Artichoke Protects Your Cells Integrity Against Carcinogens

By Kyle J. Norton

Substances such as radionuclides, or radiation which can cause the onset of cancer with or without changing the cell DNA are considered carcinogens.

Contrast to general belief, unhealthy lifestyle, long term exposure to the sunlight, certain medical treatment such as radiation, and medications such as immune suppressive drugs, chemicals exposure may also be considered carcinogens.

However, most people exposed to the substances have never developed cancer.

According to the World Health Organization (WHO), substances that cause cancer in human can be classified into 5 categories.
Group 1: Carcinogenic to humans
Group 2A: Probably carcinogenic to humans
Group 2B: Possibly carcinogenic to humans
Group 3: Unclassifiable as to carcinogenicity in humans
Group 4: Probably not carcinogenic to humans

The most dangerous carcinogens are placed in group 1. Therefore, if you are handling any of the substances in the group 1 (the list can be found here), please make sure you protect yourself according to the standard guidelines.

Artichoke is a perennial thistle of Cynara cardunculus species of the Cynara genus, belonging to the family Carduoideae native to Southern Europe around the Mediterranean.

The herbal plant has been used in traditional medicine for as liver protective and detoxified agent, and to treat digestive disorders, abdominal pain gas and bloating, etc.

Researchers on the finding natural therapeutics which block carcinogen in the induction of cancer investigated the antigenotoxic and antioxidant effects of the extract of artichoke Cynara cardunculus L. (CCE).

In the Saccharomyces cerevisiae mutagenicity/antimutagenicity assay, CCE significantly reduced the frequency of 4-nitroquinoline-N-oxide-induced genetics alternation in the diploid Saccharomyces cerevisiae tester strain D7.

In the simultaneous toxicity and clastogenicity/anticlastogenic assay, CCE exerted an anticlastogenic effect against N-nitroso-N'-methyl urea-induced clastogenicity in the plant species.

According to the free radical scavenging assay, CCE also exhibited considerable antioxidant activity against carcinogens.

In order to reveal more information of artichoke, anticarcinogenic activity researchers examined the genotoxicity of C. scolymus L. leaf extract (LE) and the ability to modulate the genetic toxicity of the alkylating agent ethyl methanesulfonate (EMS), using the comet assay on Chinese hamster ovary cells.

LE demonstrated strong anti genotoxicity after 1- and 24-h treatments by 4 different doses: 0.62, 1.25, 2.5, and 5.0 mg/mL, according to the results from pretreatment, simultaneous treatment, and post-treatment.

All doses also protect the cell against DNA damage induced by the alkylating agent ethyl methanesulfonate (EMS), after exposure for 1 and 24 h.

Interestingly, the lowest dose of LE was more protective than higher concentrations.

The genotoxic and antigenotoxic effects were attributed to the bioactive flavonoids and phenolic compounds found abundantly in artichoke.

Taken altogether, artichoke processed bioactive compound flavonoids and phenolic may be considered a functional remedy for the prevention and treatment of carcinogenic toxicity, with no side effects, pending to the validation of larger sample size and multicenter human study.

Bilberry Protects the Retinal Ganglion Cell Integrity Against Vision Loss

By Kyle J. Norton

Retinal ganglion cell (RGC) is a type of neuron in the innermost cellular layer of the retina of the eye. The neurons receive visual information and pass them to the brain.

Believe or not, the neurons also extract different aspects of the image before passing them to the brain.

According to the biological information from an animal model, retinal ganglion cells distributed evenly across the retina, with the density ranges from ∼3000 cells per mm2 to ∼600 cells per mm2 depending on the areas of distribution.

Certain neurodegenerative diseases have been found to induce the gradual loss of retinal ganglion cells. Alzheimer's' disease and disease associated with multiple system atrophy the loss of retinal ganglion cells showed a pattern resembling glaucoma.

However, in patients with Parkinson’s disease (PD) and Huntington’s disease, the loss of RGCs are found to resemble that of mitochondrial optic neuropathies.

Any structural damage of the retinal nerve fiber layer and optic nerve head, or functional damage can affect the RGC integrity.

