Ginger Inhibits the Production of Advanced Glycation End (AGEs) Products Against the Onset of Chronic Diseases, According to Studies

Advanced glycation end is a by produces formed by protein or fat combine with sugar caused by cooking meat at high temperature.

The products have been found to affect metabolic health.

Most common causes of AGEs are from the diet, particularly in people who follow the Western diet with high saturated and trans fat, a lot of red meats and processed foods.

Low levels of AGEs may not be a problem, however, overexpression of AGEs can induce oxidative stress, leading to inflammation and damage proteins, lipid and sometimes alternation of cells DNA.

In other words, AGEs initiates the production and free radicals and free radical a chain reaction that can not be stopped until all electrons in the outer ring paired or inhibited by antioxidants.

Accelerated formation and accumulation of advanced glycation end-products have been found to increased hyperglycemia or oxidative stress, leading to age-related and chronic diseases such as diabetes mellitus, chronic renal failure, neurodegenerative diseases, osteoarthritis and also non-diabetic atherosclerosis and chronic heart failure.

Some researchers suggested that modern life byproducts AGEs are the results of the promotion of the processed food diet.

Dr. Chetan Sharma, Amarjeet Kaur, the lead scientist in the assessment of AGEs, wrote, "As modern diets are largely heat processed, they are more prone to contain high levels of advanced glycation end products (AGEs)".

And, "The chemistry of the AGEs formation, absorption and bioavailability and their patho-biochemistry particularly in relation to different complications like diabetes and aging,... The emerging evidence about the adverse effects of AGEs makes it necessary to investigate the different therapies to inhibit AGEs".

The results strongly suggested the link of modern diet with highly processed food is a leading cause of chronic diseases.

Ginger (Zingiber officinale) or ginger root, the second superfood used for thousands of years by mankind, is the genus Zingiber, belonging to the family Zingiberaceae, native to Tamil.

The root has been used in traditional and Chinese medicine for the treatment of dyspepsia, gastroparesis, constipation, edema, difficult urination, colic, etc.

Researchers on finding a medical remedy in the inhibition of advanced glycation end (AGE) compared tea flavonoid (-)-epicatechin and [6]-Shogaol, a bioactive compound found in ginger against methylglyoxal (MGO) induced AGE formation.

Where Methylglyoxal (MGO) is a reactive dicarbonyl species considered a precursor of advanced glycation end products (AGEs).

Application of tea flavonoid (-)-epicatechin and [6]-Shogaol showed a strong effect in decreasing about 41% of MGO at a concentration of 10 μM within 24 h.

Both compounds also exerted a similar effect in vivo.

According to the urine samples collected from epicatechin and [6]-shogaol + epicatechin treated mice, both compounds also demonstrated a significant activity of adduction against mono-MGO.

Dr. Huang Q, the lead scientist in the joint study at the Nanjing Normal University, said, " this is the first study demonstrating the additive MGO trapping efficacy of [6]-shogaol and epicatechin and that [6]-shogaol and epicatechin retained their MGO trapping capacity in mice".

The results suggested that  [6]-shogaol + epicatechin prevent the production of AGEs by inhibiting the precursor (MGO).

Additionally, in order to reveal more information of ginger effect on AGEs, researchers investigated [6]-shogaol (6S) and [6]-gingerol (6G) on Methylglyoxal (MGO) which has been found in the contributes to the protein glycation and formation of advanced glycation end products (AGEs).

Similar to the above study, both [6]-shogaol (6S) and [6]-gingerol (6G) markedly trapped MGO in vitro. 

In the depth analysis, researchers found that 6S or 6G trapping MGO. was associated with the α-carbon of the carbonyl group in the side chain of the compounds in a time-dependent manner.

The findings showed the potential effects of 6S and 6G on the prevention of protein glycation through inhibiting the MGO.

Dr. Zhu Y, the lead scientist said, "...regular consumption of ginger root extract may attenuate the progression of MGO-associated diabetic complications in patients'.

Taken all together, ginger used alone or combined with green tea may be considered function remedies for the production of advanced glycation end products, pending to the confirmation of large and multicenter human study.

Tomato, the Anti Hyperlipidemia Functional Food for Type 2 Diabetes

Hyperlipidemia is a condition of abnormally high blood cholesterol in the bloodstream. The condition is also a member of the cluster of metabolic syndrome associated with the risk type II diabetes and cardiovascular disease.

Cholesterol is a waxy substance with a double-edged sword. On one hand, cholesterol is essential for the body to build strong cellular membranes, production of steroids hormones, and vitamin D and aided digestion, on another hand, it also causes a number of diseases if overexpressed in the blood.

