Cucumber (Cucumis sativus) Cucurbitacin B (CuB) The Potential and Natural Anti Chronic Myeloid Leukaemia Functional Food

Chronic myeloid leukemia (CML) also known as chronic myelogenous leukemia is a type of leukemia associated with certain blood-forming cells of the bone marrow.

Chronic myeloid leukemia (CML) is a slow growing cancer compared to the acute forms of leukemia.

In the US, most cases of chronic myelogenous leukemia are found in adults age of 65 and older.

According to the American Cancer Society, approximately, 8,990 new cases will be diagnosed with CML (5,250 in men and 3,740 in women) in 2019. CML also will cause the death of 1,140 patients (660 men and 480 women) in the same years.

The lifetime risk of CML is 1 in 526 in the United States.

CML besides sharing the common of symptoms of other types of cancer, the condition also induce symptoms of easy bleeding, pain or fullness below the ribs on the left side, pale skin and sweating excessively during sleep (night sweats).

Today, due to new drug discovery and improvement of technology, the 5-year survival rate for CML is 68% compared to only 31% in the early 1990s.

Out of many risk factors such as radiotherapy, gender, and aging, some researchers suggested that obesity may be one of the main factors that elevated the risk of the disease.

Dr. Strom SS, the lead researchers in the cancer team at the University of Texas M. D. Anderson Cancer Center wrote, " Obesity, measured as body mass index, has been identified as a possible risk factor for several solid tumors as well as some adult hematopoietic malignancies".

And, "Obesity at all ages was found to be an independent risk factor, with a significant dose-response effect. Among participants > or =45 years, cases gained significantly more weight each year between ages 25 and 40 compared with controls (0.78 versus 0.44 kg/y, P < 0.001) with the association strongest among those who gained >1 kg/y between 25 and 40 years of age (OR, 3.63; 95% CI, 1.46-9.04). Our results suggest that obesity and adulthood weight gain play important roles in CML risk".

The findings strongly suggested that obese are more likely to develop CML, compared to those with normal weight.

Cucumber plant is a species of Cucumis Sativus, belonging to the family Cucurbitaceae and native to Western Asia. It is a creeping vine with roots in the ground and grows up with the support of frames.

On finding a natural and potential compound for the treatment of chronic myeloid leukemia, researchers examined the Cucurbitacin B (CuB) antiproliferative effects on various leukemia cell lines.

Administration of CuB showed strong inhibition against various leukemia, lymphoma cell lines, primary mononuclear bone marrow cells derived from patients with acute myeloid leukemia or myelodysplastic syndrome.

CuB induced myeloid leukemic cells apoptosis through S-phase cell cycle arrest, enlarged cell size, multinucleation, and enhanced expression of a monocytic- and granulocytic-specific CD11b involved in numerous adhesion-related associations between cells such as monocytes, macrophages, natural killer (NK) cells, and granulocytes.

Furthermore, CuB prominently altered the cytoskeletal network of leukemic cells, leading to cell death.

In order to obtain additional information about cucurbitacin B (CuB) anti myeloid leukemia activity, researchers explored the antitumor effect and action mechanism of cucurbitacin B (CuB) on human T-cell leukemia Jurkat cells.

According to the tested assays, CuB inhibited the proliferation of Jurkat cells in a dose-dependent manner and induced G 2 /M phase arrest.

CuB induced around 10% of cells undergoing apoptosis, after treatment for 24 h.

Moreover, CuB induced cancer cell autophagy observed by the accumulation of LC3-II, an indication of cleaning out damaged cells and regenerating new ones.

The results of autophagy were found to be activated by the protein associated with a significantly increased rate of recurrence and deaths.

Taken altogether, cucumber processed a higher number of CuB may be used for the treatment of leukemia, pending to the confirmation of the larger sample size and multicenter human study.

Intake of CuB in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Phytochemical Alliin Balances the Gut Microbiota and Proinflammatory Cytokines for Optimal Immunity

Gut microbiota refers to the population of microbes living in the gut. The community contains tens of trillions of microorganisms with over 1000 different species of bacterias.

The gut microbiota plays a vital role in human health, including the key processes of nutrition-enhancing the digestive functioning, lipid, and energy metabolism by inducing the production of short chain fatty acids (SCFAs), phenolics, and other molecules,  and pathogen resistance by promoting the immune function.

In a healthy individual, the levels of gut microbiota are balanced that allow the body to function in an optimal way.

Furthermore, gut microbiota also is found to stimulate the production of some vitamins (B and K),  digest certain foods that the stomach and small intestine have not been able to digest, particularly in the combat against the aggression of other bad microorganisms.