Symptoms of retinal ganglion cell (RGC) loss is depended on the level of damage of the retinal nerve fiber layer and optic nerve head in the retina. Symptoms can be severe eye pain accompanied by nausea and vomiting, visual disturbance, blurred vision in patients with glaucoma.

Bilberry is a species of low-growing shrubs in the genus Vaccinium, belongings to the family Ericaceae, native to Northern Europe.

The plant berry has been used as herbs in traditional medicine for the treatment of acute and chronic diarrhea, gastritis, gastric ulcer, and duodenal ulcer, enterocolitis, ulcerative colitis, anemia, cystitis, kidney disease, and psoriasis, diabetes, etc.

In the urgency on finding a plant-based compound for the treatment of retinal damage, researchers evaluated the protective effects of a Vaccinium myrtillus (bilberry) anthocyanoside (VMA) and/or its main anthocyanidin constituents (cyanidin, delphinidin, and malvidin) on retinal ganglion cells (RGCs) against damage.

In vitro, retinal ganglion cells damage induced by free radical activation by injection of 3-(4-morpholinyl) sydnonimine hydrochloride (SIN-1, a peroxynitrite donor) and in vivo, N-methyl-D-aspartic acid (NMDA)-induced retinal damage in mice, researchers showed that VMA and all three anthocyanidins significantly inhibited SIN-1-induced neurotoxicity and radical activation in RGC-5 associated with the initiation of damage of the retinal ganglion cells.

Lipid peroxidation in mouse forebrain homogenates involved the retinal damage was also reduced by all tested compounds in concentration and dose-dependent manner.

Furthermore, intravitreously injected VMA significantly inhibited the NMDA-induced morphological retinal damage and an increase in apoptosis of DNA fragments in the ganglion cell layer.

Based on the findings, researchers suggested that bioactive compounds isolated bilberry process neuroprotective effects through antioxidative properties in vitro and in vivo models of retinal diseases.

In order to obtain more information about bilberry protective property against the damage of retinal ganglion cell, scientists analyzed the bilberry extract anthocyanins on retinal ganglion cell (RGC) survival after optic nerve crush.

C57BL/6J mice selected to the study were administrated orally with anthocyanins in bilberry extract (100 mg/kg/day or 500 mg/kg/day).

According to the tested assays, all doses of anthocyanins in bilberry extract exhibited the increased significantly levels of Grp78 and Grp94 associated with endoplasmic reticulum integrity and stress-induced autophagy in mice detected in the inner nuclear layer and ganglion cell layer of the retina, surrounding the RGCs.

Gene expression of Chop, Bax, and Atf4 increased associated with cellular stress and the mediation of apoptosis in mice after optic nerve crush was also decreased significantly after oral bilberry extract administration.

In other words, retinal ganglion cell (RGC) survival was increased substantially after injecting anthocyanins in bilberry extract.

Dr. Nakamura O, the lead scientist after taking into account co and confounders wrote, "bilberry extract has a potential role in neuroprotective treatments for retinal injuries, such as those which occur in glaucoma".

Taken altogether, bilberry processed abundantly bioactive compound anthocyanin may be considered a functional remedy for the protection of retinal damage against retinal diseases, particularly in retinal degeneration, pending to the validation of larger sample size and multicenter human study.

Honey, the Functional Food For the Treatment of Diseases Associated with Chronic Inflammation

By Kyle J. Norton

Inflammation is a natural response of the immune system in which the white blood cells of the first line of defense try to protect the body by destroying the foreign pathogens which try to enter the body through wound or tissues damage.

During the acute phase of infection, overproduction of proinflammatory cytokines may cause damage to nearby healthy cells, leading to the formation of scars.

Most cases of infection are stopped at the acute phase of infection as the immune system successfully kills off all invaders in a brief battle.

Some researchers suggested the immune system has evolved to protect our body over thousands of years with an aim to kill off all pathogens within a period in the range of 3 to 8 weeks. Otherwise, it will adapt to the new change, leading to chronic inflammation.

In other words, chronic inflammation happens when the immune response lingers, causing low-grade inflammation. Chronic inflammation over time may damage the body tissues and organs.