Cholesterol is either produced by the liver or by the intake of fat.

There are 2 types of cholesterol in the body, the low-density lipoprotein (LDL, bad cholesterol) and high-density lipoprotein (good cholesterol).

Where high-density lipoprotein (HDL, good cholesterol) plays a critical role to return the cholesterol to the liver for future use. Low levels of high-density or high levels lipoproteins may result in blood cholesterol accumulated in the blood, leading to high blood cholesterol.

The optimized ratio of LDL/ HDL is any numbers that less than 4.

Dr. Joan Carles Escola`-Gil, the lead scientist in the evaluation of the Western diet risk of high blood cholesterol wrote, ". The HFHC diet caused a significant increase in plasma cholesterol, HDL cholesterol, and liver cholesterol and enhanced macrophage-derived [ 3 H]cholesterol flux to feces by 3- to 4-fold".

In other words, long-term intake of high-fat diet induced hyperlipidemia.

Tomato is red, edible fruit, genus Solanum, belongs to family Solanaceae, native to South America. Because of its health benefits, the tomato is grown worldwide for the commercial purpose
and often in the greenhouse.

With an aim to find a natural ingredient or whole food for the treatment of hyperlipidemia, researchers examined the effect of tomato and grape pomace to the cholesterol (0.3%) diet of male Wistar rats produced a dose-dependent effect.

During the experiment, researchers found that at 5% pomace of either and grape or combination showed no effects on reduced levels of hyperlipidemia.

However, 15% pomace of tomato and grape reduced serum cholesterol levels from 4.4 mmol/L to 2.5 mmol/L and 2.0 mmol/L respectively.

Interestingly, at the concentration of 15%, both tomato and grape pomace induced a redistribution of cholesterol in lipoproteins associated with an anti-atherogenic profile and reduced cholesterol concentration in very-low-density lipoprotein (VDL) (24% [tomato], 50% [grape]) and low-density lipoprotein (LDL) (3-fold, 3.6-fold).

Furthermore, both types of pomace enhanced the function of HDL in cholesterol transport by reducing the VLDL and LDL contribution.

Moreover, tomato and grape pomace reduced plasma levels of conjugated dienes involved in cholesterol oxidation in high levels by 30-50%

Additionally, injection also improved the antioxidant enzymes in the liver against lipid peroxidation.

In rats fed with hypercholesterolemic feed and treated with lycopene orally by gavage at the dose of 5, 25, 45, 65, 85, and 105, 125 mg/kg/bw-1/d-1, researchers found that injection of all doses of lycopene exerted a statistical significance in reduced the levels of cholesterol observed by inverse U shape at the 4 weeks of the experiment.

Furthermore, serum lycopene concentration was negatively correlated with the levels of serum TC, LDL-C, particularly in the cerebral LDL-C.

Serum lycopene concentration was also found to have a positive correlation with the expression of Claudin-5 associated with brain microvascular endothelial cell proliferation, migration. and the number of neurons in hippocampal CA1 and CA3.

In other words, serum lycopene showed a strong effect on the protection of neurons against hyperlipidemia in the brain that facilitates neurons damage.

Dr. Wang W, the lead scientist said, "These results suggested an inverse U-shape relation between dose and serum concentration of lycopene, and intermediate doses were most effective to protect cerebral vessels and neurons from being damaged by hyperlipidemia".

Taken altogether, tomatoes processed high amounts of lycopene may be considered supplements for the prevention and treatment of hyperlipidemia, pending to the confirmation of the larger sample size and multicenter human study.

Intake of lycopene in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Cancer is a group of diseases caused by cell growth disorderly and uncontrollably in the tissue of the affected organs.

Cancers are caused by the alternation of cell DNA. What influences the change of cells DNA?

Epidemiological studies suggested that certain risk factors such as age, family and personal history, ethnicity, exposure to toxic chemicals, unhealthy lifestyle,... the list goes on and on, but no one can explain why people with the came healthy conditions in the same family, some are susceptible to cancer while others do not.

Dr. Alexandre Corthay in the examination of the risk of cancer in patients with weakened immune system explained "Here, I present eight types of evidence in support of the cancer immunosurveillance hypothesis. 
* First, primary immunodeficiency in mice and humans is associated with increased cancer risk. 

*Second, organ transplant recipients, who are treated with immunosuppressive drugs, are more prone to cancer development. 

*Third, acquired immunodeficiency due to infection by human immunodeficiency virus (HIV-1) leads to the elevated risk of cancer. 

*Fourth, the quantity and quality of the immune cell infiltrate found in human primary tumors represent an independent prognostic factor for patient survival. 

