV-).

Moreover, curcumin treatment of HPV+ and HPV- cells resulted in apoptosis via the mitochondrial pathway of apoptosis and activation of caspases that play a critical role in programmed cell death.

Additionally, the inhibition of HPV+ and HPV- cells by curcumin was also found to suppress the expression of poor prognostic markers such as XIAP, and cancer survival such as cIAP1, and cIAP2.

More importantly, co-treatment of HNSCC cells with curcumin and cisplatin potentiated inhibition of cell viability and apoptotic effects.

Based on the findings, researchers wrote in the final report, "these data suggest an important function for curcumin, acting as a suppressor of oncoprotein Skp2 in squamous cell carcinoma cells in both HPV+ and HPV- cells".

Taken altogether, turmeric processed abundantly bioactive compound curcumin may be considered supplements for the prevention and treatment of HNSCC, pending to the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Carotenoids do not cause Skin Cancer

The skin is a large and outer layer organ of the human body that not only protects the internal tissues and organ against the invasion of foreign pathogens but also serve as the third detoxified organ after the liver and kidney.

Furthermore, the skin also plays an essential role to regulate body temperature and permit the sensations of touch, heat, and cold.

The skin contains 3 layers
* The epidermis, the outermost layer of skin.

* The dermis, beneath the epidermis.

* The deeper subcutaneous tissue (hypodermis).

Skin cancer is a medical condition caused by uncontrolled growth of abnormal skin cells and often detected at an early stage.
According to the statistics, every year, approximately 4.3 million cases of basal cell cancer (BCC) are diagnosed in the U.S. compared to over 1 million cases of  Squamous cell carcinoma (SCC) which is the second most common form of skin cancer. 


There are three most common skin cancers, depending on the cancer cells originated
* Basal cell cancer (BCC)
Basal cell cancer is the most common type of skin cancer caused by abnormal growth of the cells in the lowest layer of the epidermis, accounting more than 90% of all skin cancer in the U.S. Basal cell cancer rarely spreads and kills but can cause significant destruction and disfigurement to the affected skin area.

* Squamous cell cancer
Squamous cell cancer arises from the uncontrolled multiplication of transformed malignant cells in the middle layer of the epidermis.

*Melanoma
Melanoma, a kind of skin cancer but less common to other skin cancer is a result of malignancy of melanocytes, produced dark pigment for your skin. 

Epidemiologically, long-term exposure to the sunlight, family history, age, gender, chemical exposure, r\exposure to radiation, smoking, and history of sunburns are some prevalent factors associated with the risk of the disease.

Carotenoids is a class of mainly yellow, orange, or red fat-soluble pigments, including lycopene and carotene, found abundantly in ripe tomato, pumpkins, carrots, corn, and daffodils.

In concern of overexpression of carotenoids enhancing the risk of skin cancer, researchers investigated the associations of several carotenoids and alpha-tocopherol with the risk of nonmelanoma skin cancer.

The study included serum carotenoids and alpha-tocopherol collected at baseline from 302 subjects who had at least two BCCs in the 5 years prior to randomization in the Isotretinoin-Basal Cell Carcinoma Prevention Trial.

During 5 years of follow-up, 70 subjects did not develop nonmelanoma skin cancer, 221 developed a BCC, and 85 developed a squamous cell carcinoma (SCC).

In chemical analysis, researchers found that
* Serum levels of any of the carotenoids measured or to alpha-tocopherol are not associated with the risk of developing a subsequent BCC.

* Furthermore, serum levels of alpha-carotene, beta-carotene, lycopene, and alpha-tocopherol found in carotenoids also were not independently related to the risk of a subsequent SCC.

* However, serum lutein, zeaxanthin, and beta-cryptoxanthin were positively related to SCC risk with a risk ratio for subjects in the highest versus lowest tertiles of these micronutrients were 1.63.

In other words, overexpression of lutein, zeaxanthin, and beta-cryptoxanthin increased risk  SCC compared to no risk of carotenoids.

Based on the findings, researchers said, "Additional research is needed on the relationship of carotenoids to SCC risk in the general population and in subsets of the population who are at increased risk".

Taken altogether, dietary carotenoids does not increase the risk of skin cancer, pending to the confirmation of the larger sample size and multicenter human study.

Intake of carotenoids in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Bromelain Kills Bile Duct Cancer Cells in Vitro

Cholangiocarcinoma, also known as bile duct cancer, is a medical condition that originated from the irregular cell growth in the bile ducts.

Most cases of cholangiocarcinoma are caused by the alternation of cell DNA in the cell on the surface of the inner lining of the bile duct.