However, alterations in gut microbiota composition have been found in patients with a low-grade inflammatory state, leading to an increased risk of several diseases, including type 2 diabetes mellitus and cardiovascular disease.

Although the digestive tract can adapt to change, the loss of balance in gut microbiota may induce dysbiosis., leading to health problems of functional bowel disorders, inflammatory bowel disease, allergies, obesity, and diabetes.

The are many risk factors associated with gut microbiota imbalance. Some researchers suggested that the major causes of the condition may be a result of the promotion of a high-fat diet over the past few decades.

Dr. Duo Li, at the Institute of Nutrition and Health at Qingdao University in China, and colleagues wrote, "“Western-type diet has a strong effect on the genetic composition and metabolic activity of gut microbiota,” they wrote. “Evidence has shown that in humans gut microbiota diversity and richness are reduced when comparing such high-fat diets with more traditional diets with relatively higher proportions of carbohydrate. Such diet-induced ‘dysbiosis’ in gut-associated microbial communities has been postulated as a major trigger of metabolic impairments associated with obesity".

Alliin (S-allyl-L-cysteine-S-oxour intestinal chemical compound sulfoxide,. a derivative of the amino acid cysteine, belonging to the class of sulfur compounds, found abundantly in fresh garlic and onion.

With an aim to find a natural ingredient with immunomodulatory activity, researchers examined the effect of the garlic derivative alliin, on the mitogen-induced peripheral blood mononuclear cell (PBMC) proliferation and cytokine production in vitro.

Before treated with garlic derivative alliin, pokeweed mitogen (PWM) injection induced cell proliferation, production of proinflammatory cytokines, including IL-1beta and TNF-alpha.

Furthermore, pokeweed mitogen (PWM) treated group also showed the reduced levels of both percentage of phagocyting cells and number of latex particles associated with the immune functions of phagocytosis.

Administration of garlic derivative alliin not decreased only the production of proinflammatory cytokines induced by pokeweed mitogen (PWM) but also improved the levels of antioxidant enzymes.

In other words, alliin exerted an immunomodulatory effect on the peripheral blood cells in vitro.

On finding a natural compound for the improvement of the ratio of gut microbiota, researchers examined whether different plant-derived components with similar anti-oxidative and anti-inflammatory function such as quercetin and alliin in traditional Asian diet on the status of the gut microbiota and colonic epithelial immunity.

Traditional Asian diet significantly increased reshaped the gut microbiota composition by significantly increased the relative abundance of fecal Firmicutes and Cyanobacteria but decreased that of Bacteroidetes (P < 0.05) and increased the relative abundance of Candidatus Arthromitus, Lactococcus, Geobacillus, and Ruminococcus at the genus level that benefits the host's health.

In other words, the traditional Asian diet reduced the expression of bad bacteria and increased the good bacteria in the gut.

 The consumption of quercetin and alliin also altered 13 genes expression involved in the KEGG pathways immunomodulatory effect in improving the gut immunity through their anti-inflammatory and anti-free radical activities.

In other words, the alternate consumption of quercetin and alliin can contribute beneficial metabolic effects by optimizing gut microbiota by improving the immunologic function of colonic epithelial cells associated with the innate immune defense.

Taken altogether, alliin may be considered supplements for improvement of gut microbiota inducing immunologic function of colonic epithelial cells associated with the innate immune defense, pending to the confirmation of the larger sample size and multicenter human study.

Intake of alliin in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Herbal Turmeric Protects the Bone Cells Integrity Against Osteoporosis

Osteoporosis is a medical condition characterized by the loss of bone density.

In other words, the bone disease is a result of overexpression of bone loss due to bone resorption compared to bone made by the body, leading to risk bone fracture and breaking.

In a severe case, bones of patients with osteoporosis may become so weak, they can break from a fall or even from sneezing or minor bumps.

Microscopy, healthy bone processed a high density (fewer spaces and small holes in the honeycomb) compared to low density ( large spaces and holes in a honeycomb) in the patients with osteoporosis.

In the US, due to the widespread osteoporosis in the elderly, the disease has become a concern of the scientist community. Believe it or not, according to the statistics, osteoporosis is responsible for two million broken bones and $19 billion in related costs every year. 

As the condition continues to worsen, by 2025, osteoporosis will be responsible for approximately three million fractures and $25.3 billion in costs annually.

Additionally, 1 in 4 women and more than 1 in 8 men over the age of 50 years, with 1 in 4 men and women are having evidence of a vertebral fracture.