*Fifth, cancer cells harbor mutations in protein-coding genes that are specifically recognized by the adaptive immune system. 

*Sixth, cancer cells selectively accumulate mutations to evade immune destruction (“immunoediting”). 

*Seventh, lymphocytes bearing the NKG2D receptor are able to recognize and eliminate stressed premalignant cells. 

*Eighth, a promising strategy to treat cancer consists in potentiating the naturally occurring immune response of the patient, through blockade of the immune checkpoint molecules CTLA-4, PD-1, or PD-L1".

Herbal Bilberry, The Potential Anti Intestinal Cancer Natural Remedy with No Side Effects, According to Scientists

Intestinal cancer is the medical condition characterized by irregular cell growth in the tissue of the intestines.

Most cases of intestinal cancer start in the cells on the surface of the inner lining of the internal tissue. At the advanced stage, after penetrating deeply layers of the intestines, the cancer cells can travel a distance away from the intestine to infect other healthy tissue and organs, leading to secondary metastasis.

Certain factors such as being overweight or obese, physical inactivity, certain types of diet. smoking excessive alcohol drinking, family and personal history and the increase in age, are associated with an increased risk of intestinal cancer.

Some researchers suggested that the increased incidence of intestinal cancer in the Western world may be associated with the promotion of high-fat Western diet over the past many decades.

Dr. Manon D. Schulz, the lead scientists in the investigation of the effects of high-fat diet-mediated dysbiosis promotes intestinal carcinogenesis suggested, "Several aspects common to a Western lifestyle, including obesity and decreased physical activity, are known risks for gastrointestinal cancers. There is substantial evidence suggesting that diet profoundly affects the composition of the intestinal microbiota. Moreover, there is now unequivocal evidence linking dysbiosis to cancer development".

And, "Undoubtedly various factors contribute to the etiology of intestinal cancer, but there are compelling arguments to include high-fat diet (HFD) and composition of gut microbiota as key risk factors".

The results strongly suggested that the risk of intestinal cancer can be decreased by changing the diet pattern from the Western diet to a traditional diet with higher fruits and vegetables with a lower intake of red meat and saturated and trans fat and processed foods.

Bilberry is a species of low-growing shrubs in the genus Vaccinium, belonging to the family Ericaceae, native to Northern Europe.

The herbal plant has been used as foods and herbs in traditional medicine for the treatment of acute and chronic diarrhea, gastritis, gastric ulcer, and ulcer, enterocolitis, ulcerative colitis, anemia, cystitis, kidney disease, and psoriasis, diabetes, etc.

Researchers on finding a natural compound for the treatment intestinal carcinogenesis examined the
intestinal carcinogenesis--relationship with tissue anthocyanin levels in an animal model.

Selected Apc(Min) mice ingested C3G or Mirtoselect at 0.03, 0.1 or 0.3% in the diet for 12 weeks, and intestinal adenomas were examined.

Where Mirtoselect, is an anthocyanin mixture from bilberry, and isolated cyanidin-3-glucoside (C3G) is the most abundant anthocyanin in the diet.

C3G and Mirtoselect at the highest doses decreased the adenoma numbers by 45% (p < 0.001) or 30% (p < 0.05), respectively, compared to controls.

According to the intestinal mucosa and urinary assays, anthocyanins were found at the analytical detection limit in the plasma and at quantifiable levels.

More precisely, the total anthocyanin levels in mice on C3G or Mirtoselect were 43 ng and 8.1 microg/g tissue, respectively, in the intestinal mucosa, and 7.2 and 12.3 microg/ml in the urine. 

These results suggested that C3G and Mirtoselect in the Apc(Min) mouse processed a chemotherapeutic effect for the treatment of intestinal carcinogenesis.

Moreover, in order to reveal more information about berries anti intestinal tumorigenesis, researchers examined the different phenolic profiles on intestinal tumorigenesis in multiple intestinal neoplasia/+ mice.

Mice selected to the study were fed a high-fat AIN93-G diet (Con) or AIN93-G diets containing 10% (w:w) freeze-dried bilberry, lingonberry (LB), or cloudberry (CB) for 10 wk.

All 3 berries significantly inhibited the formation of intestinal adenomas as indicated by a 15-30% reduction in tumor numbers.

According to the Western blotting and immunohistochemistry, all 3 berries inhibited the levels of decreased levels of proteins associated with cell cycle division.

Additionally, all 3 berries exerted early protection of the cells in the intestine against the alternation of cell DNA to facilitate the onset of the disease.