At the advanced stage, cancer cells in the bile duct may travel a distance away to infect other healthy tissues and organs through the circulation of blood and fluids.

The primary bile duct cancer has been found to spread to the regional lymph nodes and adjacent organs.

According to statistics, cholangiocarcinoma is a rare cancer that occurred approximately 2500 Americans each year compared to 5000 cases of gallbladder cancer and 15,000 cases of hepatocellular cancer.

Believe it or not, the 5-year survival rate diagnosed at early-stage extrahepatic bile duct cancer is only 30%. If cancer has spread to the regional lymph nodes, the 5-year survival rate is 24%.

The most common type of cholangiocarcinoma found in the bile duct is perihilar cholangiocarcinoma, followed by the distal extra-hepatic and then the intrahepatic forms.

Epidemiologically parasitic infections, primary sclerosing cholangitis, biliary-duct cysts, hepatolithiasis, and toxins are some prevalent factors that cause the onset of cholangiocarcinoma.

Dr. Gia L. Tyson, the lead scientist wrote, "Cholangiocarcinoma is the second most common primary hepatic malignancy after hepatocellular cancer. It accounts for approximately 10–25% of all hepatobiliary malignancies".

And, "Other less-established, potential risk factors include inflammatory bowel disease, hepatitis C virus, hepatitis B virus, cirrhosis, diabetes, obesity, alcohol drinking, tobacco smoking, and host genetic polymorphisms".

However, most people with some of the above risk factors do not develop cholangiocarcinoma. 

Bromelain, a proteolytic enzyme found in pineapples (Ananas comosus) has been used in traditional medicine as an inflammatory agent and to treat pains, strains, and muscle aches and pains and ease back pain and chronic joint pain, skin diseases, etc.

With an aim to find a natural ingredient against Cholangiocarcinoma (CC), researchers investigated the anticancer effect of bromelain and papain in intra- and extrahepatic human CC cell lines.

According to the medical assays, bromelain and papain showed a similar effect inhibition of human CC cell growth, migration, invasion.

Furthermore, according to cell proliferative assays, bromelain and papain resulted in a decrease in the proliferation, invasion, and migration of CC cells.

Bromelain and papain-induced CC death by inhibiting protein associated anti-apoptotic signaling as well as their downstream signaling proteins associated with CC proliferation.

Interestingly, compared to papain, bromelain showed an overall more effective inhibition of CC.

Additional differentiation of the anti-cancer activity, researchers found that bromelain and papain have cytotoxic effects on human CC cells.


Based on the findings, Dr. Müller A the lead scientist said, "bromelain and papain have cytotoxic effects on human CC cell lines and bromelain and partially papain in comparison impair tumor growth by NFκB/AMPK signaling".
And, " Especially bromelain can evolve as a promising, potential therapeutic option that might open new insights for the treatment of human CC".

The topical application of the crude extract also induced significant improvement effects on the UV-induced skin structure damage.

By examining the efficacy of constituent of the extracts, researchers found that total CUR content was correlated with the anti-aging activity.

Based on the findings, researchers said, "The results show that C. heyneana contains antioxidant compounds and has potent anti-aging activity, indicating that it can be used as an anti-aging drug candidate or as a phyto-cosmeceutical".

Herbal Cranberry, a Potential and Natural Prevention and Treatment of Esophageal Tumor

The esophagus formed part of the digestive system is a muscular tube that connects the throat (pharynx) with the stomach located behind the trachea and heart and in front of the spine.

Cancer is a group of chronic diseases associated with irregular cell growth that start in the cell on the surface of the inner lining tissue of the specific gland and organ before penetrating into deeper layers to form tumors through unlimited cell cycle division.

Over time, most cancers have become progressively worse and spread to other organs in the body via lymph or blood.

 Esophageal cancer is not very uncommon in the US caused by malignant cells of the esophagus due to cell DNA alternation.

Primary esophageal cancer has been found to spread to the bottom of the esophagus, stomach, and chest.

According to the statistics, compared to the US, esophageal cancer is more often diagnosed in other parts of the world and the seventh most common cause of cancer death among men.

Believe it or not, every year, the disease causes the death of 16,000 individuals, including 13,000 men and 3,000 women,

Compared to other cancers the 5-year survival rate for people with esophageal cancer is only 19%.

The exact causes of esophageal cancer are unidentified. However, according to the epidemiological studies, aging, male gender, smoking, excessive alcohol drinking, genetic preposition, obesity, thermal injury to the esophagus are some prevalent factors that increase the risk of the disease.