Out of many risk factors associated with the onset of osteoporosis, some researchers suggested that the disease may be correlated to the promotion of Western diet over the past few decades. 

Dr. Seon-Joo Park, the lead scientist wrote, "(In the comparison of ) "Three major dietary patterns,... using factor analysis based on baseline intake data: traditional (high intake of rice, kimchi, and vegetables), dairy (high intake of milk, dairy products, and green tea), and western (high intake of sugar, fat, and bread).".

And, "..., western dietary pattern (p-trend = 0.043 in radius) demonstrated a higher risk of osteoporosis incidence than those in the lowest quintile".

Turmeric is a perennial plant in the genus Curcuma, belonging to the family Zingiberaceae, native to tropical South Asia.

The herb has been used in traditional medicine as anti-oxidant, hypoglycemic, colorant, antiseptic, wound healing agent, and to treat flatulence, bloating, and appetite loss, ulcers, eczema, inflammations, etc.

On finding a natural compound for the treatment of osteoporosis, researchers evaluated the efficacy, in term of changes in bone density, and safety of an oral formulation based on turmeric phytosome (Meriva®), in subjects suffering from low bone density.

The study included a total of 57 otherwise healthy subjects with low bone density who freely decided to follow either standard management (SM) to control low bone density (control group=28) or SM associated with a curcumin-based oral supplementation (supplement group=29).

Meriva®-supplemented group remarkably improved in the bone density of the heel measured by the Sahara densitometer accompanied by a significant decrease of ultrasounds transmission values at week 12 (-18.4%) and at week 24 (-21.0%), compared with baseline.

Furthermore, Meriva®-supplemented group also improved the bone densities of small finger and upper jaw with no adverse effects.

Based on the results researchers said, "This preliminary study suggests that a curcumin-based supplementation in combination with an appropriate lifestyle could be beneficial in the prevention and management of osteopenia".

In order to know more information about turmeric anti-osteoporosis activity, researchers examined the effects of curcumin on oxidative stress-induced osteoblast apoptosis. 

The study included an osteoblastic cell line (Saos-2) exposed to various concentrations of H2O2 with or without curcumin treatment.

Injection of curcumin protected the viability of Saos-2 cells against the exposure to H2O2 and attenuated H2O2-induced apoptosis.

Furthermore, curcumin also improved the cellular function by preserving the mitochondrial redox potential, decreasing the mitochondrial oxidative status, through its antioxidants activity.

Moreover, curcumin treatment markedly increased levels of phosphorylated protein kinase B (Akt) and phosphorylated glycogen synthase kinase-3β (GSK3β) that have been found to induced bone loss in low levels.

Whole Food Tomato Protects the Lung Against Pulmonary Oxidative Stress

Pulmonary oxidative stress is a condition of oxidative stress in the lung.

The lung is a breathing organ containing a pair of spongy, air-filled organs located on either side of the chest (thorax) by taking oxygen to the blood vessels then passing them to all parts of our body. 

Furthermore, the lung also exhales the waste product called carbon dioxide, through breathing.

Oxidative stress is a result of an imbalance of the levels of free radical compared to the antioxidant enzymes in the body.

Oxidative stress is either expressed by overexpression of free radicals or reduced levels of antioxidants or both.

Free radical can be everywhere in the environment such as the air we breathe, the foods we eat. they can also be produced by cells metabolism in the body.

Believe it or not, free radicals in moderate amount are necessary for our body to function, including NO for muscle relaxation.

Epidemiologically, oxidative stress has been found to induce lipid and protein oxidation and cell DNA damage.

According to the free radical theory, aging posits caused by the accumulation of damage inflicted by reactive oxygen species (ROS) associated with varies chronic diseases such as atherosclerosis, cancer, diabetics, rheumatoid arthritis, post-ischemic perfusion injury, myocardial infarction, cardiovascular diseases, chronic inflammation, stroke.

In the lung, oxidative stress also was shown to elevate the risk of the pathogenesis of chronic obstructive pulmonary disease.

Tomato is red, edible fruit, genus Solanum, belonging to family Solanaceae, native to South America. Because of its health benefits, the tomato is grown worldwide for the commercial purpose
and often in the greenhouse.

With an aim to find a natural and potential whole food for the treatment of diseases associated with pulmonary oxidative stress, researchers investigated the dietary supplementation with a known source of antioxidants (tomato juice, TJ) in the protection of the lung from injury caused by breathing hyperoxic gas.

The study included the examination of the results from the selected neonatal mice (C57BL6/J) breathed either 65% O2 (hyperoxia) or room air from birth until postnatal day 7 (P7d).