In some cases, the infection may be latent in which the pathogen remains within the body for a long time without producing any symptoms, such as the hepatitis virus.

Diseases associated with chronic inflammation include cancer, heart disease, rheumatoid arthritis,  type 2 diabetes, and obesity.

Honey, the rich golden liquid is the miraculous product made by bees using nectar from flowers.

The liquid is considered one of the healthy food for replacing the use of white sugar and artificial sweetener by many people.

With an aim to find a potential compound for the treatment of diseases associated with chronic inflammation, researchers at the Universiti Sains Malaysia investigated the anti-inflammatory property of honey among smokers.

The randomized, controlled, the open-label trial included 10 New Zealand white rabbits sensitized twice with a mixture of OVA and aluminum hydroxide on days 1 and 14 to induce chronic inflammation.

The honey was given from day 23 to day 25 at two different doses (25% (v/v) and 50% (v/v) of honey diluted in sterile phosphate buffer saline.

In the aerosolized honey as a rescue agent group, animals were euthanized on day 28 and in the preventive group, animals were further exposed to aerosolized OVA for 3 days starting from day 28 and euthanized on day 31.

According to the assays, aerosolized honey protected the structural changes of the epithelium, mucosa, and submucosal regions of the airway induced by OVA.

Moreover, the aerosolized honey treatment also reduced the number of airway inflammatory cells in bronchoalveolar lavage fluid and inhibited the goblet cell hyperplasia in tested rabbits.

The results strongly suggested that aerosolized honey effectively treats and manage asthma in rabbits.

In order to reveal more information about honey anti chronic inflammatory activity, researchers examined the 12-week honey oral supplementation on plasma inflammatory markers.

The study included a total of 32 non-smokers and 64 chronic smokers from Quit Smoking Clinic and Health Campus, Universiti Sains Malaysia with Smoker randomized into 2 groups: smokers with the honey group that received Malaysian Tualang honey (20 g/day daily for 12 weeks) and smokers without the honey group.

According to the blood sample collected from the subjects before the treatment, smokers had significantly higher high sensitive C-reactive protein compared to non-smokers.

Treatment with aerosolized honey showed to reduce the number of airway inflammatory cells in bronchoalveolar lavage fluid and inhibit the goblet cell hyperplasia.

Compared to the increase of proinflammatory cytokines (tumor necrosis factor-α) and high sensitive C-reactive protein in smokers group, honey treatment significantly increased the levels of tumor necrosis factor-α) but reduced levels of sensitive C-reactive protein at post-intervention than at pre-intervention.

In other words, honey treatment inhibited the progression of inflammation by reduced proinflammatory cytokines levels of sensitive C-reactive protein but increased tumor necrosis factor-α).

Taken altogether, honey may be considered a functional remedy for the treatment of chronic inflammation with no side effects, pending to the confirmation of large sample size and multicenter human study.

Phytochemical Alliin Isolated from Garlic Gobbles Up Breast Cancer Cells in Vitro

By Kyle J. Norton

Breast cancer is the medical condition caused the irregular cell growth in the breast tissue due to the alternation of cell DNA.

According to the statistic, in 2010, over 250,000 new cases of breast cancer were diagnosed in women in the U.S. and the risk of getting invasive breast cancer during the lifetime of a woman is 1/8.

Most cases of breast cancer begin in the cells on the surface of inner lining tissues of the breast either from the inner lining of milk ducts (Ductal carcinoma) or the lobules (Lobular carcinoma) that supply the ducts with milk.

At the early stage, breast cancer is a painless lump in the breast or armpit. However, at the later stage, most women with breast cancer may experience symptoms of nipple discharge (clear or bloody)
inverted or retracted nipple, scaly or pitted skin on nipple, persistent tenderness of the breast and
unusual breast pain or discomfort.

According to statistic, the 5 years survival rate of early diagnosed breast cancer is closed to 100%.

However, in the rare case aggressive inflammatory breast cancer (IBC) that induces the growth in sheets instead of lumps and invades nearby skin, the whole breast may go suddenly red and feel hot, resembling an inflammation.