Some researchers suggested that the increased incidences of esophageal cancer in the Western world over the past few decades may be correlated to the promotion of a high-fat diet.

Dr. Silvano Gallus wrote, "Over the last 30 years, the incidence of squamous-cell carcinoma has decreased in several areas of the world, but adenocarcinoma has risen by over fourfold in the US as well as in Australia and several areas of Europe; its incidence has particularly increased in men".

And, " diets high in saturated fat and cholesterol have been associated with an increased risk (of esophageal cancer)".

Cranberry is an evergreen dwarf shrub, genus Vaccinium, belongings to the family Ericaceae, native to Northern America and Southern Asia. Because of its health benefits, cranberry has been cultivated in some parts of the world for commercial profit and used in traditional and herbal medicine to treat wounds, urinary disorders, diarrhea, diabetes, stomach ailments, and liver problems.

On finding a potential compound for the treatment of gastrointestinal tumor, researchers examined the effect of a purified cranberry-derived proanthocyanidin extract (C-PAC) on esophageal adenocarcinoma (EAC) cell lines, including acid-sensitive JHAD1 and OE33 cells, and esophageal tumor xenografts in athymic NU/NU mice.

According to the tested assays, C-PAC shared a strong activity in inducing the expression of a protein associated with cell death programming mainly in JHAD1 and OE33 cells.

Autophagy that promotes cancer cell growth and anti-apoptosis of JHAD1 and OE33 cells were also inhibited by the injection of C-PAC.

In vitro, C-PAC induced cell death was attributed to deactivation of the phosphatidylinositol-3-kinase (PI3K)/Akt and the mammalian target of rapamycin (mTOR) signaling pathways expression in the promotion of cancer cellular processes, including cell survival, growth, and differentiation.

Additionally, C-PAC not only inhibited the levels of anti-apoptotic proteins such as BCL-xL, Cytochrome C, PARP), pathways involved in the occurrence and progression of cancer, but also exhibited the expression of proteins that induces cell cycle arrest.



Similarly, in vivo, oral delivery of C-PAC significantly inhibited OE19 tumor xenograft growth as the aforementioned activities.

In other words, in vitro and vivo purified cranberry-derived proanthocyanidin extract (C-PAC) eradicate the esophageal adenocarcinoma (EAC) cells through several mechanisms involved in pro-apoptosis, anti-anti-apoptosis and blocking cellular division.

Based on the finding, researchers after taking other factors into account wrote, " C-PAC is a potent inducer of EAC cell death and is efficacious in vivo at non-toxic behaviorally achievable concentrations, holding promise for preventive or therapeutic interventions in cohorts at increased risk for EAC, a rapidly rising and extremely deadly malignancy".


Taken altogether, cranberry extract may be considered a remedy for the prevention and treatment of esophageal cancer, pending to the confirmation of the larger sample size and multicenter human study.

Healthy Food Onion Inhibits Lung Cancer in Vivo

The lung is a vital organ formed part of the respiratory system located near the backbone on either side of the heart that inhales oxygen from the air to the bloodstream and exhales carbon dioxide from the bloodstream.

Lung cancer is a medical condition caused by the abnormal growth of the cells in the lung's tissue. Most common form of primary lung cancers is derived from epithelial cells. 

According to the statistic, in the US, approximately 541,000 Americans are living with lung cancer at some points in their lives. In 2018, approximately 234,000 new cases of lung cancer were diagnosed, representing about 13 percent of all cancer diagnoses.

The exact causes of lung cancer are unknown. Epidemiologically, the prevalent factors associated with an increased risk of lung cancer are
*Smoking
Smoking is a major cause of lung cancer by causing oxidation stress to the lung due to chemical substances (carcinogens) in the cigarette over time.

* Second-hand smoke
Prolonged exposure to second-hand smoke can also increase the risk of lung cancer.

* Exposure to certain chemicals
 Risk of lung cancer is increased for people who inhale or expose certain industrial chemical such as radon gas, arsenic, chromium, nickel, etc. in the workplace.

* Family history
Risk of lung cancer increased if one of the members of the direct family has lung cancer.

*  Lung disease
Risk of lung cancer is increased if you have or previously had certain types of lung disease

* Excessive alcohol drinking
Alcohol intake of more than 2 drinks per day is associated with a small increased risk of lung cancer.

* Quit smoking
According to statistic, approximate 87% of lung cancer deaths are caused by smoking.