At P56d, TJ protects the lung cell integrity and viability by increasing the levels of total antioxidant capacity (TAC), decreasing oxidative stress and reversing the hyperoxia-induced increase in bronchiolar smooth muscle.

Furthermore, injection of TJ alone also decreased the protein involved in the production of pro-inflammatory cytokines.

In hyperoxia, TJ increased TNF-α expression and did not alter the hyperoxia-induced increase in leukocyte number.

In other words, TJ protects the lung against the oxidative stress by inhibiting and exhibiting certain protein associated with the proinflammation without increasing the production of immune white blood cells via its pulmonary antioxidants.


Based on these results, Dr. Bouch S, the lead scientist said, "TJ supplementation during and after neonatal exposure to hyperoxia protects the lung from some but not all aspects of hyperoxia-induced injury".

In other to reveal additional information about tomato anti pulmonary oxidative stress effect, researchers examined bioactive compound lycopene extracted from tomatoes in pulmonary fibrosis (PF) in an animal model.

The study included 60 Sprague-Dawley rats induced pulmonary fibrosis  (PF) by BLM randomly divided into 3 groups of 20 rats each for a normal control group (group C), BLM-treated group (group M), and lycopene + BLM-treated group (group L).

Additionally, the rats in groups M and L were subjected to intratracheal instillation of BLM to induce PF; group C served as a sham control (intratracheal instillation of normal saline). Lycopene diluted with olive oil was administered at a dose of 5 mg/kg body weight once a day in group L after BLM instillation, and groups C and M were treated with the same amounts of olive oil.

The lung coefficients in group L treated with lycopene were reduced (day 14, P < .01) as well as the extents of alveolitis (day 7 and 14, P < .05) and PF (day 14 and 28, P < .05) compared with group M.

Lycopene treated group also showed significantly reduced levels of proteins associated with the immune systematic response against the acute phase of infection.

Furthermore, lycopene levels of oxidative stress markers in the lung observed by the decreased levels of malonyldialdehyde plasma.

In other words, lycopene protected the lung of the tested rat by reducing oxidative stress via the stimulation of the production of antioxidant enzymes.

Taken altogether, tomatoes processed a high amount of bioactive compound lycopene may be considered supplements for the prevention and treatment of diseases associated with pulmonary oxidative stress, pending to the confirmation of the larger sample size and multicenter human study.

Ginger (Zingiber officinale) or ginger root, the second superfood used for thousands of years by mankind, is the genus Zingiber, belonging to the family Zingiberaceae, native to Tamil.

The root has been used in traditional and Chinese medicine for the treatment of dyspepsia, gastroparesis, constipation, edema, difficult urination, colic, etc.

Researchers on finding a bioactive compound for the treatment of infection associated with virulence examined the effects of 6-Gingerol and 6-Shogaol against Candida albicans.

6-gingerol, 8-gingerol, and 6-shogaol effectively inhibited biofilm formation.

6-shogaol at 10 μg/ml significantly reduced C. albicans biofilm formation but had no effect on planktonic cell growth.

Also, 6-gingerol and 6-shogaol inhibited hyphal growth in embedded colonies and free-living planktonic cells and prevented cell aggregation.

Furthermore, 6-gingerol and 6-shogaol reduced C. albicans virulence in a nematode infection model without causing toxicity at the tested concentrations.

These results suggested that the antibiofilm and antivirulence activities of the ginger components, 6-gingerol and 6-shogaol, were attributed to the inhibition of the proteins associated with a multidrug
resistance gene and a cytolytic peptide toxin from Candida albicans.

In order to reveal more information about the ginger effects antibiofilm activity, researchers examined the antimicrobial, antiadhesive and antibiofilm potential of fresh and cooked ginger water extracts (FGE and CGE, respectively).

Compared to CGE, FGE displayed high potential anti-adhesive and antibiofilm in vitro. 

 20 mg DTSS /mL of FGE led to reducing the adhesive of bacterial cells to HEp-2 cells by about 50%. 

The results showed that ginger water extract processed the desired anti-adhesive and antibiofilm potential against the food-related pathogens.

Taken altogether, ginger processed a high amount of bioactive compound may be considered a functional remedy for the inhibition of biofilm activities, without inducing any side effects.

Herbal Artichoke Inhibits the Parameters Associated with the Onset of Non-Alcoholic Fatty Liver Disease (NAFLD) in Vivo and Human Study

Non-alcoholic liver disease is a medical condition caused by excessive fat accumulated in the liver of people who drink little or no alcohol.

Believe or not, non-alcoholic fatty liver disease (NAFLD) now becomes a universal disorder and the most common liver disease in the Western world.