The disease only occurs in 1-6% of all breast cancer cases in the USA. Women with IBC are first treated with chemotherapy before surgery. On average, the 5-year survival rate of women diagnosed with inflammatory breast cancer is 34% compared to the early stage of breast cancer of 100%.

Out of many risk factors associated with the risk of breast cancer, the genetic proposition is one of the factors that may predict the onset of the disease.

Dr. Turnbull C, the lead scientist said, "BRCA1 and BRCA2 are high-penetrance breast cancer predisposition genes identified by genome-wide linkage analysis and positional cloning. Mutational screening of genes functionally related to BRCA1 and/or BRCA2 has revealed four genes, CHEK2, ATM, BRIP1, and PALB2; mutations in these genes are rare and confer an intermediate risk of breast cancer".

Alliin (S-allyl-L-cysteine-S-oxide) is a phytochemical compound sulfoxide, and a derivative of the amino acid cysteine, belonging to the class of sulfur compounds, found abundantly in fresh garlic and onion.

On finding a natural potential for the treatment of breast cancer, researchers examined the combination of garlic-derived S-allyl-L-cysteine sulfoxide (alliin) and paclitaxel (PTX) effect on breast adenocarcinoma MCF-7 cells.

According to Western blotting assay, alliin and PTX worked synergistically to promote and potentiate apoptosis in MCF-7 cells by inhibiting the altered expression of importin-9 (IPO9) involved in transcription, cell signaling, and cell cycle regulation.

Furthermore, the inhibition of the IPO9 expression was also found to correlate with a significant reduction in the apoptotic cell population as well as with a decrease in F-actin content in the cancer cells, leading to alteration of cell DNA and in the cortical and nuclear/perinuclear areas of the cells.

Simultaneously, the inhibition of IPO9 by alliin and PTX was also accompanied by the increased post-translational expression of CFL1 associated with the reduction of cell proliferation and metastasis.

In other words, garlic-derived S-allyl-L-cysteine sulfoxide (alliin) and paclitaxel (PTX) induced breast cancer MCF-7 cells death by inhibiting the protein associated with cell differentiation and division through the reduction of cell proliferation and metastasis.

The combined compound exhibited anti-MCF-7 cells activity by targeting modulating the cytoskeleton present in the cytoplasm of cancer cells through some aforementioned mechanisms.

Taken altogether, alliin used alone or combined with primary medicine may be used for the prevention and treatment of breast cancer, pending to the confirmation of the larger sample size and multicenter human study.

Intake of alliin in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Herbal Turmeric's Curcumin, Protects the Blood Vessels Against the Onset of Stroke, in Vivo

By Kyle J. Norton

A stroke is a medical condition caused by poor blood flow to the brain, leading to the death of the neurons.

Besides cancer and heart diseases, stroke is the third leading cause of death in the US. According to the statistics, approximate 1/4 of all stroke victims die as a direct result of the stroke or its complications.

The causes of stroke are either associated with plaque build-up on the arteries, leading to reduce blood flow to the brain, depending on the location of an obstruction or the breaking of small blood vessels in the brain.

In other words, strokes can be classified into 2 types
* Ischemic stroke caused by a clot or other blockage within an artery which leads to the brain.
* Hemorrhagic stroke caused by the vessel in the brain rupturing that results in blood leaking into the brain.

Stroke is caused by a number of factors. Epidemiological studies suggested high blood pressure, medical conditions such as diabetes, heart disease, smoking, age, gender ethnicity and personal or family history of stroke are some of the prevalent risk factors.

Some researchers in the examination of the rise of stroke in North American suggested that stroke may be associated with the promotion of the Western diet over the past few decades.

The American Heart Association wrote, "Western" diets consisting of red and processed meats, refined grains, sweets, and desserts may be associated with a greater risk of stroke, according to a study published in this week's rapid access issue of Stroke: Journal of the American Heart Association".

Turmeric is a perennial plant in the genus Curcuma, belonging to the family Zingiberaceae, native to tropical South Asia.