*  Eating more fruits and vegetables
These foods contain a high amount of antioxidants which enhance the immune system against the forming of free radicals and prevent the alternation of cell DNA cause of abnormal cell growth.

* Avoid Second smokes
Second smoke contains the same amount of carcinogens, exposure to it can increase the risk of lung cancer.

* Avoid environmental chemicals
Avoid exposure to certain industrial chemicals such as radon gas, arsenic, chromium, nickel, etc.. In the workplace, deal with them by taking maximum precautions and prevention.

* Reduced alcohol intake
Researchers found that increased risk of lung cancer for people who have more than 2 drinks a day.

The onion is a plant in the genus Allium, belonging to the family Alliaceae, a close relation of garlic. It is often called the "king of vegetables" because of its pungent taste and found in a large number of recipes and preparations spanning almost the totality of the world's cultures. 

Depending on the variety, an onion can be sharp, spicy, tangy, pungent, mild or sweet.

On finding a potential compound for the treatment of cancer, researchers examined the effects of onionin A (ONA) derived from the onion in the inhibition of tumor-associated macrophages in the M2 phenotype.

According to the tested analysis.
* Onionin A (ONA) inhibits macrophage polarization into the M2 phenotype associated with tumorigenesis by antagonizing the cytotoxic activity immune cells.

* Onionin also exerted immunosuppressive activity and tumor proliferation, by suppressing signal transducer and activator associated with tumor proliferative promotion.

* In lung tumor, ONA administration significantly suppressed subcutaneous tumor development and lung metastasis in tumor-bearing mice.

* ONA also increased the number of infiltrating lymphocytes against the tumor proliferation in tumor tissues.

The efficacy of  ONA against the onset and progression of od lung cancer was probably involved in the regulation of tumor proliferation by inhibiting cell-cell interactions between macrophages and tumor cells.

Based on the finding, researchers said, "NA administration enhanced the antitumor effects of cisplatin in the tumor-bearing mice".

Taken altogether, onion processed a higher mount of onionin A (ONA) may be considered a remedy for the prevention and treatment of lung cancer, pending to the confirmation of the larger sample size and multicenter human study.

Herbal Ashwagandha the Anti Rheumatoid Arthritis Herbal Medicine

Rheumatoid Arthritis, a type of musculoskeletal disorders (MSDs) is a chronic disorder characterized by inflammation of the flexible (synovial) joints and tissues and organs in the body.

The disease affects more women than men and generally occurs after the ages of 50, causing the diminished quality of life of many elders.

Musculoskeletal disorders (MSDs) are medical conditions mostly caused by work-related occupations and working environments, affecting patients’ muscles, joints, tendons, ligaments and nerves and developing over time.

According to the statistics provided by the CDC, musculoskeletal disorders (MSDs) affects over 52 million adults in the US alone, including 294,000 children under age 18 with some form of arthritis or rheumatic conditions.

Rheumatoid arthritis was found to induce bone loss through elevating bone resorption without increasing bone formation.

A cross-sectional population-based study of 1042 patients with rheumatoid arthritis showed that RA patients had an increased risk of death from various causes.

Patient with rheumatoid arthritis (RA) often associates to valgus deformities of the feet and deformities of gait due to body function due to an effort to support the collapsing longitudinal arch of the foot. Ashwagandha also is known as Withania somnifera is a nightshade plant in the genus of Withania, belonging to the family Solanaceae, native to the dry parts of India, North Africa, Middle East, and the Mediterranean.

The herbal medicine has been considered as Indian ginseng and used in Ayurvedic medicine over 3000 years to treat tumors and tubercular glands, carbuncles, memory loss, and ulcers and considered as anti-stress, cognition-facilitating, anti-inflammatory, and anti-aging herbal medicine.

On finding a potent compound for the treatment of arthritis, researchers examined the effect of Cannabis sativa, Prunella vulgaris, and Withania somnifera) possess anti-inflammatory, and-rheumatic arthritis properties.

According to the tested analysis, the tested medical plants showed a significant effect on RA by targeting TNF-α through its anti-inflammatory activity.

More precisely, twenty unique molecules found in the medical plants including Withania somnifera were attributed to the utmost binding affinity, least binding energies associated with the interaction of receptor-ligand interactions of residues against RA.


Based on the findings, researchers said, " the RA patients should use reported compounds for the prescription of RA by targeting TNF-α. This report is opening new dimensions for designing innovative therapeutic targets to cure RA.".

Taken altogether, Ashwagandha may be considered a functional herb for the prevention and an adjunct therapy combined with the primary medicine for the treatment of RA, pending to the result of larger sample size and multicenter study