According to the statistics in 2015, approximately, 20 to 30% of the general population in the Western world suffer from NAFLD. The prevalent rate is substantially higher in the population with type 2 diabetes mellitus (70%) and morbid obesity (90%). 

Some researchers in the assessment of the relationship of widespread obesity in risk of NAFLD suggested that non-alcoholic fatty liver disease may be caused by the promotion of the Western diet over the past few decades.

Dr. Kristen Stephenson, in the assessment of the impact of dietary fat on the development of non-alcoholic fatty liver disease in animal model wrote, "WD mouse models consisting of high fat, cholesterol, and a combination of high-fructose corn syrup, sucrose, fructose, or glucose not only lead to metabolic syndrome but also induce NAFLD with fibrosis".


Artichoke is a perennial thistle of Cynara cardunculus species of the Cynara genus, belonging to the family Carduoideae native to Southern Europe around the Mediterranean.

The herbal plant has been used in traditional medicine as liver protective and detoxified agent, and to treat digestive disorders, abdominal pain gas and bloating, etc.

Researcher on finding a natural therapeutic for the treatment of Non-alcoholic fatty liver disease (NAFLD), the most common cause of chronic liver disease worldwide investigated the artichoke leaf extract (ALE) therapeutic utility in patients with established NAFLD. 

The randomized double-blind placebo-controlled parallel-group trial included 100 subjects with ultrasound-diagnosed NAFLD randomized to either ALE 600 mg daily or placebo for a 2-month period.

Compared to the control, the ALE treatment group showed increased hepatic vein flow, reduced portal vein diameter and liver size, according to the Doppler sonography. In other words, ALE treatment improved liver function.


Inflammatory and oxidative stress such as serum ALT, AST levels and total bilirubin were also decreased upon the injection of ALE observed by the improved AST/ALT ratio.

Furthermore, ALE supplementation also reduced total cholesterol, low-density lipoprotein cholesterol, high-density lipoprotein cholesterol, non-high-density lipoprotein cholesterol, and triglyceride concentrations. All these parameters were elevated in patients with NAFLD.

Dr. Panahi Y, the lead scientist, after taking into account other factors, wrote, " This study has shown beneficial effects of ALE supplementation on both ultrasound liver parameters and liver serum parameters (ALT, AST, APRI ratio, and total bilirubin) in patients with NAFLD".

In order to reveal more information about artichoke anti NAFLD activity, researchers elucidated the physiological responses and mechanisms of JA intervention with a comprehensive transcriptome analysis.

The study included Wistar rats fed a control diet, a 60 % fructose-enriched diet (FRU), or an FRU with 10 % JA (n 6-7) for 4 weeks with an oral glucose tolerance test was carried out on day 21. 

According to the assays, dietary JA supplementation improved the IR and hepatic TAG accumulation induced by the Fructose feeding.

JA also inhibited the gene expression of malic enzyme 1 (Me1), associated with fatty acid synthesis; decorin (Dcn), related to fibrosis; and cytochrome P450, family 1, subfamily a, polypeptide 2 (Cyp1a2) and nicotinamide phosphoribosyltransferase (Nampt), associated with inflammation.

Based on these results, researchers proposed that 10 % of JA supplementation may be beneficial for the prevention of the onset of NAFLD.

Macular degeneration is a condition caused by the blurring of the center of the vision without affecting the side or peripheral vision.

Age-related macular degeneration (AMD) is an aging process found in older adults is the leading cause of blindness in adults over the age of 55.

Most adults with age-related macular degeneration (AMD) are asymptomatic. However, it can be detected by professions during annual eye check-up.

Untreated AMD, over time, can lead the increased size of blur-area and interfere with the vision, including reading.

AMD can be classified into 2 types
* The dry form is a milder form found in most patients with AMD with gradual degeneration of the central retinal tissues that make up the macula.

* The wet form is the serious form of AMD account for only 10% in all patients

However, the dry form can develop into the wet form over time.

Conventionally, as of today, there is no effective treatment for AMD. Most treatments such as from low vision aids and lifestyle control are to delay the progression and improve quality of life.

In some cases, wet AMD may be treated with medications injected into the eye to stop leaking blood vessels, depending on the disease severity.

Besides aging and exposure to UVB, smoking has been found to increased risk AMD by 2-5 fold.

Dr. Sara Velilla, the lead scientist said, "Age-related macular degeneration (AMD) is one of the main socioeconomic health issues worldwide. AMD has a multifactorial etiology with a variety of risk factors. Smoking is the most important modifiable risk factor for AMD development and progression".