The herb has been used in traditional medicine as anti-oxidant, hypoglycemic, colorant, antiseptic, wound healing agent, and for the treatment of flatulence, bloating, and appetite loss, ulcers, eczema, inflammations, etc.

On finding a natural compound for the treatment of stroke prevention scientists evaluated the protective effects of curcumin, the best-known member and is mainly extracted from turmeric preventive effect on stroke.

The study included stroke-prone spontaneously hypertensive rats (SHRsp) randomly divided into either a control group (n = 10, saline or curcumin (100 mg/kg/day)) or curcumin group (n = 10) administrated daily.

According to the tested assays, administration of curcumin significantly delayed the onset of stroke and increased the survival of SHRsp by decreasing the expression of ROS and improving endothelial-dependent relaxation of carotid arteries.

However, according to the RT-PCR and immunoblotting, curcumin showed no statistical significance in the presence of UCP2 inhibitor genipin by blocking the decrease of ROS and increase of NO production.

Where genipin is a chemical compound found in gardenia fruit extract.

In other words, in order for curcumin to exert its stroke preventive activity, curcumin cannot be consumed together with gardenia fruit.

Additionally, in the study to examine the curcumin effect during the stroke, researchers found that curcumin encapsulated in exosomes, nano-vesicles (<200 nm), shows therapeutic effects in brain diseases, such as stroke.

Taken altogether, turmeric processed a high amount of bioactive compound curcumin may be considered supplements for the prevention of stroke, pending to the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Ginger, A Natural Treatment of Post-Stroke Urinary Incontinence

By Kyle J. Norton

Scientists may have found a natural ingredient for the treatment of poststroke urinary incontinence with no side effects, according to study.

Post-stroke is a condition in which a patient continues to recovery from stroke by making changes in the physical, social and, emotional aspects of his/her life.

Urinary incontinence is a condition of leakage of urine involuntarily caused by the loss or weakened of the urinary sphincter.

In other words, a person urinates when he/she does not want to.

According to the statistic, urinary incontinence affects over 30 percent of females aged 30-60 compared to 1.5-5 percent of men.

The exact cause of the loss control of urinary sphincter is not identified. However, epidemiological studies suggested gender, smoking, obesity, and persistent stress are some of the potential risk factors associated with the condition.

Treatment of urinary incontinence is totally depending on the severity of the syndrome. Bladder control such as delaying the event, double voiding and toilet timetable and floor exercise have been found to be effective to many patients urinary incontinence.

Some patients may require the assistance of medication, including anticholinergics and Imipramine accompanied by bladder control and floor exercise.

Some researchers suggested the increase in age may be correlated to the onset of UTI as a large percentage of older adults were found to have UTI compared to the general population.

Dr. George A. Kuchel, the lead scientist wrote, "The prevalence of urinary incontinence (UI) increases with age. Moderate to severe UI affects 7% of women 20 to 39 yr of age, 17% 40 to 59 yr of age, 23% 60 to 79 yr of age, and 32% 80 yr of age".

And, "Nursing home residents have especially high rates of UI, ranging from 60 to 78% in women and 45 to 72% in men".

Ginger (Zingiber officinale) or ginger root, the second superfood used for thousands of years by mankind, is the genus Zingiber, belonging to the family Zingiberaceae, native to Tamil.

The root has been used in traditional and Chinese medicine for the treatment of dyspepsia, gastroparesis, constipation, edema, difficult urination, colic, etc.

With an aim to find a natural ingredient for the treatment of urinary incontinence in post-stroke patients researchers evaluated the efficacy and safety of ginger-salt-indirect moxibustion for urge urinary incontinence after stroke.

The study included cases of cerebral apoplexy, including cerebral infarction and cerebral hemorrhage, with urination disorders induced by neurogenic bladder at the restoration stage.

Patients were randomly divided into a treatment group treated with ginger-salt-partitioned moxibustion at Shenque (CV 8) and routine acupuncture, and a control group treated with routine acupuncture.

The treatment was given 5 times each week and the therapeutic effects were observed after treatment of 3 consecutive weeks.

39 cases in the treatment group and 36 cases in the control group completed all treatments.

The ginger-salt-partitioned moxibustion treatment group showed an improvement of urinary times of each day compared to control.