The findings strongly suggested the use of berries as chemopreventive components for the treatment of intestinal carcinogenesis. 

Taken altogether, bilberry may be considered a functional remedy for the prevention and treatment of
intestinal cancer,  pending to the confirmation of large sample size and multicenter study.

Infertility is a reproductive disease characterized by the failure to produce a pregnancy after 12 regular unprotected sexual intercourse, including pregnancy that can not be carried to term.

Ginger (Zingiber officinale) or ginger root, the second superfood used for thousands of years by mankind, is the genus Zingiber, belonging to the family Zingiberaceae, native to Tamil.

The root has been used in traditional and Chinese medicine fro the treatment of dyspepsia, gastroparesis, constipation, edema, difficult urination, colic, etc.

In the course of the continued efforts to discover a natural remedy for the treatment of infertility, researchers investigated the ginger activity on male reproductive functions.

The randomized double-blind, placebo-controlled trial included 100 infertility treatment candidates admitted to Royan Institute for Reproductive Biomedicine, Tehran, Iran.

Patients were randomly assigned to receive one of two treatments: ginger and placebo. Patients were given a 3-month oral treatment (members received capsules containing 250 mg of ginger powder twice a day in ginger and a placebo in another group).

At the end of the experiment, according to the sperm DNA fragmentation (SDF) analysis, the ginger treatment group showed a significant lower of the average positive percentage of SDF compared to the placebo group.

However, there were no significant differences in other semen parameters between the two groups.

The finding clearly suggested that ginger improves the SDF profile in the infertile men.

In order to reveal more information about ginger anti-male infertility activity, researchers investigated the preventive effects of ginger and turmeric rhizomes on some biomarkers of male reproductive function in L-NAME-induced hypertensive rats.


Male Wistar rats selected to the study were divided into seven groups (n = 10): normotensive control rats; induced (L-NAME hypertensive) rats; hypertensive rats treated with atenolol (10 mg/kg/day); normotensive and hypertensive rats treated with 4% supplementation of turmeric or ginger, respectively.

After 14 days of pre-treatment, the animals were induced with hypertension by oral administration of L-NAME (40 mg/kg/day).

Application of L-NAME showed a significant decrease in serum total testosterone and epididymal sperm progressive motility without affecting sperm viability in hypertensive rats.

Hypertensive rats exerted an increase of oxidative stress by decreasing the level of antioxidant enzymes produced by the rats.

Furthermore, hypertensive rats also displayed a decreased testicular and epididymal NO level with concomitant elevation in arginase activity that affects the sperm counts.

Dietary supplementation with turmeric or ginger efficiently normalized the alterations in biomarkers of reproductive function in hypertensive rats.

Dr. Akinyemi AJ, and colleagues wrote, "The inhibition of arginase activity and increase in NO and testosterone levels by both rhizomes could suggest a possible mechanism of action for the prevention of male infertility in hypertension".

Taken altogether, ginger used alone or combined with turmeric may be considered functional remedies for the prevention and treatment of male infertility, pending to the confirmation of large sample size and multicenter human study.

Bioactive Ingredient 3,3'-Diindolylmethane Gobbles Up Colorectal Cancer Cells in Vitro

Colorectal cancer is a medical and chronic condition caused by irregular cell growth in the colon or rectum.

Colorectal cancer is one of the slowest growth of cancer. Therefore, Most cases of colorectal cancer are discovered in the later stage, causing a high rate of mortality.

According to the study, at the advanced stage, the median survival of patients is 24 months (range 16–42). The one-year and 2-year survival rate is 65% and 25%, respectively.
Most cases of colorectal cancer start in the cells on the surface of the inner lining of the colon or rectum. At the early stage, due to the small size of the tumor, most patients are asymptomatic.

However, at the advanced stage, the oversize tumor may suppress the nearby blood vessels and nerve cells, leading to persistently severe bleeding and pain.

Patients at this stage may also experience symptoms of a change in bowel habits, including diarrhea or constipation or consistency of the stool, that lasts longer than four weeks.

3,3'-Diindolylmethane or DIM are phytochemicals derived from the digestion of indole-3-carbinol, belonging to the group of Indoles, found abundantly in broccoli, Brussels sprouts, cabbage, and kale, etc.

Researchers on finding a potential and natural compound for the treatment of colorectal cancer examined diindolylmethane (DIM) effect on cyclin D1, which was aberrantly overexpressed in various cancer cells and tumors.

Where Cyclin D1 is a protein associated with the progression of cancer through the G1 phase of the cell cycle in cellular division.

Injection of DIM interfered the Cyclin D1 function in the promotion of colorectal cancer proliferation, without affecting the cyclin D1 mRNA expression.