Tomato is red, edible fruit, genus Solanum, belongs to family Solanaceae, native to South America. Because of its health benefits, the tomato is grown worldwide for the commercial purpose

With an aim to find a natural treatment of eye disease, researchers examined the combinations of antioxidants including lutein+zeaxanthin (1 μM; 0.2 μM), zinc (10 µM), carnosic acid (2 µM), beta-carotene (2 µM), and standardized tomato extract containing lycopene and other tomato phytonutrients added to the culture media.against age-related macular degeneration (AMD).

Injection of the combination protected the eye against age-related macular degeneration (AMD) by yielding an antioxidative, anti-inflammatory, and antiangiogenic effect in M1 and M2 macrophages.

The results of the effects were attributed to the upregulation of antioxidative genes (HMOX1, SOD1) expression and the inhibition of pro-angiogenic genes and pro-inflammatory genes (SDF-1, TNF-alpha, IL-6, MCP-1).

Compared to the combination, lutein monotherapy or a combination of lutein and zinc had less effect on the expression of these genes.

Dr.Rinsky B, the lead scientist said, "Combinations of supplements can modify the expression of genes and proteins that may be relevant for the involvement of macrophages in the pathogenesis of AMD".

Futhermore, in oxidative stress-induced damage to the retinal pigment epithelial (RPE) on age-related macular degeneration and neovascular retinopathies (e.g., retinopathy of prematurity, diabetic retinopathy, researchers investigated xanthophyll carotenoids, lutein, and zeaxanthin, isolated from toma in damage of retinal pigment epithelial (RPE) cells.

Also, administration of carotenoids, lutein, and zeaxanthin showed to protect the macula (and the broader retina) via their antioxidant and photoprotective activities.

In undifferentiated ARPE-19 cells, lutein and lycopene, but not β-carotene, protected the cell integrity and viability by inhibiting the overexpression of retinal cell growth.

Moreover, pre-incubation of ARPE-19 cells with lutein or lycopene protected against tBHP-induced cell loss and cell co-exposure of lutein or lycopene with tBHP essentially neutralized tBHP-dependent cell death at tBHP concentrations up to 500 μM.

Where tert-Butyl hydroperoxide (tBHP) is an organic peroxide used to induce oxidation processes.

In other words, lutein and lycopene protected the RPE against oxidative stress-induced cell loss

Taken altogether, tomatoes may be considered a natural remedy for the prevention and treatment of age-related macular degeneration (AMD), pending to the confirmation of the larger sample size and multicenter human study.

Intake of lutein and lycopene in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

This randomized double-blind, placebo-controlled trial with a 1:1 allocation included 100 selected infertility candidates who were admitted to Royan Institute for Reproductive Biomedicine, Tehran, Iran.

Patients were randomly assigned to receive either ginger given a 3-month oral treatment (members received capsules containing 250 mg of ginger powder twice a day in ginger or placebo.

By analyzing the sperm concentration, motility, and SDF from the standardized semen samples before and after treatment, according to the World Health Organization, researchers found that
according to the multivariate analysis, patients receiving ginger (17.77, 95%CI: 6.16-29.39) was found to have a lower average positive percentage of SDF in compared with placebo, after three months of treatment without affecting the semen parameters.

In other words, ginger improves male fertility by decreasing SDF in infertile men.

Where SDF assay is a better diagnostic potential in evaluating the male infertilities, particularly in those with unexplained cause of infertility.

Researchers in the differentiation of the role of sperm DNA fragmentation in male factor infertility
wrote, "The significant role of SDF in male factor infertility is supported by current evidence. The beneficial role of SDF testing in the selection of varicocelectomy candidates, evaluation of patients with unexplained infertility and recurrent pregnancy loss, selection of the most appropriate assisted reproductive technique with the highest success rate for infertile couples, and assessment of infertile men with modifiable lifestyle factors or gonadotoxin exposure has been recently proposed".


Taken altogether, ginger may be considered a functional remedy for the improvement of male fertility, pending to the confirmation of larger sample sized and multicenter human study.

Phytochemical Sinigrin, The Antibacterial Functional Supplement

Bacterias are a class of microorganism consisted of a single cell with a simple internal structure with a number of different shapes.

Not all bacterias are harmful to human. Some are essential to human life, with benefits related to improve the digestive function, prevent overexpression of harmful disease-causing bacteria, and the synthesis of essential nutrients and vitamins.

Believe or not, human gut contains over 10 trillions of bacterias including at least 1000 different species of known bacteria with more than 3 million genes.