The mean times on awakening of the nursing personnel at night, and mean times of urgent urinary incontinence at day for the patients, were significantly lower in the treatment group.

Furthermore, cases-times of urinary incontinence of the patient at night, and increasing degree of urinary incontinence was better than that of the control group.

In other words, the ginger-salt-partitioned moxibustion treatment group was more superior in improving all parameters associated with urination disorders in post-stroke patients

Take all together, ginger used alone or combined with other alternative therapy may be considered a functional alternative for the treatment of urinary incontinence in post-stroke patients, pending to the confirmation of large sample size and multicenter human study.

Whole Food Tomato, The Potent Inhibitor Against Platelet Aggregation in Vitro and Vivo

By Kyle J. Norton

Platelet aggregation is a condition of platelets clumping together in the blood. The process also is also necessary for the cover of vascular injury.s

In other words, after sensing the bodily injury or damage, the immune system of the first line of defense stimulates the production of blood platelets to cover the site of injury with an aim to reduce the risk of infection.

Platelet disorders can lead to abnormal bleeding. Most common types of the genetic proposition of platelet disorders including
* Disorder of platelet adhesion, also known as Bernard-Soulier Syndrome is a condition in which the platelets fail to adhere together, leading to frequent bruises, nose bleeds and bleeding in the mouth and gums.

* Disorder of platelet aggregation also known as Glanzmann Thrombasthenia is a condition that the platelets not only fail to cover at the site of an injury but also prolong the bleeding.

* Disorder of platelet secretion is a condition which causes the slows down normal platelet adhesion, aggregation, and repair of the blood vessel.

* Disorder of platelets procoagulant activity, also know as Scott Syndrome is a condition in which the platelets fail to promote activation of the blood clotting proteins.

Conventionally, certain medication with blood thinning activity such as aspirin (ASA) and other drugs containing aspirin non-steroidal anti-inflammatory drugs and some antibiotics may have a strong implication of the formation of platelets coagulation.

Tomato is red, edible fruit, genus Solanum, belonging to family Solanaceae, native to South America. Because of its health benefits, the tomato is grown worldwide for the commercial purpose
and often in the greenhouse.

With an aim to find the natural ingredient for the treatment of platelet aggregation, researchers examined the anti-human platelet aggregation induced by ADP, collagen or arachidonic acid in aqueous extract of tomato.

The study included sugar-free tomato extract (TE) obtained by a heat-stable fraction with molecular mass <1,000 Da from tomatoes.was tested against the human plasma and rabbit lung.

100 g tomatoes produced 72.2 ± 4.7 mg of TE used in the experiment inhibited platelet aggregation in a dose-dependent manner.

The TE maximum effect in the inhibition of platelet aggregation  was found to be at an IC50 value of was 0.4 ± 0.02 mg/ml,

Furthermore, compared to the efficacy of TE, Fruitflow(®)-2 showed similar amounts of catechin also exerted a similar inhibitory potency against platelet aggregation activity.

In order to find more information about tomato anti-cardiovascular disease activity, researchers at the University of Newcastle examined the effect of an aqueous tomato fraction (tomato extract) against platelet aggregation in vitro.

According to the tested results from assays, platelet-rich plasma prepared from human blood incubated in the presence of tomato extract, collagen-induced, and adenosine diphosphate-induced platelet aggregation was significantly inhibited by the extract, compared to those with nontreatment plasma without affecting the basal cAMP levels in platelets.

Where cAMP plays a critical role in the regulation and signaling in platelets.

Interestingly, the extract inhibition of platelet aggregation was found to be dose-dependent and increased with longer incubation times.

However, the extract showed no effect anti-platelet aggregation induced by the presence of arachidonic acid.

In other words, the tomato extract inhibits platelet aggregation associated with other mechanisms involved in cathepsin G-induced human platelet activation such as the phospholipase C/protein kinase C pathway instead of cAMP levels.

Taken altogether, tomatoes may be considered supplements for the prevention and treatment of platelet aggregation, pending to the confirmation of the larger sample size and multicenter human study.

Intake of tomato extract in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.