The results suggested that DIM modulated cyclin D1 expression by controlling the levels of protein synthesized from its mRNA.

In other words, DIM.inhibited cyclin D1 by triggering ER stress in human colorectal cancer cells through decreasing protein synthesis and growth arrest in the G1 phase of the cell cycle.

Additionally, in order to reveal more information about DIM anti-colorectal cancer property, researchers investigated the effect of DIM on the migratory and invasive activities of the human colorectal cancer cell lines DLD-1 and HCT116.

Injection of DIM significantly inhibited the migratory ability of the DLD-1 and HCT116 cells at 24 and 48 hours.

DIM also significantly inhibited the invasion rate of the DLD-1 and HCT116 cells in a dose-dependent manner.

Following DIM treatment, researchers found that DIM anti-colorectal cancer property was associated with attenuating the mRNA expression levels of urokinase receptor and matrix metalloprotease 9 (MMP9) involved cancer cell adhesion and enhancing the expression of tumor-suppressive mRNA.

Furthermore, DIM significantly reduced the mRNA and protein levels of FOXM1 in the promotion of cellular division or proliferation in DLD-1 and HCT116 cells. 

Based on these results, Dr.Jin H, the lead scientist in the summary of the report, wrote, "DIM can influence the cell migratory and invasive properties of human colorectal cancer cells and may decrease the invasive capacity of colorectal cancer through downregulation of uPA and MMP9 mediated by suppression of the transcription factor FOXM1".

Taken altogether, 3,3'-Diindolylmethane derived from the digestion of indole-3-carbinol may be considered a supplement for the prevention and treatment of colorectal cancer, pending to the confirmation of the larger sample size and multicenter human study.

Intake of 3,3'-diindolylmethane in form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Phytochemical Sinigrin Improves the Metabolic Parameters in Patients with Types 2 Diabetes, Cardiovascular Diseases

Metabolic parameters are a measurable factor used for the prediction of type 2 diabetes, heart diseases, and stroke.

Metabolic factors including hyperglycemia, hyperlipidemia, and hyperkalemia are important considerations used for the treatment of patients with type 2 diabetes and nephropathy.

Where hyperglycemia, hyperlipidemia, and hyperkalemia are conditions of abnormally high levels of sugar, cholesterol and potassium level in the bloodstream, respectively.

In other words, the levels of lipid and carbohydrate homeostasis, esterified and total cholesterol, free fatty acids and triacylglycerols are considered measurable factors for the onset and treatment of types 2 diabetes and cardiovascular diseases.

For example, in a retrospective study used a national electronic database to examine the metabolic parameter of Malay patients with type 2 diabetes, researchers in the comparison of some of metabolic parameters  traced before, during and after Ramadan, found that systolic blood pressure, low‐density lipoprotein cholesterol and triglycerides showed insignificant change in all participants, except blood glucose which was most affected during Ramadan.
By knowing the parameters that affect the patients has been found to enhance the management and treatment in patients with types 2 diabetes.

Researchers in seeking a potential natural compound for the improvement of metabolic parameters examined the sinigrin (SIN) and allyl isothiocyanate (AITC) preventive and medicinal effect.

The study included the injection of glucosinolate sinigrin 50 μmol/kg b. w./day and its degradation product allyl isothiocyanate 25 μmol/kg b. w./day and 50 μmol/kg b. w./day in an animal rat model in vivo after their single (4 h) and 2 weeks oral administration.

AITC alone after 4 h of its ingestion caused liver triacylglycerols increment at both doses and glycemia only at the higher dose.

Interestingly, multiple SIN treatment showed its putative bioconversion into AITC.

SIN and AITC multiple administration strongly disturbed lipid and carbohydrate homeostasis, increasing esterified and total cholesterol, free fatty acids and lowering triacylglycerols in the blood serum.

Additionally, AITC at both doses elevated insulinemia and liver glycogen enhancement. 

The finding strongly suggested that SIN and AITC improve metabolic parameters through multi-directions including liver, pancreas, and intestine.

Dr. Okulicz M, the lead scientist said, "This effect is potentiated with a time of action of the examined compounds, whose influence is rather adverse for the majority of metabolic pathways (liver steatosis at the short duration and insulinemia, cholesterolemia at long time treatment)".

Taken altogether, sinigrin a bioactive ingredient in cruciferous vegetables may be used to improve metabolic factors against the onset of metabolic syndrome, pending to the confirmation of large sample size and multicenter human study.

However, intake of bioactive in the form of supplement should be taken with extreme care, as liver toxicity may be induced if overdosed.