In a healthy individual, the levels of microorganisms in the gut are balanced.

The gut microbiota plays a critical role in breaking down foods that stomach and small intestines cannot digest, stimulating the production of vitamin K and D, protecting the intestinal against damage caused by bad bacteria and increasing the immune functioning. 

Bacterias that cause disease are called pathogenic microorganisms.

Epidemiological studies suggested that Mycobacterium tuberculosis bacteria is one of the bacteria that causes tuberculosis, killing over 2 million people a year, mostly in sub-Saharan Africa.

Other pathogenic bacterias include Streptococcus and Pseudomonas, and Shigella, Campylobacter, and Salmonella.

According to the statistics, pathogenic bacterial infection is a leading cause of death of infants in developing countries.

On the finding a bioactive ingredient with antibacterial activity, researchers examined allyl isothiocyanate (AIT) is derived from the glucosinolate sinigrin in E. coli O157:H7 numbers in food products.

Injection of AIT showed a greater antimicrobial activity at low pH values. In other words, at pH 4.5 and 5.5 the MIC was 25 microL/L, while at pH 8.5, 500 microL/L was required to inhibit bacterial growth. 

Furthermore, bacterial decomposition was found at the highest rate in water at 37 degrees C.

Moreover, in bacterial enzymes (the thioredoxin reductase and acetate kinase), AIT at 10 to 100 microL/L was able to significantly inhibit both enzymes, but only 1 microL/L was needed to decrease the activity of thioredoxin reductase.

Interestingly, in a Nanocomposite carboxymethyl cellulose films tested on Escherichia coli O157:H7-containing sinigrin (SNG) prepared by stirring 2% (w/v) carboxymethyl cellulose (CMC) and 2% (w/v) glycerol (as a plasticizer) in distilled water with or without SNG (an antimicrobial precursor) as a 99% pure reagent (pSNG) or as a crude extract (cSNG), researchers found that
* In beef pieces measuring 6 × 5 × 2 cm(3) (L × W × H) were dipped in an E. coli O157:H7 broth suspension containing >8 log10 CFU ml(-1) and were drained for 3 min over a sterile cloth, the CMC and NCMC films without SNG were not antimicrobial against E. coli O157:H7; however, NCMC and CMC films with SNG were highly antimicrobial.

* After 5 days at 8°C, E. coli O157:H7 was reduced more than 4 log10 by the NCMC•pSNG film.

* The reduction remained almost the same until 18 days at 8°C when E. coli O157:H7 was reduced >5 log10 CFU g(-1) meat.

The findings strongly support the use of sinigrin for food storage against bacteria.

Taken altogether, sinigrin a bioactive ingredient in cruciferous vegetables may be considered a functional supplement for the treatment of bacterial infection, pending to the confirmation of large sample size and multicenter human study.

However, intake of bioactive in form of supplement should be taken with extreme care, as liver toxicity may be induced if overdosed.

Arthritis is a group of diseases associated with the inflammation of the joints. According to the epidemiological studies osteoarthritis (OA) and rheumatoid arthritis (RA) are 2 most common types of arthritis.

According to the information provided by the arthritis foundation, arthritis affects over 54 million people in adults of which diagnosed by doctors. Osteoarthritis is the most prevalent type in the US, affecting 31 million Americans.

Depending on types and the location affected,  arthritis can be classified into over 100 conditions, but the most common types of arthritis include
* Osteoporosis is a condition of thinning of bone and bone tissues as a result of the loss of bone density over a long period of time.

* Osteoarthritis (OA), is a condition caused by the wear and tear on a joint due to aging, affecting over 25 million people in the United States alone.

* Rheumatoid Arthritis is a chronic disorder as a result of inflammation, affecting mostly the flexible (synovial) joints and tissues and organs in the body. 

* Polymyalgia arthritis is a condition of inflammatory rheumatic disease induced pain, stiffness, and tenderness in large muscles, including muscles shoulders and pelvic girdle.

* Fibromyalgia in the newly proposed criteria for the classification of fibromyalgia are 1) widespread pain in combination with 2) tenderness at 11 or more of the 18 specific tender point sites(a) as a result in responding to pressure.

As of today, there is no cure for arthritis, medications used in conventional medicine are to reduce pain, and symptoms and improve quality of life for patients to carry out daily activity.

3,3'-Diindolylmethane or DIM are phytochemicals derived from the digestion of indole-3-carbinol, belonging to the group of Indoles, found abundantly in broccoli, Brussels sprouts, cabbage, and kale, etc.

In the urgency to discover a natural compound for the treatment of diseases associated with osteoclastogenesis, researchers investigated the therapeutic efficacies of DIM on experimental arthritis.

The study included a rat model of adjuvant-induced arthritis (AIA), with daily AIA paw swelling.

According to the clinical and histologic indices of inflammation and tissue damage, administration of DIM attenuated AIA in animal models.

Where Receptor activator of nuclear factor kappa-Β ligand (RANKL) is a protein involved the activation-induced cytokine, osteoprotegerin ligand, and osteoclast differentiation factor.

Dr. the lead researcher wrote, "DIM has shown anti-arthritis activity in animal models via inhibiting the expression of RANKL, and thus may offer potential treatments for arthritis and associated disorders".



Taken altogether, 3,3'-Diindolylmethane derived from the digestion of indole-3-carbinol may be considered a supplement for the prevention and treatment of arthritis, pending to the confirmation of the larger sample size and multicenter human study.

Herbal Turmeric, the Best Functional Herb Which Processes Anti Diseases Associated with Chronic Low Grade Inflammation Potential

Inflammation is a natural response induced by the immune system in which the white blood cells in the first line of defense try to destroy the harmful pathogens before they can enter the body to multiply in the acute phase of infection

However, overproduction of pro-inflammatory cytokines in this phase can cause damage to the nearby healthy cells, leading abnormal production of collagen ratio in the initiation of scars forming.

Most pathogens are stopped in the acute phase of infection, depending on the location and the size of the wound and tissues damage.

If the immune system can not kill off all invaded pathogens with a set of period of time, it will compromise, leading to the low-grade chronic inflammation with the persistent production of pro-inflammatory cytokines in the affected area.

Long-term low-grade inflammation ultimately leads to tissues and organs damage, inducing a number of chronic illness, including diabetes and cardiovascular disease.

Symptoms of low-grade chronic inflammation include body pain as in arthritis, constant fatigue and insomnia as in cancer, depression, anxiety and mood disorders due to pain. gastrointestinal complications like constipation, diarrhea, and acid reflux, unintended weight loss or gain depending on the infected tissues and organs.

According to statistics, chronic inflammatory diseases affects approximately 350 million people worldwide and nearly 43 million people in the United States or almost 20% of the population. The prevalence of diseases associated with chronic inflammation is anticipated to increase persistently for the next 30 years, causing a financial burden on the economy and health care sector.

Turmeric is a perennial plant in the genus Curcuma,  belonging to the family Zingiberaceae, native in demagogical South Asia.

The herb has been used in traditional medicine as anti-oxidant, hypoglycemic, colorant, antiseptic, wound healing agent, and to treat flatulence, bloating, and appetite loss, ulcers, eczema, inflammations, etc.

With an aim to find a natural compound for the treatment of diseases associated with chronic inflammation, including cancers, researchers examined the effects of the turmeric through its bioactive components.

According to the review of previous studies, turmeric oil, present in turmeric, can enhance the bioavailability of curcumin.

In other words, essential oil isolated from turmeric in the form of curcumin-free turmeric (CFT) components is more effective for the treatment of chronic inflammatory disease compared to other extracts.

In the components analysis, turmerin, turmerone, elemene, furanodiene, curdione, bisacurone, cyclocurcumin, calebin A, and germacrene were all contributed to the efficacy of the curcumin-free turmeric (CFT).

In examining the effect of ar-turmerone (ATM) and turmerones (TUR, a mixture of 5 sesquiterpenes including ATM) against disease associated with inflammation, researchers found that both CUR and ATM inhibited lipopolysaccharide (LPS)-induced expression of inducible forms of inflammation by exhibiting the production of free radicals and protein involved overexpression proinflammatory cytokines.

Furthermore, ATM prevented LPS-induced translocation of certain inflammatory genes.

In a colitis model, oral administration of TUR significantly protected the large bowel against 2% dextran sulfate sodium (DSS)-induced shortening of the large bowel by 52-58% and colon carcinogenesis.

Moreover, at a low dose, TUR markedly suppressed adenoma multiplicity by 73%, compared to the range of 63-69% of CUR.

Also, the combination of CUR and TUR at both low and high doses showed a complete abolish of tumor formation.

Based on the findings, Murakami A, the lead scientist in the chronic inflammatory disease team wrote, "we consider that its use in combination with CUR may become a powerful method for prevention of inflammation-associated colon carcinogenesis".

Taken altogether, turmeric in the form of essential oil, CUR, and TUR may be considered herbal remedies for the prevention and treatment of diseases associated with chronic inflammation, pending to the confirmation of the larger sample size and multicenter human study.

Intake of turmeric in form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.
